An increasing number of industrial, agricultural, and commercial chemicals in the aquatic environment leads to various deleterious effects on organisms, which is becoming an increasingly serious problem in China. In this study, the comet assay was conducted to investigate the genotoxicity to human body caused by organic concentrates in the drinking water sources of Nanjing City from Yangtze River of China, and health and ecology risk due to expose to these organic pollutants were evaluated with the multimedia environmental assessment system (MEAS). For all the water samples, they were collected from four different locations in the drinking water sourcr samples, es of Nanjing City. The results of the comet assay showed that all the organic concentrates from the water samples could induce different levels DNA damages on human peripheral blood lymphocytes, and a statistically significant difference (p〈0.01) was observed compared with the solvent control, which demonstrated the genotoxicity was in existence. According to the ambient severity (AS) of individual compound, we had sorted out the main organic pollutants in the drinking water source of the four waterworks, and the results showed that there was some potential hazard to human body for all the source water, namely the total ambient severity (TAS) of health for each water source was more than 1. However, the TAS of ecology for each water source was less than 1, which indicated that it was safe to ecology. The results of this investigation demonstrate the application of the comet assay and the MEAS in aquatic environmental monitoring studies, and the comet assay found to be fast, sensitive, and suitable for genotoxicity monitoring programs of drinking water source.  相似文献

Abstract

Paclobutrazol (PBZ) is a plant growth regulator (PGR) widely used in fruit and vegetable cultivation. However, due to the severe toxicity of PBZ, a sub-ppm level maximum residue limit (MRL) was established worldwide. Therefore, it is significant to propose a rapid, sensitive and high throughput screening method for monitoring the PBZ residues in foods. In this study, a simple and sensitive indirect competitive Enzyme-linked immunosorbent assay (icELISA) was established for PBZ detection in fruits basing polyclonal antibody. For both economy and pollution prevention, a microwave-solvent-free method was used to synthesize the PBZ hapten with high efficiency. The detection conditions, such as coating antigen concentration, antibody concentration, organic reagent concentration, ionic strength and pH, were optimized. Under the optimized conditions, this method showed high sensitivity and specificity. The detection range is 1.27-138.23?ng/mL, half-maximum inhibition concentration (IC50) is 13.26?ng/mL, and the IC20 was lower than the reported ELISAs for PBZ. Additionally, this method had high accuracy and precision. The recoveries were ranged from 88.78% to 96.80% in PBZ spiked apple samples with RSD below 4%. All the results showed that the polyclonal antibody based icELISA could be useful for PBZ screening in fruit samples.  相似文献

为探索一种简便、直接检测环境诱变物对人体靶细胞遗传毒性的方法,笔者以甲醛作为染毒剂,对人体口腔颊黏膜细胞彗星实验的染毒时间、染毒温度和2种用于检测交联作用的方案进行了研究。结果显示:经7 5μmol L甲醛37℃染毒30和60min后,DNA的断裂程度较染毒15min更大;而经该浓度甲醛在4,23和37℃下染毒30min后均可引起DNA的断裂,但37℃下染毒对DNA的断裂作用更大。在2种应用于检测交联作用的方案中,加大电泳条件的交联检测方案还可检测兼具交联和断裂效应的诱变物。   相似文献

甲醛诱导Hela细胞凋亡的研究   总被引:3,自引:1,他引:2  
细胞凋亡是当今环境毒理学领域的研究热点之一.为了探讨甲醛能否诱导细胞凋亡,以Hela细胞为实验材料,通过不同浓度的甲醛处理24h后,应用形态学观察、四甲基偶氮唑蓝(MTT)实验和流式细胞仪检测了Hela细胞凋亡情况.实验结果表明低浓度甲醛(≤0.2mmol·L-1)处理Hela细胞24h后,对细胞生长有显著的抑制作用(p<0.05),但对细胞形态改变不大,且对细胞核DNA没有损伤;而高浓度甲醛(≥0.4mmol·L-1)处理Hela细胞24h后,对细胞生长有极显著的抑制作用(p<0.01),细胞形态逐渐变的模糊不清,并有少量细胞萎缩成圆形,且会造成细胞核DNA有规律的断裂,细胞凋亡率随甲醛浓度的增加而增大.以上实验结果显示,高浓度甲醛处理Hela细胞24h后,会诱导细胞凋亡;并且随甲醛浓度的升高凋亡率逐渐增大,呈一定的剂量-效应关系.  相似文献

采用鼠伤寒沙门氏菌回复突变试验(Ames试验),用TA97,TA98,TA100和TA102菌株,加与不加S9,剂量分别设为每皿1000μg,500μg和250μg;小鼠骨髓多染红细胞微核试验,剂量设为雄性、雌性小鼠给药剂量均为250,500和1000mg*kg-1;小鼠睾丸精母细胞染色体畸变试验,剂量设为625mg*kg-1,1250mg*kg-1和2500mg*kg-1.结果表明,Ames试验中,各测试浓度的诱发回变菌落数均未超过自发回变菌落数的2倍;小鼠骨髓多染红细胞微核试验和小鼠睾丸精母细胞染色体畸变试验,各剂量组和溶剂对照组的微核率进行统计学处理,未见有显着性差异(P>0.05),表明各项试验结果均为阴性.因此,己酸二乙氨基乙醇酯无致突变作用.  相似文献

A simple method – direct agar diffusion assay – was optimised for rapid assessment of heavy metal toxicity to marine chromogenic and non-chromogenic bacteria. The procedure involved spotting of a 10 microliter test solution on the seeded agar plate and incubation of the plates at 30°C to accelerate bacterial growth. Under optimum conditions, test results were obtainable within 12–18 hr instead of 96 hr incubation time generally required for a marine bacterial assay by conventional agar plate methods. A range of sixteen heavy metals, each at 5 different concentrations was tested. Toxicity was demonstrated by the formation of a clear zone of growth inhibition around the point of application. Toxicity of tested chemicals could be easily demonstrated at concentrations as low as 0.1 g per spot on the agar plate. A dose dependent relation between metal concentration (g/spot) and the diameter of the clear zone on agar plate was observed, suggesting potential of this method as an easy and economical tool in quantitative toxicology studies.  相似文献

Effluents from wastewater treatment plant (WWTP) have been reported to have a broad spectrum of endocrine disrupting compounds (EDCs). The majority of studies have focused on the occurrence of estrogenic activity, while ignoring nuclear hormone receptors (NRs) pathways. In the present study, a battery of in vitro yeast bioassays and a cell bioassay, including antagonistic and agonistic effects on estrogen receptor (ER), androgen receptor (AR), progesterone receptor (PR), estrogen- related receptor (ERR) and aryl hydrocarbon receptor (AHR), were conducted to evaluate the removal efficien- cies of EDCs by different treatment processes of a WWTP located in Beijing. Estrogenic, anti-estrogenic, anti- androgenic, anti-progesteronic, anti-ERR and the activa- tion of AHR activities were detected in samples from all treatment processes and the receiving water. The concen- tration of estrogenic contaminants with estradiol (E2) equivalent concentrations ranged from 0.82 x 10-9 to 3.54 x 10 9g Ee_EQ.L-1. The concentration of anti-estrogenic contaminants with 4-hydroxytamoxifen (4-OHT) equiva- lent concentrations ranged from 1.24 × 10-6 to 2.36 x 10-6 g 4-OHT-EQ.L-1. The concentration of anti-androgenic contaminants ranged from 2.21 x 10-s to 3.52 × 10-6g flutamide-EQ. L-1. The concentration of anti-progesteronic contaminants ranged from 3.15 x 10^-5 to 2.71 x 10^-4g RU486-EQ. L-1. The concentration of anti-ERR contami- nants ranged from 7.09 x 10-5 to 6.50 x 104 g 4-OHT-EQ × L^-10. The concentration of AHR activators ranged from 1.7 × 10-10 to 3.4 × 10^-10g TCDD-EQ-L-1. These processes including secondary clarifier, coagulation, as well as coal and sand filtration could eliminated 67.2% of estrogenic contaminants, 47.0% of anti-estrogenic contaminants, 98.3% of anti-androgenic contaminants, 88.4% of anti- progesteronic contaminants, 65.4% of anti-ERR contami- nants and 46.9% of AHR activators. WWTP effluents contain multiple receptor disruptors may have very complex adverse effects on exposed organisms.  相似文献

卤代乙酰胺类(HAc Am)消毒副产物(DBPs)是饮用水中新兴含氮类DBPs之一。为探究HAc Ams的致突变性,选择小鼠淋巴瘤细胞作为受试细胞,分别考察了一氯代乙酰胺、二氯代乙酰胺、三氯代乙酰胺、一碘代乙酰胺、二碘代乙酰胺(DIAc Ams)共5种HAc Ams对小鼠淋巴瘤细胞Tk基因突变的影响。结果表明,在0.05~20μmol·L~(-1)暴露浓度下,5种HacAms类DBPs对小鼠淋巴瘤细胞呈现出一定的细胞毒性,随着暴露浓度的升高,细胞毒性增强。暴露4 h后,5种HacAms中只有DIAc-Ams呈现出显著的致突变性(暴露剂量高于5μmol·L~(-1)),且以小集落为主。上述研究结果为研究HacAms对哺乳动物细胞的致突变作用提供了基础数据。  相似文献
