镧对铅胁迫下小麦幼苗抗氧化酶活性的影响   总被引:20,自引:0,他引:20

在铅胁迫的条件下 ,研究了镧对小麦幼苗生长期地上部分及根系超氧化物岐化酶 (SOD)、过氧化氢酶 (CAT)的活性、脂质过氧化产物丙二醛 (MDA)的含量以及对植物生长的影响 .实验在严格控制的人工气候室进行 1 2d .Pb(NO) 3 处理量为 0 ,5 0 ,1 0 0mg·l- 1.在第二天至实验结束的研究结果表明 ,加La组与对照组比较有明显差异 ,根系的SOD和CAT各增加 0 4 5— 1 6 5倍和 32 2— 77 77% ,MDA减少 1 1 0 5—2 7 4 9% .当有铅胁迫时 ,在 0— 5d内 ,与对照组比较 ,镧使小麦幼苗的地上部分及根系的SOD和CAT有少量的但有统计学意义的增加 (p <0 0 5 ) ,MDA含量则减少 .表明La(NO3 ) 3 使抗氧化酶提高 ,增加了小麦抗铅胁迫的能力 ,但是只是在胁迫初期和胁迫不很严重的条件下 ,才具有这一作用 .  相似文献

PM2.5引起的健康危害日益受到广泛关注,但其确切的损伤机理仍不完全清楚,目前也缺乏有效拮抗PM2.5损伤的天然活性物质。因此本文对北京市PM2.5造成CHO细胞的损伤作用及天然物质的拮抗作用进行研究,结果可为PM2.5污染治理和疾病的预防提供科学依据。通过CCK-8法测定细胞存活率;通过流式细胞仪测定细胞凋亡;用荧光探针DCFH-DA法测定ROS;提取细胞内总蛋白并测定其中SOD含量;以及通过Western Blot法分别测定了p-akt/akt、p65及Bad的相对表达量。结果显示:(1)10~40μg·m L-1的PM2.5可明显降低CHO细胞存活率,并呈现极显著的剂量-效应关系(r=-0.964,P0.01);15μg·m L-1PM2.5可以显著增加细胞凋亡、引起细胞内ROS增加、SOD含量下降;p-akt/akt、p65表达量增加说明Akt通路与NF-κB通路均被激活。(2)20μmol·L-1阿魏酸、50μmol·L-1绿原酸和10μmol·L-1荭草素预处理可拮抗PM2.5引起的细胞存活率下降以及细胞凋亡率增加,同时降低细胞内ROS并增加SOD含量,下调p-akt/akt、p65及Bad的相对表达量。其中20μmol·L-1阿魏酸的保护作用最佳。结果说明,PM2.5引起的CHO细胞损伤与细胞凋亡与Akt和NF-κB通路的激活有关;三种天然活性物质具有拮抗PM2.5造成的细胞损伤的能力,其保护机理是通过其降低PM2.5引起的细胞内氧化应激反应,进而降低被PM2.5激活的Akt通路与NF-κB通路和下调Bad蛋白表达量来实现的。  相似文献

The gene expression of environmental organisms is useful as a biomarker of environmental pollution. One of its advantages is high sensitivity. We identified the cDNA of a novel cadmium-responsive gene in the soil collembolan Folsomia candida. The deduced protein, designated “metallothionein-like motif containing protein” (MTC), was cysteine-rich and contained a metallothionein-like motif with similarity to metallothionein, but had a much longer sequence than metallothionein and contained repeated sequences of amino acids. Expression of MTC mRNA was sensitively induced by cadmium exposure at 0.3 mg/kg of dry food, a concentration at which toxic effects are not observed, but expression was not affected by γ-ray exposure (an inducer of oxidative stress). These findings suggest that MTC is involved in cadmium-binding processes rather than in oxidative-stress responses. In conclusion, we suggest that gene expression of MTC may be a candidate biomarker for detecting low levels of cadmium contamination in soil.  相似文献

In this work we measured a set of antioxidative and photoprotective compounds (chlorophylls, carotenoids, tocopherol, ascorbate and glutathione), which were suggested previously as stress markers in conifer needles, at two spruce forest sites at different elevation in Saxony, Germany. Most variables differed significantly between current and 1-year-old needles, but only the content of the xanthophyll cycle per mg total chlorophyll and the oxidation state of glutathione were significantly different between the sites. We applied principal component analysis (PCA) to address the question if underlying accumulated variables are similar to the ones found in spruce needles across Alpine elevation profiles and/or for pines in Mediterranean ecosystems. Four principal components (accumulated variables, PC) representing 68% of the total variance of the dataset were extracted. PC 1 encompassed total chlorophyll, lutein, and β-carotene contents, PC 2 combined the epoxidation state of xanthophylls, ascorbate content and redox state, and glutathione content, PC 3 represented the content of xanthophylls and the redox state of glutathione, and PC 4 encompassed the content of α-carotene and the epoxidation state of xanthophylls. Only PC 3 was significantly different between sites. The PCA structure shows many similarities to corresponding findings in studies on spruce in mountain forests in the Alps and pines in Mediterranean systems. This corroborates the interpretation of PCs as indicative for underlying physiological processes. However, separation of the two investigated sites by PCs was in the present case study not superior to the separation by single input variables.  相似文献

Abstract

Sixty female prawn seed collectors and 60 female control subjects from Sajenakhali and Sandeshkhali blocks of Sunderbans, West Bengal, India, were randomly selected to evaluate and compare musculoskeletal disorders and physiological stress. The control group was engaged in domestic work involving minimum hand-intensive activities. The modified Nordic musculoskeletal questionnaire and rapid entire body assessment were used. Most subjects suffered from discomfort in different body parts, especially in the lower back (98%), knees (88%), shoulders (75%), ankles (70%) and feet (67%). This study reveals that female prawn seed collectors suffer from significant physiological load and extreme physiological stress due to prolonged working hours in a standing posture and excessive work pressure. Consequently, all these factors affect female prawn seed collectors ‘ health and work performance.  相似文献

Nurses are often obliged to work in a 12-hr shift work system. We have decided to check whether such a working system constitutes an excessive load for nurses. On the basis of a questionnaire survey among nurses working in an 8-hr daytime shift system (169 nurses) and in a 12-hr shift (536 nurses), the amount of physical workload, work stress, and after-work activities were compared. Data analysis has shown that a 12-hr shift system is characterized by less significant physical workload but greater mental load. The nurses working in a 2-shift system were more tired after work, but they could spend more time on leisure activities and do housework. The data suggest that there are no significant contraindications for nurses to work in a 2-shift system.  相似文献

Most research on workplace aggression focuses on the antecedents and consequences of aggression for individual workers. The current study examines how shared workplace aggression concerns relate to internal and external organizational outcomes. Drawing on the work stress, social identity, and social contagion literatures, we propose relationships between unit‐level aggression concerns and unit‐level measures of performance and employee attitudes in a public school sample (2989 employees; 163 schools). We also propose that these relationships differ depending on the resource context of the school. Consistent with our expectations, schools in which teachers had strong shared concerns about aggression also had poorer shared job attitudes and poorer student outcomes, as indicated by average standardized test scores at the school. The impact of shared concerns about aggression on school‐level standardized test scores was stronger for resource‐rich schools than for schools with fewer resources. The current study contributes to organizational research as it establishes that workplace aggression concerns have adverse consequences for the whole organization, not only for the individuals experiencing the events. Our findings demonstrate that employees' shared concerns about aggression have an impact on organizational outcomes and suggest a prevention‐focused approach to workplace aggression. We theorized that organization–environment adaptation, the interaction between external demands and personnel resources, predicts distress and morale. We tested this hypothesis in 29 stations within one state police department, and combined three data sources. We measured environmental demands for policing via census data pertaining to the station precinct (e.g., per cent unemployed; per cent in public housing). We assessed resources via personnel numbers within stations. Outcomes were employee's perceptions of staff distress and morale at the station (N = 247), assessed twice, 14 months apart. Using hierarchical linear modeling, we found that environmental demands predicted perceptions of workgroup distress and morale and that the relationships were moderated by personnel resources. For distress, when resources were low, demands were positively associated with distress; when resources were high, demands were negatively associated with distress. For morale, when resources were high, demands were positively associated with morale; there was no relationship when resources were low. Results show that aversive and pleasurable reactions at work may be traced to how resources are employed to manage external demands. Results support a macro‐level shift in modeling distress and morale, incorporating external demands, and strategic management decisions regarding personnel resourcing. Our research suggests that rather than being a result of individual failure to adapt, compromised work ability may result from an organizational failure to adapt to the environmental context. 按照毒性试验标准方法研究了1-癸基-3-甲基咪唑氯盐([DMIM]Cl)、1-十二烷基-3-甲基咪唑氯盐([C12MIM]Cl)对水稻幼苗生长的影响,测定了水稻根的半数抑制浓度(IC50-5d)、超氧化物歧化酶(SOD)活性、过氧化氢酶(CAT)活性、过氧化物酶(POD)活性、水稻根部丙二醛(MDA)含量及对水稻根系的生理影响.结果表明:两种离子液体(ILs)对水稻幼苗根部生长均具有抑制作用,随着浓度的增大抑制率增加,[DMIM]Cl和[C12MIM]Cl的根长IC50-5d分别为0.10 mg·L-1和0.054 mg·L-1,水稻幼苗根部对两种ILs均有氧化应激反应,在[DMIM]Cl和[C12MIM]Cl处理下,3种抗氧化酶活性均受到严重抑制,丙二醛(MDA)含量升高,加剧了水稻幼苗根部细胞膜脂质过氧化作用.[DMIM]Cl和[C12MIM]Cl处理导致根系活力降低,根系质膜透性增大.  相似文献
