To investigate the bacterial community structure features of soak solutions used to preserve bamboo slips that were excavated from Han dynasty tomb located in Laoguanshan of Chengdu and to reveal the diversity of bacteria in these soak solutions, PCR-DGGE was employed. Subsequently, the major DGGE bands were excised and sequenced to analyze the phylogeny of bacteria. The richness (S), Shannon-Wiener index (H), and Simpson index (D) of deionized water (0#) without the soaked bamboo slips were higher than those of the other samples. Among the bamboo slip soak solution samples, there were significant differences in these indicators; the bacterial genetic diversity of sample 121# was the highest and that of sample 1# was the lowest. Principal Component Analysis (PCA) showed that there were comparatively large differences among the samples, and the similarity between sample 1# and others was the lowest. Based on the sequence analysis, the major community of bacteria in soak solution were belonged to Alphaproteobacteria, Betaproteobacteria, Gammaproteobacteria, Firmicutes, Bacteroidetes, and Actinobacteria, including Cupriavidus, Aquabacterium, Comamonas, Albidiferax, Hyphomicrobiaceae, Azospirillum, Nevskia, Streptococcus, Staphylococcus, Sediminibacterium, and Propionibacterium, among which Cupriavidus of the β-Proteobacteria class was detected in all samples. The bacterial community structure of the soak solutions that were collected from different bamboo slips was quite complex and significantly different. The analysis of the main bacterial community revealed the potential bacteria species that may trigger the damage in bamboo slips; the result provided a reference to prevent waterlogged bamboo slips from microbial diseases in the future. © 2018 Science Press. The microbial community structures of two mesophilic anaerobic chemostats, one fed with glucose, the other with starch as sole carbon sources, were studied at various dilution rates (0.05–0.25 d–1 for glucose and 0.025–0.1 d–1 for starch) during two years continuous operation. In the glucose-fed chemostat, the aceticlastic methanogen Methanosaeta spp. and hydrogenotrophic methanogen Methanoculleus spp. predominated at low dilution rates, whereas Methanosaeta spp. and the hydrogenotrophic Methanobacterium spp. predominated together when dilution rates were greater than 0.1 d–1. Bacteria affiliated with the phyla Bacteroidetes, Spirochaetes, and Actinobacteria predominated at dilution rates of 0.05, 0.1, and 0.15 d–1, respectively, while Firmicutes predominated at higher dilution rates (0.2 and 0.25 d–1). In the starch-fed chemostat, the aceticlastic and hydrogenotrophic methanogens coexisted at all dilution rates. Although bacteria belonging to only two phyla were mainly responsible for starch degradation (Spirochaetes at the dilution rate of 0.08 d–1 and Firmicutes at other dilution rates), different bacterial genera were identified at different dilution rates. With the exception of Archaea in the glucose-fed chemostat, the band patterns revealed by denaturing gradient gel electrophoresis (DGGE) of the microbial communities in the two chemostats displayed marked changes during long-term operation at a constant dilution rate. Antibiotics are newly emerging organic pollutants in manure, soil, vegetables and water. Animal manure application might be leading to the accumulation of antibiotics in the farmland. However, the effect of sulphamethazine (SMZ) on the soil microbial community was scarcely investigated. This study was aimed to evaluate the impact of SMZ on poultry manure, on the structure and function of microbial community, carbon mineralisation, and changes in nitrogen forms in soil via an incubation experiment lasting 56?d. The treatments consisted of poultry manure at 1% wt (PM), PM containing 20?mg?kg?1 SMZ (PM?+?20SMZ) and PM containing 100?mg?kg?1 SMZ (PM?+?100SMZ), along with the untreated soil (control). Solid phase extraction was performed to measure the SMZ concentration in soils using high-pressure liquid chromatography. The cumulative CO2-C was increased in all treated soils over the incubation period compared to the control. The PM?+?100SMZ had the highest increase in cumulative CO2-C from the soil at 56?d of incubation. The treatment of PM?+?20SMZ showed a short-term decrease in nitrification rate in the soils at 1?d by altering the microbial community composition with 17% dissimilarity and decreasing the abundance of bacteria compared to PM-treated soil. A new approach was developed to evaluate the implications of the spatiotemporal variability of green vegetation for the dispersion of livestock that is required to access quality forage in semi-arid Africa. Maximum NDVI (Normalized Difference Vegetation Index) at 1 km2 resolution was determined for concentric rings (0–31 km radii) around 227 individual sample locations within the study area for 14 dates (between 1 April to 1 November) annually over the 2000–2010 period. A sigmoidal curve was fitted to points within the maximum NDVI × distance radii space to determine the asymptote distance (AD) – the radius at which further dispersion from the sample location does not lead to significant gains in access to green forage. AD was found to: increase with latitude (or increasing aridity); decline as the rainy season proceeds; and show no trend over the 2000–2010 period. These results introduce much-needed empirical data to current debates surrounding the scales of governance to support livestock mobility. 相似文献

为研究半干旱黄土区高效植被恢复途径,选取陕西省吴起县境内的合沟封禁流域和柴沟人工造林流域为研究区,在两流域内,沿顺坡和横坡方向,选择一系列20 m×20 m的样地,各组成2条样带。在每个样地内的对角线交点和对角线四分之一处选取5个样方进行植被调查,草本样方和乔灌样方的尺寸分别为1 m×1 m和5 m×5 m。共获得草本样方185个、乔灌木样方80个,其中,合沟封禁流域105个,均为草本样方,柴沟人工造林流域草本样方与乔灌样方各80个。运用植物生态学和植物群落学方法,调查各样方内的植物种类、株数、株高、盖度、地上生物量以及树木基茎、树高、冠幅等植被群落特征指标,并在此基础上对两流域的植被群落特征进行对比分析。结果表明:(1)两流域的植被种类共61种,其中柴沟人工造林流域46种,合沟封禁流域51种,共存物种36种;(2)经过10年生态恢复,柴沟人工造林流域已形成乔-灌-草群落,合沟封禁流域仍为草本群落,且两流域草本群落均以铁杆蒿(Artemisia sacrorum Ledeb.)和茭蒿(Artemisiagiraldii Pamp.)为主要建群种;(3)柴沟人工造林流域的植被盖度、平均高、生物量均明显高于合沟封禁流域的相应指标。两流域植物群落Simpson指数和Shannon-Weiner指数差异较小,Sorensen指数为0.742,主要草本植物种的指标差异不显著。以上特征反映出,在半干旱黄土区,适宜的人工造林可加速该区植被恢复的进程。 相似文献

Land use conversion is an important factor influencing the carbon gas exchange between land and atmosphere. The effect of land use conversion on soil organic carbon mineralization and microbial function is important for soil organic carbon sequestration and stability. This research studied the effects of land use conversion on soil chemical properties, organic carbon mineralization and microbial community structure after two years of conversion from double rice cropping (RR) to maize-maize (MM) and soybean-peanut (SP) double cropping systems in southern China. The results showed that soil pH significantly decreased by 0.50 (MM) and 0.52 (SP, P = 0.002), and dissolved organic carbon significantly increased by 23%- 35% (P = 0.016). No significant difference was found in soil organic carbon mineralization rate with the land use conversion, though the accumulated mineralization decreased after 13 days of incubation (P = 0.019). Land use conversion from paddy to upland significantly changed soil microbial community structure. The total PLFAs, bacterial, gram-positive bacterial (G+), gram-negative bacterial (G-) and actinomycetic PLFAs decreased significantly (P < 0.05), the ratio of fungal PLFAs to bacterial PLFAs (F/B) increased significantly (P = 0.006). But no significant differences in microbial groups were found between MM and SP. The accumulated mineralization at the beginning period of the incubation were significantly positively correlated with soil actinomycetic PLFAs (P = 0.034). After 13 days of incubation, soil F/B showed a positive correlation with the accumulated mineralization (P = 0.004). However, soil microbial community structure(P = 0.014)and total PLFAs(P = 0.033)showed a positive correlation with the accumulated mineralization after 108 days of incubation. Our results indicated that after conversion from paddy soils to drained soils, soil pH and total nitrogen are the key factors regulating the variations in soil microbial community structure and biomass, and then influencing soil organic carbon mineralization. 相似文献

通过在实际重力流排水管道中选取10个典型的腐蚀区域作为研究对象,通过对排水管道对应腐蚀区域的生物膜和水体进行采样,探究实际排水管道不同空间位置上微生物群落分布的差异性以及水质因素对微生物群落多样性分布影响.采用高通量测序方法反映腐蚀区域微生物群落的多样性.结果表明,在排水管道中10个典型腐蚀区域中的微生物群落分布结构基本相似,在门水平上主要是变形菌占绝对优势,其次是放线菌门,在纲水平上主要是a-变形菌纲,其次是β-变形菌纲两者相对含量之和占到43.56%,在属水平上的硫酸盐还原菌对管道具有严重的局部腐蚀,包含有Desulforhabdus,Desulfuromonas,Desulfobacter.RDA环境影响因子研究发现,在门水平上硫化氢、溶解氧与微生物群落存在一定的相关性,在纲水平上与微生物群落存在相关性的主要是铵根离子、pH值和COD. 相似文献

2010年12月、2011年1月、2月、3月、5月和8月,利用样带法对珠海市鹤洲湿地鸟类物种多样性进行研究,利用Shannon-Wiener指数分析了生境类型的鸟类群落种数、数量、物种多样性、均匀性指数等特征。结果表明,共记录到96种,隶属12目31科66属。鸟类多样性指数(H')进行了计算,12月3.6956,1月3.7685;2月3.6551;3月3.4703;5月3.2437和8月3.1941。该湿地鸟类多样性动态表现出明显的季节性。 相似文献

对富营养化水体常见浮游植物进行了分离和培养,测定了其磷脂脂肪酸(PLFA)。同时,检测了惠州西湖浮游植物的磷脂脂肪酸;结合惠州西湖浮游植物群落组成数据,分析了浮游植物磷脂脂肪酸特征及其与浮游植物群落结构的关系。结果表明:结合PLFA与细胞丰度和生物量的分析,α-亚麻酸(18:3ω3)的质量浓度与蓝藻(除湖丝藻外)呈正相关;二十碳四烯酸ARA(20:4ω6)和二十二碳六烯酸DHA(22:6ω3)的质量浓度与硅藻呈正相关;二十碳五烯酸EPA(20:5ω3)和16:3ω3的质量浓度与绿藻呈正相关等,其中16:3ω3为绿藻门独有的脂肪酸。这些均与室内实验得到的结果相符合。本研究表明,磷脂脂肪酸可以作为生物标志物来分析浮游植物组成,它将是一种可行的研究浮游植物群落结构的新方法。 相似文献
