通过分子克隆技术,从费氏中华根瘤菌(Sinorhizobium fredii)HN01基因组文库中克降到一个lrp基因.该基因与已报道的苜蓿中华根瘤菌(Sinorhizobium meliloti)1021的lrp基因在核苷酸水平上有89%相似性,在氨基酸水平上有99%相似性.利用自杀质粒pK18mob构建含有lrp基因部分片段的重组质粒,通过三亲本结合后导入原始菌株HN01中,经过同源单交换,获得发生正向插入突变的突变株GXHNLTA和反向插入突变的突变株GXHNLTB.将lrp基因ORF连接到载体pLAFR3上,获得用于互补的质粒pGXHNL100,将该质粒通过三亲本接合导入突变株中,获得互补株GXHNWA、GXHNWB.对野生型菌株、突变株、互补株进一步研究发现,在以脯氨酸、亮氨酸、丝氨酸等氨基酸为唯一碳、氮源的MM培养基中培养时,突变体GXHNLTA、GXHNLTB的生长均滞后于出发菌株HN01.植株试验表明,突变菌株GXHNLTA、GXHNLB比野生菌株HN01开始结瘤的时间提前1 d,在结瘤效率、单株瘤数、瘤重、固氮酶活性方面并无显著差别.  相似文献

以赤潮微藻米氏凯伦藻(Karenia mikimotoi)和塔玛亚历山大藻(Alexandrium tamarense)为受试对象,对一株芽孢杆菌WZ01所产的生物絮凝剂去除赤潮微藻的性能进行了研究。结果表明:芽孢杆菌WZ01所产生物絮凝剂对人工培养的米氏凯伦藻和塔玛亚历山大藻藻液均有去除能力,适宜的絮凝剂质量浓度为40.0~60.0 mg/L,絮凝时间为90~120 min,pH=7.5~8.5,在20~40℃范围内生物絮凝剂对2种微藻的絮凝率无显著区别;Ca~(2+)和Mg~(2+)浓度对生物絮凝剂去除2种微藻有一定影响,适宜浓度为4.0~6.0 mmol/L;适宜条件下生物絮凝剂对人工培养的米氏凯伦藻和塔玛亚历山大藻藻液的絮凝率均超过80.0%。研究表明,芽孢杆菌WZ01所产的生物絮凝剂在去除赤潮微藻方面有应用潜力。  相似文献

Stopping declines in biodiversity is critically important, but it is only a first step toward achieving more ambitious conservation goals. The absence of an objective and practical definition of species recovery that is applicable across taxonomic groups leads to inconsistent targets in recovery plans and frustrates reporting and maximization of conservation impact. We devised a framework for comprehensively assessing species recovery and conservation success. We propose a definition of a fully recovered species that emphasizes viability, ecological functionality, and representation; and use counterfactual approaches to quantify degree of recovery. This allowed us to calculate a set of 4 conservation metrics that demonstrate impacts of conservation efforts to date (conservation legacy); identify dependence of a species on conservation actions (conservation dependence); quantify expected gains resulting from conservation action in the medium term (conservation gain); and specify requirements to achieve maximum plausible recovery over the long term (recovery potential). These metrics can incentivize the establishment and achievement of ambitious conservation targets. We illustrate their use by applying the framework to a vertebrate, an invertebrate, and a woody and an herbaceous plant. Our approach is a preliminary framework for an International Union for Conservation of Nature (IUCN) Green List of Species, which was mandated by a resolution of IUCN members in 2012. Although there are several challenges in applying our proposed framework to a wide range of species, we believe its further development, implementation, and integration with the IUCN Red List of Threatened Species will help catalyze a positive and ambitious vision for conservation that will drive sustained conservation action.  相似文献

The human rights related controversies surrounding Anglo Platinum's Mogalakwena mine in South Africa's Limpopo Province hit the world stage in 2008, attracting public scrutiny and instigating much debate in the realm of international business and human rights discussions. We provide an in-depth analysis of the controversies at Mogalakwena, and Anglo Platinum's responses. This case study contributes to ongoing debates on mining company-community relations in the context of the emerging business and human rights agenda, specifically with respect to the notion of human rights ‘due diligence’. We argue that the Mogalakwena case study illustrates the risks of companies adhering to and emphasizing the technical and logistical facets of due diligence, without sufficient attention to the relational, communicative and emergent aspects, which are closely tied to corporate culture. We highlight the complexities of company-community relationships, with particular reference to the issues of legitimate community representation and participatory decision-making, and discuss the role that organizational culture plays in exacerbating or mitigating corporate-community conflict. The case study demonstrates how legal challenges often exacerbate rather than resolve the conflicts that arise due to a clash between corporate and community cultures. We suggest the need for corporate leaders to become more conscious of this cultural dimension of effective social management, particularly when interacting with communities whose cultures are markedly different from those of business corporations.  相似文献

We use a relatively new approach to endogenously detect the volatility shifts in the returns of four major precious metals (gold, silver, platinum and palladium) from 1999 to 2013. We reveal that the turbulent year of 2008 has no significant effect on volatility levels of gold and silver however causes an upward shift in the volatility levels of palladium and platinum. Using the consistent dynamic conditional correlations, we show that precious metals get strongly correlated with each other in the last decade which reduces the diversification benefits across them and indicates a convergence to a single asset class. We endogenously detect the shifts in these dynamic correlation levels and reveal uni-directional volatility shift contagions among precious metals. The results show that gold has a uni-directional volatility shift contagion effect on all other precious metals and silver has a similar effect on platinum and palladium. However, the latter two do not matter in terms of volatility shift contagion. Thus, investors that hedge with precious metals should, in particular, monitor the volatility levels of gold and silver.  相似文献

Location matters for the value of capital assets. The value of changes in natural capital wealth can depend on whether natural capital asset prices are measured locally and then aggregated or whether average values are applied over aggregate representative areas. Spatial heterogeneity of resource characteristics and institutions impact approximations of the intertemporal welfare function and accounting price function because when spatial aggregation precedes valuation it implies greater arbitrage opportunities leading to more inelastic shadow (accounting) price functions than when valuation is done locally and then aggregated. Aggregation of observed values across varying resource and institutional characteristics can lead to omitted variables bias. We illustrate these results in the context of groundwater in the Kansas High Plains Aquifer and demonstrate that the accounting price function is less elastic when the accounting price is measured locally. Failure to measure locally and then aggregate could lead to undervaluing scarce resources and overvaluing plentiful ones, which biases wealth accounts in favor of passing the non-declining wealth sustainability test.  相似文献

从采集的多种土壤样品中.获得了19株在3种筛选平板上均生长良好的候选氯氰菊酯降解菌.通过生理生化特性鉴定和16S rDNA序列的测定及比对.将其中的氯氰菊酯降解菌CY22-7鉴定为中华根瘤菌属(Sinorhizobium sp.).同时,对氯氰菊酯降解菌CY22-7的降解特性研究结果表明:(1)氯氰菊酯降解菌CY22-7以5%(体积分数)的接种鼍接种到氯氰菊酯起始质量浓度为100 mg/L的氯氰菊酯乙醇培养基后,于200 r/min、30℃摇床中培养.经过6 d的培养,氯氰菊酯降解菌CY22-7降解了约60%的氯氰菊酯.(2)加人外源营养物质有利于促进氯氰菊酯的降解.其中,葡萄糖和酵母提取物的促进作用最为明显.(3)氯氰菊酯降解菌CY22-7降解氯氰菊酯的最适温度为30℃,最佳pH为6.0.  相似文献

采用铝基无机有机复合絮凝剂(PAC-JY01)对含有腐殖酸与高岭土的模拟水样进行强化混凝处理,取混凝后出水进行超滤,通过测定超滤前后出水水质以及超滤膜运行状况,探讨PAC-JY01对超滤膜工艺效能的影响.结果表明,在强化混凝-超滤联用工艺中PAC-JY01的最佳投加量为3mg/L,最佳pH=6.在最佳条件下,UV254和浊度的去除率分别达到79.30%、99.70%;在实验运行条件下,较大的絮体容易在膜表面沉积而造成膜污染.  相似文献

将从自杀性重组质粒pDB30上分离的3.6kb含卡那霉素抗性基因和发光酶基因(luxAB)的BglⅡ酶切片段,克隆到小Mr的高拷贝质粒pIJ2925上,构成新的质粒,pHNLX1011;然后将pHNLX1011经BglⅡ部分酶切,酶连在含蔗糖酶基因(sacB)的自杀性重组质粒载体pMH1701上,载体分别采用BglⅡ酶切和BamHⅠ酶切.连接转化后共得到4种重组质粒类型的菌株,它们具有不同发光活性;将它们分别与快生型大豆根瘤菌HN01进行三亲本杂交,发现它们的转座频率均较pDB30高.对经转座标记后的根瘤菌进行发光活性检测,只有pHNC3质粒所转座的根瘤菌浇具有较强发光活性.所得发光根瘤菌衍生菌株能在野大豆和栽培大豆黑农39根际形成正常根瘤,能用X光片记录下根瘤的发光活性.  相似文献
