Biodegradability testing was performed in an aqueous environment under anaerobic conditions after inoculation with digested sludge from municipal wastewater treatment plant. In cross-linking with 1,2:3,4-diepoxybutane in limits 0.8–9.1% weight, biodegradability degree decreased from 76.8 to 62.2%; when 1,2:7,8 diepoxyoctane in quantities 1.1–13.2% weight was used, biodegradability degree dropped more prominently – from 72.3 to 22.8%. There is obviously a direct connection between growing cross-link degree (assessed by so-called fixation index) and decreasing readiness to biodegradation, apparently owing to build-up of a network forming an obstacle to access of micro-organisms and enzymes.  相似文献

In recent years there has been an increasing interest in the use of autohydrogenotrophic bacteria to treat nitrate from wastewater. However, our knowledge about the characteristics of extracellular polymeric substances (EPS) releasing by these activities is not yet very advanced. This study aimed to investigate the change in EPS compositions under various pH values and hydrogen flow rates, taking into consideration nitrogen removal. Results showed that pH 7.5 and a hydrogen flow rate of 90 mL/min were the optimal operating conditions, resulting in 100% nitrogen removal after 6 hr of operation. Soluble and bound polysaccharides decreased, while bound proteins increased with increasing pH. Polysaccharides increased with increasing hydrogen flow rate. No significant change of bound proteins was observed at various hydrogen flow rates.  相似文献

Spatial and temporal changes in the biochemical composition of particulate organic matter in a semi-enclosed marine system (Stagnone di Marsala, Mediterranean Sea) were studied, on a monthly basis, from January to December 1994, in order to assess nutritional value of suspended particles for benthic suspension feeders. According to previous findings, the study site displayed a strong oligotrophy. Chlorophyll-a accounted for a very low fraction of the total suspended matter pool (0.1%), whereas at least 75% of POC was of detrital/heterotrophic origin. POC: PON ratio values indicate that bacterioplankton biomass accounted for a significant fraction of the total POC pool, displaying values comparable to those of the phytoplankton biomass (phytoplankton to bacterial biomass ratio was about (1). Temporal and spatial changes in the biochemical composition of particulate organic matter were rather limited and related to its sources, the main of which is represented by detrital particles released by the Posidonia oceanica (L.) beds. the comparison between our results and those encountered in other coastal lagoons indicates that the low abundance of suspension-feeding organisms observed in the study area is related to the “quality depression” of particles due to the dilution of high quality compounds (i.e., biopolymeric carbon) in a largely inorganic matrix. This result leads us to conclude that, to reach the same amount of high quality particulate food, a suspension feeder mollusc in the Marsala lagoon would need to filter a sea water volume around 3 times higher than in other Mediterranean coastal lagoons.  相似文献

We report that green algae in lakes and rivers can serve as precursors of halobenzoquinone(HBQ) disinfection byproducts(DBPs) produced during chlorination. Chlorination of a common green alga, Chlorella vulgaris, produced 2,6-dichloro-1,4-benzoquinone(2,6-DCBQ), the most prevalent HBQ DBP in disinfected water. Under varying p H conditions(p H 6.0–9.0), 2,6-DCBQ formation ranged from 0.3 to 2.1 μg/mg C with maximum formation at p H 8.0. To evaluate the contribution of organic components of C. vulgaris to 2,6-DCBQ formation, we separated the organics into two fractions, the protein-rich fraction of intracellular organic matter(IOM) and the polysaccharide-laden fraction of extracellular organic matter(EOM). Chlorination of IOM and EOM produced 1.4 μg/mg C and 0.7 μg/mg C of 2,6-DCBQ, respectively. The IOM generated a two-fold higher 2,6-DCBQ formation potential than the EOM fraction, suggesting that proteins are potent 2,6-DCBQ precursors. This was confirmed by the chlorination of proteins extracted from C. vulgaris: the amount of 2,6-DCBQ produced is linearly correlated with the concentration of total algal protein(R2= 0.98). These results support that proteins are the primary precursors of 2,6-DCBQ in algae, and control of green algal bloom outbreaks in source waters is important for management of HBQ DBPs.  相似文献

Microorganisms, plants, and animals have successfully colonized cold environments, which represent the majority of the biosphere on Earth. They have evolved special mechanisms to overcome the life-endangering influence of low temperature and to survive freezing. Cold adaptation includes a complex range of structural and functional adaptations at the level of all cellular constituents, such as membranes, proteins, metabolic activity, and mechanisms to avoid the destructive effect of intracellular ice formation. These strategies offer multiple biotechnological applications of cold-adapted organisms and/or their products in various fields. In this review, we describe the mechanisms of microorganisms, plants, and animals to cope with the cold and the resulting biotechnological perspectives.  相似文献

•Tryptophan protein, and aromatic protein I/II were the key identified proteins. •Cysteine was more correlated with methane production than other amino acids. •The presence of cysteine can promote methane production and degradation of VFAs. •The presence of cysteine can lower ORP and increase biomass activity. •Predominant Tissierella and Proteiniphilum were noted in pretreated sludge samples. Many studies have investigated the effects of different pretreatments on the performance of anaerobic digestion of sludge. However, the detailed changes of dissolved organic nitrogen, particularly the release behavior of proteins and the byproducts of protein hydrolysis-amino acids, are rarely known during anaerobic digestion of sludge by different pretreatments. Here we quantified the changes of three types of proteins and 17 types of amino acids in sludge samples solubilized by ultrasonic, thermal, and acid/alkaline pretreatments and their transformation during anaerobic digestion of sludge. Tryptophan protein, aromatic protein I, aromatic protein II, and cysteine were identified as the key dissolved organic nitrogen responsible for methane production during anaerobic digestion of sludge, regardless of the different pretreatment methods. Different from the depletion of other amino acids, cysteine was resistant to degradation after an incubation period of 30 days in all sludge samples. Meanwhile, the “cysteine and methionine metabolism (K00270)” was absent in all sludge samples by identifying 6755 Kyoto Encyclopedia of Genes and Genomes assignments of genes hits. Cysteine contributed to the generation of methane and the degradation of acetic, propionic, and n-butyric acids through decreasing oxidation-reduction potential and enhancing biomass activity. This study provided an alternative strategy to enhance anaerobic digestion of sludge through in situ production of cysteine.  相似文献

Parasporin, a Bacillus thuringiensis parasporal protein, is unique in having a strong cytocidal activity preferential for human cancer cells. In this study, we characterized parasporin activities associated with three novel geographical isolates of B. thuringiensis. Parasporal inclusion proteins of the three isolates were highly toxic to human uterus cervix cancer cells (HeLa), but not to non-cancer uterine smooth muscle cells (UtSMC). Inclusions of the isolates lacked insect toxicity and hemolytic activity against sheep erythrocytes. Ouchterlony immunodiffusion tests revealed that the proteins of the three isolates are immunologically closely related to parasporin-1 (Cry31A), but dissimilar to the three other existing parasporin groups. Our results provide evidence that the parasporin-1-producing organism is a common member in B. thuringiensis populations occurring in natural environments of Japan.  相似文献

A promising alternative to conventional single phase processing, the use of sequential anaerobic-aerobic digestion, was extensively investigated on municipal sewage sludge from a full scale wastewater treatment plant. The objective of the work was to evaluate sequential digestion performance by testing the characteristics of the digested sludge in terms of volatile solids (VS), Chemical Oxygen Demand (COD) and nitrogen reduction, biogas production, dewaterability and the content of proteins and polysaccharides. VS removal efficiencies of 32% in the anaerobic phase and 17% in the aerobic one were obtained, and similar COD removal efficiencies (29% anaerobic and 21% aerobic) were also observed. The aerobic stage was also efficient in nitrogen removal providing a decrease of the nitrogen content in the supernatant attributable to nitrification and simultaneous denitrification. Moreover, in the aerobic phase an additional marked removal of proteins and polysaccharides produced in the anaerobic phase was achieved. The sludge dewaterability was evaluated by determining the Optimal Polymer Dose (OPD) and the Capillary Suction Time (CST) and a significant positive effect due to the aerobic stage was observed. Biogas production was close to the upper limit of the range of values reported in the literature in spite of the low anaerobic sludge retention time of 15 days. From a preliminary analysis it was found that the energy demand of the aerobic phase was significantly lower than the recovered energy in the anaerobic phase and the associated additional cost was negligible in comparison to the saving derived from the reduced amount of sludge to be disposed.  相似文献
