为了进一步探索种群水平的生态风险评估方法,本文利用β-N-Acetyl-D-glucosaminidase(NAGase)的变化量来监测农药对摇蚊种群发育的影响。从花翅摇蚊Chironomus kiiensis体内分离纯化得到电泳纯的NAGase,并通过免疫大白兔制得NAGase的多克隆抗体。运用间接非竞争ELISA法检测抗体特异性,结果表明其与共同存在于水体的一些生物的NAGase的交叉反应率为隆线溞4.41%、老年低额溞3.12%、多刺裸腹溞3.40%、中华薄壳介4.17%、日本沼虾3.23%、白纹伊蚊7.50%、小球藻0.5%。运用抗体测得毒死蜱、氰戊菊酯和阿维菌素3种杀虫剂对于摇蚊NAGase释放量的12 d-EC50分别为1.2012、0.0043和0.6281μg·L-1,以NAGase活力作为测试终点,测得相应的12 d-EC50:1.4765、0.0051和0.6756μg·L-1,两者差异不显著,但均显著低于以死亡作为测试终点的12 d-LC50:4.8171、0.0954和2.1340μg·L-1,且毒力大小均为氰戊菊酯阿维菌素毒死蜱。上述结果表明,利用NAGase多克隆抗体可以特异性地检测农药对花翅摇蚊种群发育的影响。  相似文献

Laboratory studies were conducted to determine the adsorption/desorption equilibrium of carbofuran between four divergently textured soils and distilled water and an aqueous solution of 0.01?mol?L?1 β-cyclodextrin using a batch equilibrium technique. The determined adsorption isotherms for silt loam soils were of L-shaped, for sandy loam soil S-shaped, in agreement with a Freundlich isotherm. In the presence of β-cyclodextrin, the adsorption of carbofuran to the four soils was lower than with distilled water. Positive hysteresis was observed in all soils with distilled water, negative hysteresis when using a solution of β-cyclodextrin as desorbent. The results indicate the potential use of β-cyclodextrin for remediation of pesticide-contaminated soils.  相似文献

制备了不同β-环糊精(β-CD)含量的醋酸纤维素(CA)纳米纤维膜,并对纤维膜的结构形貌、尺寸分布、孔隙率、比表面积以及纤维膜的力学性能进行表征分析。随后研究了不同β-CD含量、初始浓度、吸附时间下,纳米纤维膜对甲基红染料的截留性能和吸附性能。结果表明:过量(≥30%)的β-环糊精会使纳米纤维产生珠节,纤维的形貌和力学性能变差;吸附试验表明,β-CD的加入提高了CA纳米纤维膜对染料的吸附,MR吸附率可达93.48%,最大平衡吸附量可达181.74 mg/g,比纯CA纳米纤维膜提高了将近40%;此外,染料的截留实验表明,纳米纤维膜对染料的截留效果较差,最大截留率为50.35%。  相似文献

Using the polymerase chain reaction (PCR), it was possible to amplify a single copy fragment of the β-globin gene from 2–32 human embryonic cells obtained from arrested preimplantation embryos. For the detection of β-thalassaemia mutations, allele specific priming of the PCR using nested primers was employed using approximately 10 pg of DN A from individuals known to carry these mutations. This approach was successful in detecting the presence or absence of five Asian Indian β-thalassaemia mutations that were selected for this study. In spite of meticulous precautions against contamination, false-positive amplification was observed, a problem that will have to be overcome before this approach can be used in clinical practice.  相似文献

为实现灵敏、快速、特异性地检测水环境中的17β-雌二醇 (E2) ,以甘蔗渣衍生的碳量子点作为荧光信号,核酸适配体 (aptamer) 作为识别元素,构建了一种可以特异性检测E2的荧光探针,通过荧光强度的变化来定量检测E2并对检测效果进行分析。结果表明:核酸适配体能成功修饰在碳量子点的表面形成稳定的荧光探针;200 mg·L−1的碳量子点与1 μmol·L−1的aptamer为荧光探针的最佳构建比例;相对荧光强度与0~10 μg·L−1质量浓度的雌二醇成正比,且最低检测限为0.42 μg·L−1;该荧光探针可成功应用于水体中E2的检测,回收率为93.6%~106.5%。与传统的仪器检测方法相比,该荧光探针检测E2具有良好的选择性和重现性,还具有操作简单、成本低的优点。本研究成果可为核酸适配体构建的荧光探针在水环境检测中的推广应用提供参考。  相似文献

重量法是测量PM_(2.5)的传统手工经典方法,而β射线法加动态加热系统联用光散射法作为自动在线监测技术的常用方法被广泛使用。将5030-SHARP型β射线法加动态加热系统联用光散射法PM_(2.5)自动监测设备监测结果与手工法测量结果进行比对、分析,结果表明:β射线法与重量法监测结果具有一致性,测量结果不存在显著性差异;β射线法与重量法相对偏差值与温度、相对湿度、风速等气象因子的相关性不明显;β射线法与重量法监测结果的差异性与PM_(2.5)质量浓度、季节等条件有关;随着PM_(2.5)质量浓度逐渐升高,β射线法与重量法相对偏差的绝对值|RD|也随之增大,不同季节的|RD|平均值差异较大,春季、冬季的|RD|平均值大于夏季、秋季。  相似文献

为了探讨污水灌溉与镉胁迫对菠菜抗氧化酶活性的影响,采用盆栽试验的方法,分别用清水和生活污水配制不同浓度(0、1、5、10、50mg·L-1)的Cd2+溶液,对菠菜进行受试处理,每5d灌喷1次,共处理5次.分别采用邻苯二酚比色法、L-苯丙氨酸比色法、水杨酸比色法和高锰酸钾滴定法,对菠菜叶片多酚氧化酶(PPO)、苯丙氨酸解氨酶(PAL)、β-1,3葡聚糖酶(β-1,3glucanase)和过氧化氢酶(CAT)活性进行了测定.结果表明:1)清水处理系列:随着镉处理浓度的增大,与清水对照相比,PPO、PAL活性呈先升高后降低的趋势,CAT活性略有升高,β-1,3葡聚糖酶活性略有降低,各暴露组与清水对照组相比,均呈现显著差异(p<0.05).2)污水处理系列:随着镉处理浓度的增大,与污水对照相比,PPO、PAL活性呈先升高后降低的趋势,CAT和β-1,3葡聚糖酶活性呈逐渐降低趋势;各暴露组与污水对照相比,均呈现显著差异(p<0.05).3)相同Cd2+浓度下,与清水处理相比,各浓度污水处理PAL和PPO活性均明显降低(p<0.05);而β-1,3葡聚糖酶明显升高(p<0.05).以上结果显示:单一镉污染和生活污水与镉复合污染对菠菜不同抗氧化系统酶具有不同影响,这可能与污水中各成分与镉的相互作用有关.  相似文献

The removal of 17β-estradiol (E2) in laccase catalyzed oxidative coupling processes was systematically studied in this work. We focused on the influence of pH and natural organic matter (NOM) on the performance of the enzymatic treatment processes. It was found that the optimal pH for E2 removal was between 4 and 6. The removal of E2 was slightly inhibited in the presence of NOM. Enzymatic transformation of E2 was second-order in kinetics with first-order to both the concentrations of the enzyme and contaminant. Mass spectrum (MS) analysis suggested that coupling products were formed through radical-radical coupling mechanism. The results of this study demonstrated that laccase catalyzed oxidative coupling process could potentially serve as a treatment strategy to control steroid estrogens.  相似文献

Hull-less barley is a special food crop rich in various nutrients in Qinghai-Tibet Plateau. Six-pearling is often used to produce commercial hull-less barley rice in order to improve its rough taste and inferior cooking quality. This study evaluated the differences in the nutritional, cooking, sensory, and storage qualities of hull-less barley rice with different pearling times to obtain suitable processing conditions for the production of high-quality hull-less barley rice. With increasing pearling time, the contents of vitamin B6, vitamin E, dietary fiber, iron, zinc, phenols, and flavonoids significantly decreased to a large extent, protein and vitamin B3 decreased slightly, the contents of total starch and β-glucan increased significantly, and γ-aminobutyric acid (GABA) initially increased and then decreased. The peak, trough, and final viscosity of whole grain barley were 1 860.50 cP, 914.50 cP, and 2 150.00 cP, respectively, and increased after six-pearling to 4 219.00 cP, 2 628.00 cP, and 5 074.00 cP, respectively. At the same time, the water absorption and volume expansion of the pearled hull-less barley increased significantly. The hardness of pearled hull-less barley reduced from 4 708.50 g to 2 282.00 g, its adhesiveness increased from 0.00 to -7.33, and its taste and sensory quality exhibited better. The activities of α-amylase, polyphenol oxidase, lipase, and catalase in pearled hull-less barley slightly decreased. Hull-less bran flour is rich in a variety of nutritional components and could be a potential resource with great developmental value. In general, hull-less barley rice obtained from three-pearling has high nutritional value, high cooking quality, low enzyme activity, and low energy consumption; therefore, it can be used to produce high-quality barley rice.
