针对S.xinjiangensis分类地位的争议,从新疆再次分离获得34株快生大豆根瘤菌,在16SrDNAPCR-RFLP分析和SDS全细胞蛋白电泳分群的基础上,进行了16SrDNA全序列相似性和DNA同源性分析.所测4个菌株和S.fredii模式菌株USDA205的16SrDNA全序列有很高的相似性.而DNA同源性分析说明新分离的菌株代表与原定的S.xinjiangensis为一个DNA同源群.其模式菌株CCBAU110与S.fredii的2株参比菌株USDA194、2048之间的DNA同源性分别为31.5%和28.7%.S.fredii的模式菌株USDA205与新分离的菌株代表及原定的S.xinjiangensis的模式菌株和2个参比菌株之间的DNA同源性为20.8%~39%,远低于70%.属于种水平上的差异.按照国际细菌分类委员会以DNA同源性≥70%且△Tm≤5℃作为定种的标准,S.xinjiangensis是Sinorhizobium属中不同于S.fredii的一个独立的种.表3参20  相似文献

为了探讨纳米水滑石与Hela细胞的生物相容性,采用单细胞凝胶电泳法检测了纳米水滑石对Hela细胞DNA的损伤,并采用H2DCF-DA荧光法检测了纳米水滑石对Hela细胞活性氧簇的影响.结果发现,随着纳米水滑石浓度的升高(0~800μg·mL-1),Hela细胞尾部DNA含量、尾距及胞内活性氧簇含量均呈逐渐升高趋势;较低浓度(50、100μg·mL-1)纳米水滑石处理后,Hela细胞尾部DNA含量、尾距及胞内活性氧簇含量与对照组无显著性差异(p>0.05),而较高浓度(200、400、800μg·mL-1)纳米水滑石处理后,Hela细胞尾部DNA含量、尾距及胞内活性氧簇含量与对照组差异显著(p<0.05,p<0.01).以上结果表明,较高浓度(≥200μg·mL-1)的纳米水滑石可对Hela细胞DNA产生损伤,而较低浓度(≤100μg·mL-1)的纳米水滑石则具有较好的生物相容性,有可能作为载体应用于医药领域.  相似文献

By means of chorion biopsy together with restriction endonuclease analysis of fetal DNA, first trimester diagnoses were successfully made in 33 fetuses at risk for Bart's hydrops fetalis. Seven pregnancies with Hb H or hydrops fetalis were therapeutically terminated before 4 months of gestation. Of the 26 pregnancies intended to continue, 18 have come to term with normal deliveries; one with threatened abortion was terminated at the end of the first trimester and, seven are progresssing normally.  相似文献

1. The HGPRT (Hypoxanthine‐Guanine‐Phospho‐Ribosyl‐Transferase) assay with Chinese Hamster V79 cells was used to measure the mutagenic effects of UV irradiation, potassium dichromate and nickel chloride. The agents were tested separately and in the combinations of UV plus nickel and dichromate plus nickel.

2. UV, Cr(VI) and Ni(II) were confirmed to be mutagenic in the V79 cell assay. The combination of UV(5J/m2) and Ni(II) (0.5 mM) caused a mutation rate 11.2 times above that corresponding to the sum of the individual mutation rates of these agents. The combined action of Cr(VI) (0.1 mM) and Ni(II) (0.5 mM) produced a mutation rate 2.8 fold above that corresponding to the sum of the individual rates of the separate agents.

3. The enhancing effect of nickel chloride on the mutagenicity of UV or Cr(VI) is interpreted by an interference of Ni(II) with the repair of DNA lesions.  相似文献

2012年1月~2012年12月采集乌鲁木齐大气PM2.5样品,使用质粒DNA评价法研究了不同季节PM2.5的氧化能力,并进行氧化性毒性与相应气象因素和质量浓度之间的相关性研究.结果表明,乌鲁木齐大气PM2.5的质量浓度具有冬季最高,春季和秋季次之,夏季最低的季节性变化特征;PM2.5全样和水溶部分氧化能力的季节差异较大,对质粒DNA的氧化性损伤具有冬季最大,春季和夏季之次,秋季最低.冬、春、夏、秋季大气PM2.5全样的TD30(PM2.5对质粒DNA造成破坏达到30%所需要的颗粒物的剂量)平均值分别为440,491,503,515μg/mL,水溶部分分别为474,721,666,600μg/mL.绝大部分PM2.5样品全样的TD30值均小于水溶部分样,表明全样的毒性大于相应的水溶部分样.全样TD30值与平均温度显著(P<0.05)正相关,表明寒冷的天气/季节可能造成PM2.5的高毒性.水溶样TD30值与风速显著(P<0.01)正相关,与相对湿度显著负相关.这表明,高的风速和低的相对湿度可能跟较低和较高的PM2.5的毒性有关.PM2.5氧化性损伤能力的大小与其质量浓度之间的相关性不明显,表明仅以颗粒物的质量浓度来评价大气颗粒物氧化性损伤能力大小的方法并不能真实地反映其对人体健康的危害程度,起决定作用的还是颗粒物的化学组成及其表面吸附的有害成分.  相似文献

随机选用了10条RAPD引物,采用3种商品TapDNA聚合酶体系,以5个不同物种的动植物总DNA为模板,进行RAPD扩增,实验发现,在对同一模板、采用同一引物进行的RAPD扩增中,仅仅因为使用了不同的商品TaqDNA聚合酶,获得的扩增产物差别极大;不同商品来源的PCR反应缓冲体系,对扩增产物也有一定影响,但相对较小,这说明,不同的商品酶体系对RAPD扩增产物影响很大,因此,影响RAPD扩增产物稳定性的一个关键因素是商品TaqDNA聚合酶本身;研究中前后一致地使用同一商品TaqDNA聚合酶体系,是成功进行RAPD分析的基础。  相似文献

Accidental spills and chronic leaks of fuel oil or other hydrocarbon material (e.g., coal tar) often result in subsurface accumulation of nonaqueous phase liquid (NAPL), which can be a subsequent source of contamination in groundwater. Linking hydrocarbons in groundwater to a source NAPL has been difficult when using standard target analytes (e.g., BTEX) because of differences in partitioning properties of the analytes between the source NAPL and groundwater. Because aqueous solubility is predicted to be the controlling influence in the partitioning of hydrocarbons from NAPL to groundwater, a solubility-based approach to matching dissolved hydrocarbons in groundwater to their source NAPL has been developed and validated for two sites with commonly encountered types of NAPL contamination. Specifically, a gasoline LNAPL and a coal tar DNAPL from two separate sites (West Virginia and California) and groundwater interfaced with these NAPLs were analyzed for approximately 50 gasoline-range hydrocarbons consisting of paraffin, isoparaffin, (mono-) aromatic, naphthene, and olefin compounds (PIANO). Solubility characteristics of selected alkyl aromatic hydrocarbons from the PIANO analysis were used to identify a set of diagnostic hydrocarbons, expressed as hydrocarbon ratios, which were found to be useful in distinguishing the source(s) of hydrocarbons in groundwater. At the West Virginia site, the diagnostic ratios in a downgradient groundwater sample were similar to those of a gasoline NAPL at that site, indicating the source of hydrocarbons to the groundwater was the upgradient gasoline NAPL. The diagnostic ratios of the groundwater in contact with the gasoline NAPL and the remote groundwater were also similar, providing evidence that the diagnostic ratios were retained during transport in the aquifer. At the California site, diagnostic ratios in a cross-gradient groundwater sample differed from those of the coal tar NAPL at that site, indicating that the remote groundwater hydrocarbons did not originate from the coal tar contamination. Environmental factors such as selective degradation of specific isomers and various geological conditions (e.g., soil mineralogy, and organic content) may confound the application of this solubility-based fingerprinting approach. Thus, it is recommended that multiple diagnostic pairs be simultaneously evaluated when considering this fingerprinting approach for specific sites and product types.  相似文献

于2010年12月以及2011年6月采集了大亚湾9个站点的表层水样,利用PCR-变性梯度凝胶电泳(PCR-DGGE)技术对浮游细菌DNA指纹进行了分析.结果显示,大亚湾海域浮游细菌DNA指纹比较丰富,冬季和夏季样品中分别共得到32条和33条条带,每个样品得到的条带数为18~25条.DNA指纹的Shannon-Weaver多样性指数(H')和Pielou均匀度(J)的变化范围分别为2.48~2.88和0.80~0.93.DNA指纹条带数与温度和盐度明显正相关,而与p H、DO、透明度呈明显负相关关系.结果说明PCR-DGGE技术可以在一定程度上反映细菌的群落结构和多样性,但需要与其他技术相结合方能较全面反映细菌的结构与功能.  相似文献

The objective of the study was to evaluate the potential risk of DNA damage due to exposure to a mixture of the most widely used pesticides, namely endosulfan, chlorpyriphos and thiram at an environmentally relevant concentration (5 μM each) and the DNA protective capacity of sulforaphane (SFN) (10–30 μg/mL). DNA damage in human lymphocytes was ascertained with Single Cell Gel Electrophoresis (SCGE), also called Comet Assay. For positive control, H2O2 at 100 mM was used. The pesticide mixture produced DNA damage at the concentration used in the lymphocytes. SFN was able to offer a statistically significant (P < 0.01), concentration-dependant protection to DNA damage between 10–20 μg/mL in both the pre-incubation and co-incubation strategies. The results indicate that exposure to low levels of these pesticide mixtures can induce DNA damage, and the presence of SFN in diet may reduce the incidence of genetic damage, especially in farm workers. However, it is not clear whether SFN is involved in quenching of the free radicals generated by the pesticide mixture or it is involved in DNA repair mechanism.  相似文献
