Photochemical reaction of particle-bound 1-nitropyrene under simulated atmospheric condition

Photoreaction of particle-bound 1-nitropyrehe (1-NP) under simulated atmospheric condition has been studied. When 1-NP coated on the surface of silica gel was suspended in a stream of nitrogen, oxygen or air, respectively, and exposed to intensive light of xenon lamp, photochemical reaction took place. The result of kinetic study showed it to be of the second order. The photoreaction products were identified by off-line HPLC/MS as 1,6-pyrenequinone, 1,8-pyrenequinone, trinitro-hydroxypyrene, dinitrohydroxypyrene, 6,6'- dipyrenenone, etc. The mutagenicity of photochemical reaction products is higher in oxygen and air, and lower in nitrogen than that of the precursor 1-NP, as detected by the Ames Bioassay.     

Prenatal diagnosis of alpha-1-antitrypsin deficiency by PCR of linked polymorphisms: A study of 17 cases

We describe our experiences of prenatal diagnosis of alpha-1-antitrypsin deficiency using closely linked polymorphisms detected by Southern blotting and the polymerase chain reaction. Prenatal diagnosis was carried out for 17 pregnancies from 15 couples.

Variation in the levels of pregnancy- specific β-1-glycoprotein in maternal serum from chromosomally abnormal pregnancies

Human pregnancy-specific β-1-glycoprotein (SP1) was assayed retrospectively in stored maternal serum (MS) samples from 82 chromosomally abnormal pregnancies and 377 matched controls. The median MSSP1 concentration in 48 Down's syndrome pregnancies was significantly elevated at 1.17 multiples of the control median (MOM), and significantly reduced (0.5 MOM) in a group of eight cases of unbalanced translocations. There was no significant difference in median SP1 concentrations in cases of trisomy 18, trisomy 13, balanced translocations, or sex chromosome abnormalities. A comparison with human chorionic gonadotrophin results in the same series of samples indicates that SP1 is a less sensitive predictor of Down's syndrome pregnancies.     

Fumarylacetoacetase activity in cultured and non-cultured chorionic villus cells,and assay in two high-risk pregnancies

We describe further development of the fumarylacetoacetase (FAA) assay for the prenatal diagnosis of tyrosinaemia type 1 using chorionic villus sampling (CVS). We have established a reference range for FAA activity in cultured villus cells and have confirmed previously reported data on the FAA activity in uncultured chorionic villus cells. This should allow confirmation of results using CVS, without the need for further invasive procedures. We report the FAA enzyme stability at −70°C, +4°C, and at room temperature, and we have shown no obvious difference in enzyme activity with gestational age. We have analysed cultured and non-cultured CVS activity of FAA in two pregnancies at risk of tyrosinaemia type 1. In both, the fetus was designated unaffected, and these results were confirmed postnatally.     

Prenatal testing for inherited immune deficiencies by fetal blood sampling

We attempted to develop a prenatal diagnosis in fetuses at risk for immunodeficiency by fetal blood sampling performed under fetoscopy at 18–22 weeks of gestation. In order to obtain normal values, we first investigated thirty-five control fetuses whose blood punctures were undertaken for the diagnosis of haemoglobinopathies. Surface markers and in vitro mitogen-induced proliferation of the fetal lymphocytes were studied using micromethods. We then examined two fetuses at risk for two different types of severe combined immunodeficiency and established their immunological integrity, hence avoiding an unjustified termination of pregnancy. This immunological integrity was confirmed after birth.
