AbstractThe aim of this study was to produce Bacillus thuringiensis-based biopesticide using starch-producing industry wastewater (SIW) fortified with soybean medium and optimize the formulated product using different adjuvants. This study was necessary as low endotoxin concentration is obtained in formulated biopesticide when SIW alone is used as fermentation medium. The fermentation runs were conducted using SIW alone and SIW fortified with 25% soybean (w/v) medium in 2000?L and 150?L bioreactor, respectively. SIW supplemented with soybean medium showed an increase in cell count (from 1.95?×?108 to 1.65?×?109 CFU mL–1), spore synthesis (from 1.5?×?108 to 1.35?×?109 CFU mL–1) and endotoxin concentration (from 436 to 1170?μg mL–1) when compared to SIW medium alone. The fermented broth was concentrated using continuous centrifugation and adjuvants were added for biopesticide formulation in order to enhance its resistance against UV rays and rainfastness. Entomotoxicity of the formulation produced using fermented broth of SIW fortified with soybean (38,000?IU μL–1) was higher than that obtained by SIW medium alone (21,000?IU μL–1), commercial biopesticide Foray 76B (20,000?IU μL–1) and Btk sander’s (12,500?IU μL–1). 相似文献
The present study is aimed at assessing the ability of Bacillus sp.JDM-2-1 and Staphylococcus capitis to reduce hexavalent chromium into its trivalent form.Bacillus sp.JDM-2-1 could tolerate Cr(Ⅵ) (4800 μg/mL) and S.capitis could tolerate Cr(Ⅵ) (2800 μg/mL).Both organisms were able to resist Cd2+ (50 μg/mL),Cu2+ (200 μg/mL),Pb2+ (800 μg/mL),Hg2+ (50 μg/mL) and Ni2+ (4000 μg/mL).S.capitis resisted Zn2+ at 700 μg/mL while Bacillus sp.JDM-2-1 only showed resistance up to 50 μg/mL.Bacillus sp.JDM-2-1 and S.capitis showed optimum growth at pH 6 and 7,respectively,while both bacteria showed optimum growth at 37℃.Bacillus sp.JDM-2-1 and S.capitis could reduce 85% and 81% of hexavalent chromium from the medium after 96 h and were also capable of reducing hexavalent chromium 86% and 89%,respectively,from the industrial effluents after 144 h.Cell free extracts of Bacillus sp.JDM-2-1 and S.capitis showed reduction of 83% and 70% at concentration of 10 μg Cr(Ⅵ)/mL,respectively.The presence of an induced protein having molecular weight around 25 kDa in the presence of chromium points out a possible role of this protein in chromium reduction.The bacterial isolates can be exploited for bioremediation of hexavalent chromium containing wastes,since they seem to have the potential to reduce the toxic hexavalent form to its nontoxic trivalent form. 相似文献
The productivity and efficiency of cellulase are significant in cellulose hydrolysis. With the accumulation of volatile fatty acids (VFAs), the pH value in anaerobic digestion system is reduced. Therefore, this study will find out how the pH and the amount of acetate influence the enzymatic hydrolysis of cellulose. The effects of pH and acetate on cellulase produced from Bacillus coagulans were studied at various pH 5-8, and acetate concentrations (0-60 mmol/L). A batch kinetic model for enzymatic cellulose hydrolysis was constructed from experimental data and performed. The base hypothesis was as follows: the rates of enzymatic cellulose hydrolysis rely on pH and acetate concentration. The results showed that the suitable pH range for cellulase production and cellulose hydrolysis (represents efficiency of cellulase) was 2.6-7.5, and 5.3-8.3, respectively. Moreover, acetate in the culture medium had an effect on cellulase production (K1= 49.50 mmol/L, n=1.7) less than cellulose hydrolysis (K1=37.85 mmol/L, n=2.0). The results indicated that both the pH of suspension and acidogenic products influence the enzymatic hydrolysis of cellulose in an anaerobic environment. To enhance the cellulose hydrolysis rate, the accumulated acetate concentration should be lower than 25 mmol/L, and pH should be maintained at 7. 相似文献
The use of microorganisms as biological control agents (BCAs) has become an effective alternative to chemical means of controlling plant pathogens. The antagonistic and inhibitory activity of 71 Bacillus spp. strains, which were isolated from different Mexican sites, were tested against several phytopathogen fungi, Fusarium oxysporum, Fusarium equiseti, Fusarium avenaceum, Bipolaris spp. and Alternaria spp. From the antagonism study, the strain ELI149 showed a marked inhibition of growth against all tested fungi; therefore crude metabolites from this strain were extracted using ethyl acetate and amberlite resin and probed against the same fungi as well as strains of Mucor sp., Penicillium spp. and Paecilomyces spp. The results indicated that amberlite was more suitable for extraction of secondary metabolites with antifungal activity. Finally, observation of cell damage in the tested pathogenic fungi showed marked morphological changes on reproductive structures in all tested fungi indicating that antibiosis was the mechanism of the antagonistic effect. These results suggest that metabolites from the Bacillus strains have a wide spectrum of antibiotic activities, which can be used as biocontrol agents for controlling fungal plant diseases of agricultural importance. 相似文献
One of the most common types of microbial pesticides used is Bacillus thuringiensis. Although generally considered safe, exposures to B. thuringiensis pesticide do occur where management by healthcare providers is sought. The objective of this investigation was to describe B. thuringiensis pesticide exposures reported to Texas poison centers. Cases were all B. thuringiensis pesticide exposures reported to Texas poison centers during 2000–2010. The distribution of cases was determined for various demographic and clinical factors. Of 115 total cases, 64% occurred during May–July. The rate per 1,000,000 population was 14.3 in central Texas and 4.4 in the rest of the state. Of the patients, 56% were of an age 20 years or older and 55% were male. The exposure routes were 57% by ingestion, 30% by dermal contact, and 11% by inhalation. The patient was managed on site in 93% of the exposures and only one exposure was judged to be potentially serious. B. thuringiensis pesticide exposures reported to Texas poison centers were more frequently reported from the central region of the state. The majority of exposures occurred during April–July, involved adults and males, occurred by ingestion or dermal route, were managed on site and did not result in serious outcomes. 相似文献
This study was carried out to assess the effectiveness of the combined use of earthworm (Alma millsoni) and bacterium (Bacillus sp.) in the bioremediation of spent engine oil (SEO) contaminated soils. A. millisoni were collected from the Botanical Garden of University of Ibadan, Nigeria. The stock culture of hydrocarbonoclastic Bacillus sp. was used for the bioremediation study. A set-up of eight pots containing 1000?g soil sample and 20?g of cow dung were mixed with 100, 75, 50 and 0?mL SEO respectively. Each of the set up was subjected to bioremediation agents; A. millisoni alone, Bacillus sp alone, A. millisoni and Bacillus sp, no treatment (control) in duplicate. Treatment of 100?ml SEO contaminated soil with combined A. millisoni and Bacillus sp resulted in significantly (P?0.05) lower levels of TPH and heavy metals (Ni and Pb), higher microbial activity and C:N ratio than the other treatments. The effectiveness of the bioremediating agents as indicated by reduction in the indices of petroleum hydrocarbon pollution followed the pattern: vermi-bacterium > bacterium > vermi-remediating agents. Earthworms exposed to 100?mLSEO-contaminated soil had higher CAT, SOD, and GPx activities compared to the control. Three acid-producing strains, AFB-1, AFB-2 and AFB-3, were isolated during this study, and their roles in anaerobic digestion of waste activated sludge (WAS) were evaluated. Data of 16S rRNA method showed that AFB-1 and AFB-2 were Bacillus coagulans, and AFB-3 was Escherichia coli. The removal in terms of volatile solids (VS) and total chemical oxygen demand (TCOD) was maximized at 42.7% and 44.7% by inoculating Bacillus coagulans AFB-1. Besides, the optimal inoculum concentration of Bacillus coagulans AFB-1 was 30% (v/v). Solubilization degree experiments indicated that solubilization ratios (SR) of WAS reached 20.8%±2.2%, 17.7%±1.48%, and 11.1%±1.53%. Volatile fatty acids (VFAs) concentrations and compositions were also explored with a gas chromatograph. The results showed that VFAs improved by 98.5%, 53.0% and 11.6% than those of the control, respectively. Biochemical methane potential (BMP) experiments revealed that biogas production increased by 90.7% and 75.3% when inoculating with Bacillus coagulans AFB-1 and AFB-2. These results confirmed that the isolated acid-producing bacteria, especially Bacillus coagulans, was a good candidate for anaerobic digestion of WAS.
