Pollution by toxic metals including cadmium (Cd) and hypoxia are important stressors in estuaries and coastal waters which may interactively affect sessile benthic organisms, such as oysters. We studied metabolic responses to prolonged hypoxic acclimation (2 weeks at 5% O2) in control and Cd-exposed (30 d at 50 μg L−1 Cd) oysters Crassostrea virginica, and analyzed the effects of these stressors on abundance of Vibrio spp. in oysters. Hypoxia-acclimated oysters retained normal standard metabolic rates (SMR) at 5% O2, in contrast to a decline of SMR observed during acute hypoxia. However, oysters spent more time actively ventilating in hypoxia than normoxia resulting in enhanced Cd uptake and 2.7-fold higher tissue Cd burdens in hypoxia. Cd exposure led to a significant decrease in tissue glycogen stores, increase in free glucose levels and elevated activity of glycolytic enzymes (hexokinase and aldolase) indicating a greater dependence on carbohydrate catabolism. A compensatory increase in activities of two key mitochondrial enzymes (citrate synthase and cytochrome c oxidase) was found during prolonged hypoxia in control oysters but suppressed in Cd-exposed ones. Cd exposure also resulted in a significant increase in abundance of Vibrio parahaemolyticus and Vibrio vulnificus levels during normoxia and hypoxia, respectively. Overall, Cd- and hypoxia-induced changes in metabolic profile, Cd accumulation and bacterial flora of oysters indicate that these stressors can synergistically impact energy homeostasis, performance and survival of oysters in polluted estuaries and have significant consequences for transfer of Cd and bacterial pathogens to the higher levels of the food chain.  相似文献

Abstract

Cometabolic degradation of the herbicide molinate was tested using two microorganisms, Arthrobacter sp., strain M3 and Streptomyces griseus strain M2; the latter classified on the basis of the presence of the enzymatic cofactor SF‐420. The strains M3 and M2, inoculated in a basic salts medium with glucose as carbon source and added with 100 mg L‐1 of molinate, degraded respectively 35 and 51% of the herbicide in 36 days.

Increasing concentrations of molinate, ranging from 50 to 200 mg L‐1 in glucose medium, did not affect the final ATP yield of the strain M2, but decreased the final growth yield and the ATP synthesis rate. Moreover, the onset of coenzyme SF‐420 synthesis was progressively delayed.

In contrast, surprisingly, SF‐420 final yield and production rate were increased by progressive increasing concentrations of molinate in the mineral medium.  相似文献

Abstract

A laboratory study was conducted to determine the effect of four experimental insecticides, DOWCO429X, DPX43898, tefluthrin and trimethacarb, on enzyme activities and levels of adenosine 5'‐triphosphate (ATP) in mineral and organic soils. DOWCO429X decreased urease activity in organic soil after 7 days while a stimulatory effect was observed with most treatments after 14 days. No inhibition on acetylene (C2H2) reduction by nitrogenase was evident with any of the insecticides in either soil. With the exception of DOWCO429X and tefluthrin at 7 days in organic soil, none of the insecticide treatments inhibited dehydrogenase activity in either soil. Dehydrogenase activity, measured by formazan formation, was greater in many samples in sandy loam than the control throughout the experiment. No inhibitory effect was observed on amylase activity after 2 or 3 days in sandy soil. A stimulatory effect was apparent in many samples after 2 days in organic soil. All insecticide treatments in sandy soil reduced invertase activity at 2 days. However, none of the experimental insecticides inhibited invertase activity after 3 days. A stimulatory effect in invertase activity was apparent in most cases at 2 days in organic soil and no difference was observed after 3 days. Phosphatase activity in insecticide treated samples was equal to or greater than that of control in sandy soil after 2 h. With the exception of DPX43898, the insecticides depressed phosphatase activity in most organic soil samples. The insecticides did not affect ATP levels in either soil. Results indicated that the chemical treatments at the levels tested did not significantly affect activities of enzymes or level of ATP in both soils.  相似文献

研究了不同浓度镍、锌、铝对常见绿藻羊角月芽藻(S.capricornutum)生长速度、蛋白质含量、ATP水平、葡萄糖-6-磷酸脱氢酶(G6PDH)、酸性磷酸酶及硝酸还原酶活性的影响.结果表明3种金属离子在所试浓度范围内对羊角月芽藻的生长速度均有抑制作用,但单位藻培养物中蛋白质含量却随着金属离子浓度的增加而增加;高浓度金属离子对酶活性有明显抑制作用;藻细胞中ATP水平随着金属离子浓度的增加而下降.提出金属离子对藻类生物产生影响的机理可能是:高浓度金属离子的存在,打破了生物最佳的各种营养元素生物可利用性的平衡.并讨论了藻类生长及生化活性对金属离子反应的相关性.  相似文献

分析松花江流域沉积物中三磷酸腺苷(ATP)微生物量的分布,结果表明,ATP分布具有显著的空间特征,在河流发源地及部分城市地区河流段沉积物中具有高的ATP含量,统计分析(主成分分析,多元回归树分析和响应模型分析)表明:ATP是沉积物的主要成分性质之一,在本研究所涉及11个因子中,影响ATP分布变异的第一因子是空间位置(纬度,39.79%),然后是环境因子(总氮,18.49%;硝氮,16.24%),其中ATP随纬度呈二次曲线变化(P<0.001),与总氮呈显著正相关(P=0.027),和硝氮呈显著负相关(P<0.01).同时ATP也随有机质、总磷和海拔的增加而呈增加趋势(P<0.05).沉积物ATP与多种营养元素及其不同形态具有显著相关性,表明通过ATP活性微生物量可以反映水体环境的营养水平.  相似文献

The energetic parameters, such as glycogen, glucose, proteins, lactate and adenosine triphosphate (ATP) content and lactate deshydrogenase (LDH) activity in tissues and blood of carps from simazine (2-chloro-4,6-bis(ethylamino)-s-triazine) contaminated waters were investigated. In order to confirm the field results, a laboratory experiment was designed in which carps were exposed to simazine at the concentration level of 45 μ g. L? 1 (10-fold of the amount found in natural waters) for 90 days. Fish from a contaminated reservoir showed low glycogen concentrations in hepatopancreas and muscle, while fish in another contaminated reservoir showed high LDH activity together with an increase in lactate content in muscle. Laboratory findings did not confirm field results, and fish exposed to simazine did not show alterations in the parameters studied. The results suggest that carps were not stressed by the presence of the simazine at the concentration levels found in both studies and the mechanisms of defense covered the energetic demand.  相似文献

Fe~(3+)掺杂TiO_2/凹凸棒复合材料的光催化性能   总被引:2,自引:1,他引:1  
采用酸催化的溶胶-凝胶法制备了一系列Fe3+掺杂TiO2/凹凸棒(Fe3+-TiO2/ATP)复合光催化剂材料。在可见光条件下,以亚甲基蓝(MB)溶液的光催化降解反应,评价了样品的光催化性能,研究了TiO2负载量、Fe3+掺杂量和焙烧温度对复合材料光催化性能的影响。在单因素实验的基础上,采用正交实验设计(L25(53))优化了催化剂的制备条件。光催化实验结果表明,MB在复合材料上的光催化降解反应遵循Langmuir-Hinshelwood(L-H)动力学模型。正交实验结果表明,当TiO2负载量为15%、Fe3+掺杂量为0.5%和焙烧温度为550℃时,得到的复合材料对MB的光催化降解效率最佳,测得表观反应速率常数kapp为6.09×10-3min-1,反应4 h后MB的降解率(Dt)可达75.88%,相同实验条件下与P25(1.51×10-3min-1)相比较,反应速率提高了4.03倍,降解率提高了45.05%。另外,复合材料的沉降性能优于P25,易于分离,是一类有应用前景的复合光催化剂。  相似文献

• Long amplicon is more effective to test DNA damage induced by UV. ATP in bacteria does not degrade instantly but does eventually after UV exposure. • After medium pressure UV exposure, ATP degraded faster. The efficacy of ultraviolet (UV) disinfection has been validated in numerous studies by using culture-based methods. However, the discovery of viable but non-culturable bacteria has necessitated the investigation of UV disinfection based on bacterial viability parameters. We used quantitative polymerase chain reaction (qPCR) to investigate DNA damage and evaluated adenosine triphosphate (ATP) to indicate bacterial viability. The results of qPCR effectively showed the DNA damage induced by UV when using longer gene amplicons, in that sufficiently long amplicons of both 16S and gadA indicated that the UV induced DNA damages. The copy concentrations of the long amplicons of 16S and gadA decreased by 2.38 log/mL and 1.88 log/mL, respectively, after exposure to 40 mJ/cm2 low-pressure UV. After UV exposure, the ATP level in the bacteria did not decrease instantly. Instead it decreased gradually at a rate that was positively related to the UV fluence. For low-pressure UV, this rate of decrease was slow, but for medium pressure UV, this rate of decrease was relatively high when the UV fluence reached 40 mJ/cm2. At the same UV fluence, the ATP level in the bacteria decreased at a faster rate after exposure to medium-pressure UV.  相似文献

通过测定有机物生物降解过程中微生物的能量变化--ATP含量的变化来表征有机物的好氧生物降解性。采用受度物的初始浓度为100mg/L溶解性有机碳(DDC),接种生物量为500mg/L,试验周期14d,取得了较好的试验结果。给出了测试程序,采用峰值时间、峰高指数,IA指数作为定量评价指标,从而建立了TP评价方法,并对46种有机物和7种废水的好氧生物降解性进行了评价。  相似文献
