Organic solvents, such as dimethylsulfoxide (DMSO) and methanol are widely used as vehicles to solubilise lipophilic test compounds in toxicity testing. However, the effects of such solvents upon innate detoxification processes in aquatic organisms are poorly understood. This study assessed the effect of solvent exposure upon cytochrome P450 (CYP)-mediated xenobiotic metabolism in Daphnia magna and zebrafish larvae (4 d post fertilisation). Adult D. magna were demonstrated to have a low, but detectable, metabolism of ethoxyresorufin in vivo and this activity was not modulated by pre-exposure to DMSO or methanol (24 h, up to 0.1% and 0.05% v/v, respectively). In contrast, the metabolism of ethoxyresorufin in zebrafish larvae was significantly reduced by both solvents (0.1% and 0.05% v/v, respectively) after 24 h of exposure. In zebrafish, these observed decreases in activity towards ethoxyresorufin were accompanied by decreased expression of a variety of genes coding for drug metabolising enzymes (corresponding to CYP1, CYP2, CYP3 and UDP-glucuronyl transferase [UGT] family enzymes), measured by quantitative PCR. Reduction of gene expression and CYP1 enzyme activities by methanol (0.05% v/v) in zebrafish larvae was partially reversed by co-exposure with Aroclor 1254 (100 μg L−1). Overall this study suggests that relatively low concentrations of organic solvents can impact upon the biotransformation of certain xenobiotics in zebrafish larvae, and that this warrants consideration when assessing compounds for metabolism and toxicity in this species. 相似文献

为探讨华南珠江三角洲和湘桂走廊及其周边地区的酸沉降硫源,分别对大气降水、大气SO2、气溶胶和工业用煤、重油及其燃烧产物进行了硫同位素组成测定.研究结果表明,珠江三角洲和湘桂走廊地区的大气降水硫同位素组成有明显的差异,其大气降水δ34S值的变化范围分别为1.9‰~10.3‰和-4.8‰~-0.1‰.湘桂走廊地区大气降水明显的富集轻硫同位素(32S),而珠江三角洲地区则富集重硫同位素(34S).在煤、重油燃烧过程中硫同位素分馏效应研究基础上,将该地区酸雨硫源区分为4种类型,即人为成因硫、天然生物硫、海雾硫和远距离传输硫.在定量计算不同硫源对酸雨贡献的基础上.提出人为成因硫是珠江三角洲和湘桂走廊地区最强的污染硫源,而生物硫在夏季贡献突出,其贡献率分别为47%和52%.传输硫在冬季贡献率可达49%.上述结果,对研究治理华南地区的酸雨具有重要意义. 相似文献

We analyzed 13 years of hourly measurements of SO2, NOx, and O3, at forest ecosystem research sites in SE Germany. A quasi-continuous data record was obtained by combining data sets from
two locations. Before interpreting trends in the combined data set, we analyzed if the change of location introduced a systematic
bias. We employed autocorrelation functions, Hurst statistics, complexity analysis, and recurrence quantification and found
that the partial data sets exhibited no indication of the presence of any bias. For SO2, we also compared the data from the forest sites with data obtained in nearby cities and also found no indications for any
systematic effects. Applying nonparametric trend statistics we found a significant decrease of the SO2. Most of the observed decrease is due to the reductions of SO2 emissions in eastern Germany, but reductions in western Germany and the Czech Republic also played important roles. Extracellular, oxidative soil enzymes like monophenol oxidases and peroxidases play an important role in transformation of xenobiotics and the formation of organic matter in soil. Additionally, these enzymes may be involved in the formation of non-extractable residues (NERs) of xenobiotics during humification processes. To examine this correlation, the fate of the fungicide 14C metalaxyl in soil samples from Ultuna (Sweden) was studied. Using different soil sterilization techniques, it was possible to differentiate between free, immobilized, and abiotic (“pseudoenzyme”-like) oxidative activities. A correlation between the formation of metalaxyl NER and soil organic matter content, biotic activities, as well as extracellular phenoloxidase and peroxidase activities in the bulk soil and its particle size fractions was determined. Extracellular soil-bound enzymes were involved in NER formation (up to 8% of applied radioactivity after 92 days) of the fungicide independently from the presence of living microbes and different distributions of the NER in the soil humic subfractions. 相似文献
