Dispersion of atmospheric contaminants in the vicinity of an isolated cubical model building was investigated in the field. A dual source/receptor technique was used in the experiments, which was proved to be very useful for the investigation of pollution dispersion. This experimental technique involved the simultaneous release of two different tracer gases from two different point sources, and the deployment of a FID (Flame Ionization Detector) co-located with a UVIC®(Ultra-Violet Ion Collector) detector. Both mean concentrations and concentration fluctuation statistics were examined. In this paper concentration fluctuation statistics are presented. The effect of the upwind source location on intermittency values and on the cumulative density function (cdf) is examined. The exact location of a source placed upwind of an obstacle has a very significant and complex effect on both mean concentrations and concentration fluctuations. As the lateral or vertical displacement between the two sources is increased, cross-correlation values between data taken simultaneously by two co-located detectors decrease rapidly.  相似文献

以开滦矿区某矿8#孤岛煤柱回采为工程背景,针对孤岛工作面长度对底板垂直应力的分布影响及下伏大巷围岩稳定性问题展开研究。采用理论分析和数值模拟相结合的手段,建立了孤岛工作面在倾向方向上的底板受力模型,采用半无限体空间理论,推导出底板垂直应力计算公式,将该孤岛工作面实际地质参数带入公式,得出了孤岛工作面长度与底板应力之间的函数曲线。研究表明:当该孤岛工作面长60 m时,对下方-480 m水平大巷产生的垂直应力最小。通过FLAC 3D数值模拟方法验证了8#孤岛工作面合理尺寸(60 m)比实际孤岛工作面长度(100 m)对下方水平大巷的采动影响小,便于维护。  相似文献

The effects of osmotic stress on the ATPase activity, the contents of —SH group and conjugated polyamines in mitochondrial membrane from wheat seedling [ Triticum aestivum L. cv. Yumai No. 18 (drought-tolerant) and cv. Yumai No.9(drought-sensitive)] roots were investigated. The results showed that ATPase activity and --SH group content decreased with polyethylene glycol(PEG) 6000( - 0.55 MPa) treatment for 7 d, in concert with the decrease of the ratio of noncovalently conjugated spermidine (NCC-Spd)/noncovalently conjugated putrescine(NCC-Put) and increase of the covalently conjugated putrescine (CC-Put). Osmotic stress injury to Yangmai No.9 seedlings was alleviated greatly with 1 mmol/L exogenous spermidine(Spd), in concert with marked increases of the ratio of NCC-Spd/NCC-Put, --SH group contents and ATPase activity in mitochondrial membrane. Under osmotic stress, the concomitant treatment of Yumai No. 18 seedlings with methylglyoxyl bis(guanylhydrazone) (MGBG), an inhibitor of S-adenosyl methionine decarboxylase(SAMDC), and phenanthrolin ( o-Phen), an inhibitor of transglutaminase(TGase), caused a significant decrease of the ratio of NCC-Spd / NCC-Put, CC-Put contents, respectively, in concert with the marked decreases of ATPase activity, --SH group content and its tolerance to osmotic stress. All the results above suggested that osmotic stress tolerance of wheat seedlings was associated with the ATPase activity, the contents of --SH group, NCC-Spd and CC-Put in mitochondrial membrane.  相似文献

高层隔震结构的分析理论和设计方法是目前隔震技术向高层建筑推广的2个关键理论问题。本文以高烈度区宿迁市已经竣工的高层隔震建筑阳光大厦为工程实例,对高层隔震结构的非线性地震响应分析方法进行了研究,包括隔震支座竖向不同拉压刚度的模拟、各种类型的隔震支座水平力学特性的准确模拟,以及不同地震动输入方式和输入角度对隔震支座受拉情况的影响分析等;在此基础上,对隔震层的设计方法进行了分析,提出了隔震层设计的基本原则,进而给出了控制隔震层设计的基本指标,包括隔震支座的长期面压、极值面压、隔震层偏心率等;最后,对高层隔震结构的相关构造要求进行了必要的说明。  相似文献

取活鲤鱼肝胰脏,经冲洗后,立即投入液氮中,然后制成匀浆,先在4℃下600g离心,保留上层液,再900g离心,保留沉淀物并用不同游泳重复悬浮洗涤离心,可得互比较纯的线粒体。将上述粒体在含1%SDS的Saline-Na2EDTA溶液中悬浮,于37℃反应15min,然后在室温用混合溶剂萃到,先后用乙醇洗涤700g,1200g离心,待用电泳检查RNA除净后,加少量蛋白醇-K除去其余蛋白质,最终获得比较的D  相似文献

A new series of 3-phenoxyazetidin-2-ones (β-lactams) were designed and synthesized for the evaluation as selective cyclooxygenase-2 (COX-2) inhibitors. In this study, the effects of a synthetic of β-lactam-structured COX-2 inhibitor with 4-(4-(methylsulfonyl)phenyl)-3-phenoxy-1-phenylazetidin-2-one on cell viability of cancerous lymphoblast isolated from patients diagnosed with acute lymphocytic leukemia (ALL) and normal lymphocytes collected from healthy donors were investigated. The viability % of cancer lymphoblast and normal lymphocyte treated 4-(4-(methylsulfonyl)phenyl)-3-phenoxy-1-phenylazetidin-2-one were tested with MTT assay. Apoptosis and necrosis were measured by double stains of annexin V and propidium iodide, and caspase-3 as a final mediator in apoptotic death measured by colorimetric assay. Mitochondria were isolated from both cancerous lymphoblast and normal lymphocytes to measure parameters of mitochondrial damage such as reactive oxygen species (ROS) formation, mitochondrial membrane potential decrease, swelling, and cytochrome c release following the administration of azithidine-2-one derivative, 4-(4-(methylsulfonyl)phenyl)-3-phenoxy-1-phenylazetidin-2-one. Our results showed that 4-(4-(methylsulfonyl)phenyl)-3-phenoxy-1-phenylazetidin-2-one inhibited proliferation of cancerous lymphoblast in a concentration-dependent manner by inducing apoptosis but not in normal lymphocytes. Treatment with azithidine-2-one derivative produced a rapid loss of mitochondrial transmembrane potential, stimulation of release of ROS and mitochondrial cytochrome c into cytosol, and subsequent induction of procaspase-9 processing. Data suggest that 4-(4-(methylsulfonyl)phenyl)-3-phenoxy-1-phenylazetidin-2-one-induced ROS production led to mitochondria-mediated death signaling that resulted in apoptosis in cancerous lymphoblast cells. The induction of apoptosis by azithidine-2-one compounds, such as 4-(4-(methylsulfonyl)phenyl)-3-phenoxy-1-phenylazetidin-2-one, may provide a mechanism for its cancer chemopreventive action in acute lymphocytic leukemia cells.  相似文献

采用C57BL6小鼠作为模型,SO2(7 mg·m-3)动式吸入染毒28 d,每天染毒6 h;SO2吸入染毒开始后的第1~5 d,每天在SO2吸入染毒结束后对小鼠进行腹腔注射Ba P(40 mg·kg-1(b.w.)),1 d注射1次.吸入染毒结束后,采用荧光定量PCR技术检测小鼠心肌细胞中由核DNA(n DNA)编码的细胞色素C氧化酶亚基CO4和由线粒体DNA(mt DNA)编码的ATP合酶亚基ATP6,以及调控线粒体呼吸链组分的核转录因子PGC1-α、NRF1和mt TFA的mRNA水平;并采用Western blot技术检测上述3种线粒体调控基因的蛋白表达.结果发现,SO2和Ba P复合暴露后,小鼠心肌线粒体氧化磷酸化复合体亚基CO4和ATP6的mRNA表达水平均显著降低,调控基因PGC1-α、NRF1和mt TFA的mRNA和蛋白水平也显著降低.提示小鼠在SO2和Ba P复合暴露后,可能通过降低心肌中NRF1表达,影响其对mt TFA的调控,进一步抑制相关基因组的转录和翻译,最终可能导致小鼠心肌线粒体的氧化磷酸化功能受损,进而引发心血管疾病.  相似文献
