以正十六烷无机盐培养基为选择培养基,从武汉石化输油管附近土壤中筛选出1株高效降解长链烷烃的菌株,命名C3,对其进行生理生化、16S r DNA鉴定,C3为不动杆菌属。在正十六烷浓度为1 000 mg/L的无机盐培养基中接入4%的种子液,放入35℃、125 r/min摇床中震荡60 h,C3对正十六烷的降解率可达100%,其降解动力学拟合结果符合Monod模型。将C3应用到柴蜡的降解,96 h后,1 000 mg/L的柴蜡混合溶液的降解率能达到91%。C3产生的生物表面活性剂经鉴定为磷脂类活性剂,排油圈直径为80 mm,CMC约为35 mg/L,能将水的表面张力降低到20.79 m N/m。该菌株对长链烷烃的降解提供了良好的菌源。  相似文献

为探究准好氧生物矿化垃圾床处理渗滤液过程中的微生物作用机理,研究建立了16S r DNA克隆文库及PCRRFLP技术以研究矿化垃圾反应器的细菌多样性。结果表明:矿化垃圾反应器细菌具有高度多样性,反应器内有36种细菌分别属于13个纲,变形菌纲占绝对优势占70%(其中β-变形占56.5%,γ-变形菌纲占10%),拟杆菌纲、鞘氨醇菌纲,芽孢杆菌纲也具有一定优势;3%的Nitrosomonas属是渗滤液氨氮转化成亚硝酸盐氮的主要功能微生物,由于亚硝酸盐氧化菌不存在或丰度极低,因此造成反应器内亚硝酸盐积累;Thauera属(17%)和Thiobacillus denitrificans属(10%)是反应器内主要优势微生物属,是反应器内反硝化脱氮的功能微生物,由于Thauera属在好氧条件下具有反硝化特性,Thiobacillus denitrificans为严格自养反硝化菌,因此反应器脱氮主要途径为好氧反硝化、自养反硝化。  相似文献

We investigated the absorption rates of zinc, copper and lead in freshwater biofilm and assessed whether biofilm bacterial populations are affected by exposure to environmentally relevant concentrations of these metals in flow chamber microcosms. Metals were rapidly accumulated by the biofilm and then retained for at least 14 days after transfer to uncontaminated water. Changes in bacterial populations were assessed by Automated Ribosomal Intergenic Spacer Analysis (ARISA) and 16S rRNA gene clone libraries. Significant differences in bacterial community structure occurred within only three days of exposure to metals and remained detectable at least 14 days after transfer to uncontaminated water. The rapid uptake of stormwater-associated metals and their retention in the biofilm highlight the potential role of biofilms in the transfer of metals to organisms at higher trophic levels. The sensitivity of stream biofilm bacterial populations to metal exposure supports their use as an indicator of stream ecological health.  相似文献

By enrichment culturing of the sludge collected from the industrial wastewater treatment pond, we isolated a highly efficient nicosulfuron degrading bacterium Serratia marcescens N80. In liquid medium, Serratia marcescens N80 grows using nicosulfuron as the sole nitrogen source, and the optimal temperature, pH values, and inoculation for degradation are 30–35°C, 6.0–7.0, and 3.0% (v/v), respectively. With the initial concentration of 10 mg L?1, the degradation rate is 93.6% in 96 hours; as the initial concentrations are higher than 10 mg L?1, the biodegradation rates decrease as the nicosulfuron concentrations increase; when the concentration is 400 mg L?1, the degradation rate is only 53.1%. Degradation follows the pesticide degradation kinetic equation at concentrations between 5 mg L?1 and 50 mg L?1. Identification of the metabolites by the liquid chromatography/mass spectrometry (LC/MS) indicates that the degradation of nicosulfuron is achieved by breaking the sulfonylurea bridge. The strain N80 also degraded some other sulfonylurea herbicides, including ethametsulfuron, tribenuron-methyl, metsulfuron-methyl, chlorimuron-ethyl,and rimsulfuron.  相似文献

为充分了解20#钢在塔河油田集输管线工况环境中的腐蚀行为,采用高温高压釜研究CO2/H2S分压比、温度和pH值对20#钢在CO2/H2S环境中的腐蚀规律的影响,并利用扫描电镜(SEM)分析腐蚀产物形貌特征。结果表明:CO2分压不变时,随着H2S分压的增加,20#钢的腐蚀速率先增大后减小,H2S分压为0.01 MPa时的腐蚀速率达到最大0.435 mm/a;随着温度的升高,20#钢的腐蚀速率逐渐增大,在100℃时腐蚀速率高达2.280 mm/a;CO2控制下20#钢的腐蚀速率随着pH值的增大而减小;H2S控制下20#钢的腐蚀速率随着pH值的增大而增大。  相似文献

The U.S. Endangered Species Act (ESA) requires that the “best available scientific and commercial data” be used to protect imperiled species from extinction and preserve biodiversity. However, it does not provide specific guidance on how to apply this mandate. Scientific data can be uncertain and controversial, particularly regarding species delineation and hybridization issues. The U.S. Fish and Wildlife Service (FWS) had an evolving hybrid policy to guide protection decisions for individuals of hybrid origin. Currently, this policy is in limbo because it resulted in several controversial conservation decisions in the past. Biologists from FWS must interpret and apply the best available science to their recommendations and likely use considerable discretion in making recommendations for what species to list, how to define those species, and how to recover them. We used semistructured interviews to collect data on FWS biologists’ use of discretion to make recommendations for listed species with hybridization issues. These biologists had a large amount of discretion to determine the best available science and how to interpret it but generally deferred to the scientific consensus on the taxonomic status of an organism. Respondents viewed hybridization primarily as a problem in the context of the ESA, although biologists who had experience with hybridization issues were more likely to describe it in more nuanced terms. Many interviewees expressed a desire to continue the current case‐by‐case approach for handling hybridization issues, but some wanted more guidance on procedures (i.e., a “flexible” hybrid policy). Field‐level information can provide critical insight into which policies are working (or not working) and why. The FWS biologists’ we interviewed had a high level of discretion, which greatly influenced ESA implementation, particularly in the context of hybridization.  相似文献

Two thermophilic digesters were inoculated with manure and started-up under mixed and stagnant conditions. The Archaea in the mixed digester (A) were dominated by hydrogenotrophic Methanobateriaceae (61%) with most of the methane being produced via syntrophic pathways. Methanosarcinales (35%) were the only acetoclastic methanogens present. Acetate dissipation seems to depend on balanced hydrogenotrophic-to-acetotrophic abundance, which in turn was statistically correlated to free ammonia levels. Relative abundance of bacterial community was associated with the loading rate. However, in the absence of mixing (digester B), the relationship between microbial composition and operating parameters was not discernible. This was attributed to the development of microenvironments where environmental conditions are significantly different from average measured parameters. The impact of microenvironments was accentuated by the use of a non-acclimated seed that lacks adequate propionate degraders. Failure to disperse the accumulated propionate, and other organics, created high concentration niches where competitive and inhibiting conditions developed and favored undesired genera, such as Halobacteria (65% in B). As a result, digester B experienced higher acid levels and lower allowable loading rate. Mixing was found necessary to dissipate potential inhibitors, and improve stability and loading capacity, particularly when a non-acclimated seed, often lacking balanced thermophilic microflora, is used.  相似文献

Permeable reactive barriers (PRBs) are an alternative technology to treat mine drainage containing sulfate and heavy metals. Two column experiments were conducted to assess the suitability of an organic carbon (OC) based reactive mixture and an Fe0-bearing organic carbon (FeOC) based reactive mixture, under controlled groundwater flow conditions. The organic carbon mixture contains about 30% (volume) organic carbon (composted leaf mulch) and 70% (volume) sand and gravel. The Fe0-bearing organic carbon mixture contains 10% (volume) zero-valent iron, 20% (volume) organic carbon, 10% (volume) limestone, and 60% (volume) sand and gravel. Simulated groundwater containing 380 ppm sulfate, 5 ppm As, and 0.5 ppm Sb was passed through the columns at flow rates of 64 (the OC column) and 62 (the FeOC column) ml d− 1, which are equivalent to 0.79 (the OC column) and 0.78 (the FeOC column) pore volumes (PVs) per week or 0.046 m d− 1 for both columns. The OC column showed an initial sulfate reduction rate of 0.4 µmol g (OC)− 1 d− 1 and exhausted its capacity to promote sulfate reduction after 30 PVs, or 9 months of flow. The FeOC column sustained a relatively constant sulfate reduction rate of 0.9 µmol g (OC)− 1 d− 1 for at least 65 PVs (17 months). In the FeOC column, the δ34S values increase with the decreasing sulfate concentration. The δ34S fractionation follows a Rayleigh fractionation model with an enrichment factor of 21.6‰. The performance decline of the OC column was caused by the depletion of substrate or electron donor. The cathodic production of H2 by anaerobic corrosion of Fe probably sustained a higher level of SRB activity in the FeOC column. These results suggest that zero-valent iron can be used to provide an electron donor in sulfate reducing PRBs. A sharp increase in the δ13C value of the dissolved inorganic carbon and a decrease in the concentration of HCO3 indicate that hydrogenotrophic methanogenesis is occurring in the first 15 cm of the FeOC column.  相似文献

Increasing recognition of the human dimensions of natural resource management issues, and of social and ecological sustainability and resilience as being inter-related, highlights the importance of applying social science to natural resource management decision-making. Moreover, a number of laws and regulations require natural resource management agencies to consider the “best available science” (BAS) when making decisions, including social science. Yet rarely do these laws and regulations define or identify standards for BAS, and those who have tried to fill the gap have done so from the standpoint of best available natural science. This paper proposes evaluative criteria for best available social science (BASS), explaining why a broader set of criteria than those used for natural science is needed. Although the natural and social sciences share many of the same evaluative criteria for BAS, they also exhibit some differences, especially where qualitative social science is concerned. Thus we argue that the evaluative criteria for BAS should expand to include those associated with diverse social science disciplines, particularly the qualitative social sciences. We provide one example from the USA of how a federal agency − the U.S. Forest Service − has attempted to incorporate BASS in responding to its BAS mandate associated with the national forest planning process, drawing on different types of scientific information and in light of these criteria. Greater attention to including BASS in natural resource management decision-making can contribute to better, more equitable, and more defensible management decisions and policies.  相似文献
