采用富集培养及平板分离方法从成都某污水处理厂活性污泥中筛选出一株具有絮凝活性的菌株SNUX-1,经形态特征及16S rRNA鉴定该菌株为约氏不动杆菌(Acinetobacter johnsonii).对该菌株絮凝活性分布及在不同生长条件下所产絮凝剂的絮凝率进行了研究,并对其絮凝剂成分进行了分析.结果表明,该菌株所产微生物絮凝剂为胞外产物,显色剂显色结果表明,该絮凝剂不含蛋白质成分,含有糖类成分;在以蔗糖为碳源、培养基初始pH值为8、以氯化铜为助凝剂的条件下培养12 h,该菌株所产微生物絮凝剂的絮凝率最高,达96.71%,可作为治理环境污染的菌种资源.  相似文献

为研究不同温度下的土壤微生物呼吸及其与水溶性有机碳(DOC)和转化酶的关系,设置了室内培养实验.采集南京市周边老山、紫金山、宝华山的土壤,研究不同土壤的微生物呼吸对温度升高的响应规律,并分析土壤DOC含量及转化酶活性.结果表明,不同土壤的累积微生物呼吸与土壤温度之间的关系均可用指数方程描述,其P值均达到极显著水平(P0.001),不同地点土壤的微生物呼吸温度敏感系数(Q10值)在1.762~1.895之间变异.累积土壤微生物呼吸的Q10值随着土壤温度升高表现出降低的趋势.培养后27 d土壤微生物呼吸的Q10值与培养后1 d的Q10值无显著差异(P0.05),这表明难分解有机质的温度敏感性与易分解有机质的温度敏感性一致.对于所有土壤而言,累积土壤微生物呼吸与DOC含量之间存在极显著(P=0.003)的线性回归关系,DOC可以解释累积土壤微生物呼吸31.6%的变异性.无论是单独分析不同土壤还是综合所有土壤的测定结果,累积微生物呼吸与土壤转化酶活性均存在极显著(P0.01)的一元线性回归关系,由此说明转化酶活性是衡量土壤微生物呼吸大小的一个较好的指标.  相似文献

微生物分子生态学技术的不断发展,使得同时分析复杂生态系统中微生物的分布和功能特征成为可能.为了研究荧光原位杂交-纳米二次离子质谱技术(fluorescence in situ hybridization-Nano secondary ion mass spectroscopy,FISH-Nano SIMS)在环境微生物生态学上的应用,本研究采用稳定同位素标记的化合物13C-C6H12O6、15N-NH4Cl作为C源和N源,分别对纯培养锰氧化细菌假单胞菌Pseudomonas sp.QJX-1(培养基加锰及不加锰两种条件下),以及浅层土壤及厌氧污泥两种环境样品进行培养.利用FISH-NanoSIMS技术检测培养后样品中微生物体内12C-、13C-、12C14N-、12C15N-的分布特征及其丰度值,进而探讨纯菌及环境样品中微生物利用同位素碳氮源的情况.结果显示所有样品细菌分布区域对应的同位素碳氮(13C、15N)的含量均显著大于其自然丰度值,这表明Pseudomonas sp.QJX-1及环境样品中的微生物均能代谢13C-C6H12O6和15N-NH4Cl.研究进一步发现,Pseudomonas sp.QJX-1在碳氮源消耗至较低浓度时才进行锰氧化;浅层土壤和厌氧污泥中可能都存在同步硝化反硝化细菌群落.FISH和Nano SIMS技术联用能同时分析环境样品中特定微生物的分布特征及代谢功能,进而能更好地掌握环境样品中微生物群落的生理生态学特征.  相似文献

处理水产养殖污水潜流湿地中的厌氧氨氧化菌群特征   总被引:2,自引:2,他引:0  
厌氧氨氧化(anaerobic ammonium oxidation,ANAMMOX)是潜流湿地净化污水的重要功能,目前关于潜流湿地厌氧氨氧化特征还不清楚.为了解处理水产养殖污水潜流湿地中的厌氧氨氧化特点,采用16S rRNA基因克隆文库和实时荧光定量PCR技术以及多样性分析等方法,对处理水产养殖污水潜流湿地中ANAMMOX菌的结构、多样性和丰度进行了研究分析.结果表明,处理水产养殖污水潜流湿地中存在Candidatus brocadia和Candidatus kuenenia这2类已知的ANAMMOX菌和3类未知的ANAMMOX菌,其优势种群为Candidatus brocadia;在不同季节中,秋季潜流湿地中ANAMMOX菌的多样性最高(H',1.21),春季最低(H',0.64);在养殖周期内,潜流湿地中ANAMMOX菌的丰度(以鲜重计,下同)在8.0×104~9.4×106copies·g-1之间,总细菌丰度在7.3×109~9.1×1010copies·g-1之间;在潜流湿地不同层面以及不同季节中,ANAMMOX菌的丰度存在着明显的差异,总细菌丰度则差异不明显,ANAMMOX菌的丰度在不同层面上的差异没有随时间变化的规律性.根据潜流湿地中ANAMMOX菌的分布特征,改变养殖污水布水方式和调整湿地结构可以提高潜流湿地的反硝化效果.该研究为进一步优化潜流湿地结构,提高净化养殖污水效率提供了依据.  相似文献

The impact of diverse composition in teams is neither straightforward nor direct, and evidence suggests that diversity can be either conducive or detrimental to team innovation. Professionally diverse healthcare teams are increasingly used to develop innovative clinical approaches and solve complex healthcare problems; however, there is evidence that collaboration across professional boundaries creates conflict and is frequently unsuccessful. Healthcare organizations consequently face a dilemma. If they embrace professional diversity in teams, they risk interprofessional hostility, but if they choose homogeneous teams, they diminish their teams' capacity to innovate. We respond to this quandary by utilizing social identity theory to better understand the mechanisms through which professional diversity can enhance team innovation. In particular, we argue that professional identity salience operates as a mediator capable of explaining both positive and negative outcomes of professional diversity, contingent on the moderating effect of openmindedness norms. Analysis of survey data from 70 healthcare teams supports our model and indicates that professional salience can both enhance and undermine team innovation, depending on the extent of team openmindedness. Irgarol 1051是一种常用于船舶防污漆的杀生活性物质。为了评价船舶防污漆杀生活性物质Irgarol 1051的海洋环境风险,根据ISO 13073-1的评价原则和程序,对其进行环境危害评价、环境暴露评价和风险表征。通过对公共数据库的文献检索获取数据,从理化性质、环境行为、生态毒性3个方面评价Irgarol 1051的环境危害。采用评估因子法计算Irgarol 1051的预测无效应浓度(PNEC)。采用质量守恒法计算Irgarol 1051在海水中的释放率,通过MAMPEC v3.0模型推导上海洋山深水港的集装箱船区、码头、航道等暴露场景的预测环境浓度(PEC)。经过比较上述暴露场景的风险商值(PEC/PNEC)发现,港口的海水相风险商值大于1,Irgarol 1051的环境风险需要关注。  相似文献

Heavy metal concentrations in muscle and their relation to thegrowth of two marine fish species,including tonguefish( Cynoglossus arel) and mullet( Mugil cephalus),were studied. The samples were collected in Bach Dang estuary andconcentrations of heavy metals( As,Cd,Co,Mn,Cu,Zn,Pb,and V) in muscle of the fisheswere determined. The result showed that the accumulated trend of heavy metal is different between fish species. The concentration of As,Zn,Mn,V,Cu,Pb,Co and Cd in tonguefish were 73. 7 ± 30. 6,22. 82 ± 4.87,3. 44 ± 2. 13,1. 61 ± 0. 15,0. 71 ± 0. 13,0. 45 ± 0. 24,0. 03 ± 0. 02 and 0. 02 ± 0. 02 mg·g-1,respectively. Meanwhile the concentration of Zn,As,V,Mn,Cu,Pb,Co and Cd in mullet were 83. 41 ±19. 68,9. 78 ± 1. 92,1. 36 ± 0. 54,1. 29 ± 0. 51,0. 65 ± 0. 12,0. 42 ± 0. 20,0. 06 ± 0. 03 and0. 03 ± 0. 01mg·g-1,respectively. Comparison of metal levels among thesespecies indicated that the concentrations of As and Mn in tonguefish were significantly higher than those in mullet,whereas Zn levels in mullet were found to be higher than that in tonguefish. There is no significant differences of Cd,Co,Cu,Pb and V levels in muscle between two species. Significant inversed relationshipsbetween concentration levels of metals and lengths of tonguefish were found for Mn,Cu and Zn,butnot for As,Cd,Pb,and V. There were no significant relationships between the heavy metal concentrations and the length of mullet. In general,decreasesof the heavy metal concentrations corresponded to the increases of fish body lengths,particularly for As,Co,Cu,Mn and V and the exception for Cd. Terefore reduced risks were associated with consuming biggermullet fish,and increased risks of As and Cd were associated with consuming bigger tonguefish in Bach Dang river mouth.  相似文献

This research aimed to study the effect of ultra-micro powder Dendrobium officinale on the molecular diversity of intestinal bacteria in mice with spleen-deficiency constipation, in order to provide scientific basis for ultra-micro D. officinale therapy. The mice with spleen-deficiency constipation in treatment groups were respectively given the traditional decoction of D. officinale and 50% dose ultra-micro powder D. officinale. The metagenome DNA of intestinal flora was extracted from intestinal contents. Then ARDRA analysis was made after PCR by universal primer of bacteria. The results showed that OTUs, Shannon index and Brillouin index were the same between the normal group and the 50% dose ultra-micro powder group, and between the model group and the traditional decoction group, but greater in the former two groups than the latter ones. The similarity coefficient (0.500 0) was bigger between the 50% dose ultra-micro powder group and the normal group than between the model group and the traditional decoction group, which was 0.181 8. Both cluster analysis and principal components analysis showed that the molecular diversity of intestinal microbiotia in mice of 50% dose ultra-micro powder group was the closest to that in the normal group. The results showed that the weakened molecular diversity of intestinal bacteria in mice with spleen-deficiency constipation can be regulated by the two kinds of D. officinale treatments to different extent. The efficacy of 50% dose ultra-micro powder D. officinale is better than the traditional decoction.  相似文献

多溴联苯醚(PBDEs)属溴代阻燃剂,曾因其优良的阻燃性能而广泛应用于电子电器、石油化工和建材纺织等工业产品中.然而,随着大量生产和使用,PBDEs已成为大气、水体、土壤和生物体等多环境介质中普遍检出且极具生态风险的有机污染物.因此,开展微生物降解研究对于典型环境中PBDEs污染风险消除和污染修复,具有重要的科学意义.本文从PBDEs环境归趋行为及其暴露风险出发,综述了PBDEs微生物厌氧降解和好氧降解的最新研究动态,比较分析了两种降解过程的降解特性与影响因素,并针对微生物,尤其是好氧微生物降解机理,阐述了bph A或etb A功能基因及其编码酶对PBDEs好氧降解过程的调控作用,同时就PBDEs微生物高效降解菌种选育、降解机理等方面的研究趋势进行了展望.  相似文献

Arbuscular mycorrhizal fungi(AMF) have great potential for assisting heavy metal hyperaccumulators in the remediation of contaminated soils. However, little information is available about the community composition of AMF under natural conditions in soils contaminated by antimony(Sb). The objective of this study was to investigate the characteristics of AMF molecular diversity, and to explore the effects of Sb content and soil properties on the AMF community structure in an Sb mining area. Four Sb mine spoils and one adjacent reference area were selected from around the Xikuangshan mine in southern China. The association of AMF molecular diversity and community composition with the rhizosphere soils of the dominant plant species was studied by Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis(PCR-DGGE). Results from all five studied sites showed that the diversity of AMF decreased with increasing Sb concentration. Principal component analysis(PCA) indicated that the AMF community structure was markedly different among these groups. Further redundancy analysis(RDA) showed that Sb contamination was the dominating factor influencing the AMF community structure in the Sb mine area. However, the multivariate analysis showed that, apart from the soil Sb content, extractable nitrogen content and organic matter content also attributed to AMF sequence distribution type. Some AMF sequences were only found in the highly contaminated area and these might be ideal candidates for improving phytoremediation efficiency in Sb mining regions. Gene sequencing analysis revealed that most species were affiliated with Glomus, suggesting that Glomus was the dominant AMF genus in the studied Sb mining area.  相似文献
