集约化蔬菜种植区地下水中反硝化细菌的分离鉴定   总被引:1,自引:0,他引:1  
从研究我国典型集约化蔬菜种植区地下水中硝酸盐的来源和浓度人手,进行富集、培养、分离、纯化,筛选出一株具有反硝化作用的菌株,通过形态学、革兰氏染色结合16SrDNA序列同源性分析鉴定,其鉴定结果为农杆菌(Agrobacterium sp.).该研究为开展地下水硝酸盐污染的生物修复储备宝贵的菌种资源,为地下水中硝酸盐污染的原位微生物修复和相关污水的生物处理提供微牛物基础,对于经济、有效的解决地下水硝酸盐污染和水资源短缺的问题有着十分重要的意义.  相似文献

Maintaining a living plant collection is the most common method of ex situ conservation for plant species that cannot be seed banked (i.e., exceptional species). Viability of living collections, and their value for future conservation efforts, can be limited without coordinated efforts to track and manage individuals across institutions. Using a pedigree-focused approach, the zoological community has established an inter-institutional infrastructure to support long-term viability of captive animal populations. We assessed the ability of this coordinated metacollection infrastructure to support the conservation of 4 plant species curated in living collections at multiple botanic gardens around the world. Limitations in current practices include the inability to compile, share, and analyze plant collections data at the individual level, as well as difficulty in tracking original provenance of ex situ material. The coordinated metacollection framework used by zoos can be adopted by the botanical community to improve conservation outcomes by minimizing the loss of genetic diversity in collections. We suggest actions to improve ex situ conservation of exceptional plant species, including developing a central database to aggregate data and track unique individuals of priority threatened species among institutions and adapting a pedigree-based population management tool that incorporates life-history aspects unique to plants. If approached collaboratively across regional, national, and global scales, these actions could transform ex situ conservation of threatened plant species.  相似文献

When characterizing environmental radioactivity, whether in the soil or within concrete building structures undergoing remediation or decommissioning, it is highly desirable to know the radionuclide depth distribution. This is typically modeled using continuous analytical expressions, whose forms are believed to best represent the true source distributions. In situ gamma ray spectroscopic measurements are combined with these models to fully describe the source. Currently, the choice of analytical expressions is based upon prior experimental core sampling results at similar locations, any known site history, or radionuclide transport models. This paper presents a method, employing multiple in situ measurements at a single site, for determining the analytical form that best represents the true depth distribution present. The measurements can be made using a variety of geometries, each of which has a different sensitivity variation with source spatial distribution. Using non-linear least squares numerical optimization methods, the results can be fit to a collection of analytical models and the parameters of each model determined. The analytical expression that results in the fit with the lowest residual is selected as the most accurate representation. A cursory examination is made of the effects of measurement errors on the method.  相似文献

Protected areas (PAs) are key elements for biodiversity conservation and ecosystem services. Brazil has the largest PA system in the world, covering approximately 220 million ha. This system expanded rapidly in the mid‐1990s to the mid‐2000s. Recent events in Brazil, however, have led to an increase in PA downgrading, downsizing, and degazettement (PADDD). Does this reflect a shift in the country's PA policy? We analyzed the occurrence, frequency, magnitude, type, spatial distribution, and causes of changes in PA boundaries and categories in Brazil. We identified 93 PADDD events from 1981 to 2012. Such events increased in frequency since 2008 and were ascribed primarily to generation and transmission of electricity in Amazonia. In Brazilian parks and reserves, 7.3 million ha were affected by PADDD events, and of these, 5.2 million ha were affected by downsizing or degazetting. Moreover, projects being considered by the Federal Congress may degazette 2.1 million ha of PA in Amazonia alone. Relaxing the protection status of existing PAs is proving to be politically easy in Brazil, and the recent increase in frequency and extension of PADDD reflects a change in governmental policy. By taking advantage of chronic deficiencies in financial and personnel resources and surveillance, disputes over land tenure, and the slowness of the Brazilian justice, government agencies have been implementing PADDD without consultation of civil society. If parks and reserves are to maintain their integrity, there will need to be investments in Brazilian PAs and a better understanding of the benefits PAs provide. Parasitic species, which depend directly on host species for their survival, represent a major regulatory force in ecosystems and a significant component of Earth's biodiversity. Yet the negative impacts of parasites observed at the host level have motivated a conservation paradigm of eradication, moving us farther from attainment of taxonomically unbiased conservation goals. Despite a growing body of literature highlighting the importance of parasite‐inclusive conservation, most parasite species remain understudied, underfunded, and underappreciated. We argue the protection of parasitic biodiversity requires a paradigm shift in the perception and valuation of their role as consumer species, similar to that of apex predators in the mid‐20th century. Beyond recognizing parasites as vital trophic regulators, existing tools available to conservation practitioners should explicitly account for the unique threats facing dependent species. We built upon concepts from epidemiology and economics (e.g., host‐density threshold and cost‐benefit analysis) to devise novel metrics of margin of error and minimum investment for parasite conservation. We define margin of error as the risk of accidental host extinction from misestimating equilibrium population sizes and predicted oscillations, while minimum investment represents the cost associated with conserving the additional hosts required to maintain viable parasite populations. This framework will aid in the identification of readily conserved parasites that present minimal health risks. To establish parasite conservation, we propose an extension of population viability analysis for host–parasite assemblages to assess extinction risk. In the direst cases, ex situ breeding programs for parasites should be evaluated to maximize success without undermining host protection. Though parasitic species pose a considerable conservation challenge, adaptations to conservation tools will help protect parasite biodiversity in the face of an uncertain environmental future.  相似文献

In the past five years, enclosures have beeninstalled in an acidic mining lake in Lusatia to investigate insitu remediation processes. They were treated with straw, withstraw and Carbokalk, or with Carbokalk alone, where Carbokalkis a by-product of the sugar industry (solid precipitate ofnon-sugars after lime clarification of extracted sugar beetjuice). Sediment samples were taken as cores to get informationabout the behaviour of organic and inorganic components insediment pore-water with depth. Vertical distributions of pH,soluble reactive phosphate, nitrate, ammonium, silica,aluminium, iron, manganese, calcium, magnesium, sodium,potassium, sulphate, and DOC were measured. The resultingprofiles, each consisting of 15 data points at differentdepths, were compared by cluster analysis. The similarities ofsediment cores from different treated enclosures relating toprofiles of chemical components were discussed. Increasedconcentrations of potassium and sulphate were found in pore-water after substrate treatment. The data imply dissolution ofpotassium iron sulphate hydroxide minerals (jarosite) after anincrease of pH caused by dissolution of lime and by sulphatereducing processes which were stimulated by organic substrateaddition.  相似文献

Diffusion experiments in compacted bentonite have been carried out in situ using the borehole laboratory CHEMLAB. The "ordinary" anion iodide and the redox-sensitive pertechnetate ion have been investigated. In spite of strongly reducing groundwater conditions, technetium was found to diffuse mostly unreduced as TcO4-, although in some spots in the compacted clay, the activity was significantly higher, which may be explained by reduction of some TcO4- by iron-containing minerals in the bentonite. The measured concentration profiles in the clay cannot be accommodated by assuming one single diffusion process. The experimental data are modeled assuming two diffusion paths, intralamellar diffusion and diffusion in external water. The apparent diffusivity for the intralamellar diffusion was found to be 8.6 x 10(-11) m2 s(-1) for iodide with a capacity factor of 0.1, while the apparent diffusivity for the diffusion in external water was found to be 5 x 10(-14) m2 s(-1) with alpha=2.26. The corresponding values for Tc were found to be Da= 6 x 10(-11) m2 s(-1), alpha=0.1 and Da= 1 x 10(-13) m2 s(-1), alpha=0.46, respectively. The diffusion constants and capacity factors obtained in this study are in accordance with data from laboratory experiments.  相似文献

The neonatal incidence rate of Down syndrome (DS) is well-known to accelerate strongly with maternal age. This non-linearity renders mere accumulation of defects at recombination during prolonged first meiotic prophase implausible as an explanation for DS rate increase with maternal age, but might be anticipated from chromosomal drive (CD) for trisomy 21. Alternatively, as there is selection against genetically disadvantaged embryos, the screening system that eliminates embryos with trisomy 21 might decay with maternal age. In this paper, we provide the first evidence for relaxed filtering stringency (RFS) to represent an adaptive maternal response that could explain accelerating DS rates with maternal age. Using historical data, we show that the proportion of aberrant live births decrease with increased family size in older mothers, that inter-birth intervals are longer before affected neonates than before normal ones, and that primiparae exhibit elevated levels of DS incidence at higher age. These findings are predicted by adaptive RFS but cannot be explained by the currently available alternative non-adaptive hypotheses, including CD. The identification of the relaxation control mechanism and therapeutic restoration of a stringent screen may have considerable medical implications.  相似文献

This paper is aimed to identify the factors that influence peoples' preference for adaptation against the impacts of sea level rise (SLR). A total of 285 respondents from three coastal villages in Bangladesh are randomly interviewed using a semi-structured questionnaire. First, employing the principal component analysis various factors that influence adaptation preferences of people are identified. These factors are related to “demographic and social aspects”, “wealth and economic standing”, “past coping and adaptive behaviour”, “climate knowledge and information” and “spatial aspect” of life. What is common in these factors is their ability to influence peoples' vulnerability. Finally, the binomial logistic regression model is employed to compute the explanatory power of these factors to predict the respondents' preference for adaptation in situ over retreat or vice versa. Model findings are robust for two scenarios of SLR, i.e. 2050–2075 (LR χ 2?=?133.65, pseudo-R 2?=?0.53, p?<?0.001) and 2080–2100 (LR χ 2?=?282.61, pseudo-R 2?=?0.85, p?<?0.001). Therefore, it is concluded that to avoid relocation of substantial number of people initiative for encouraging adaptation in situ must be taken along side establishment of safe shelter, community radio service and campaign for raising climate awareness.  相似文献

评价了Ag/SAPO-34分子筛催化剂选择性还原NO的活性,并运用漫反射红外光谱原位研究NO在Ag/SAPO-34催化剂上的选择性催化还原机理.结果表明Ag/SAPO-34有良好的低温活性,在氧气浓度为3.6%和温度为573K~673K时NO还原成N2的转化率达70%;催化剂活性随C3H6浓度的增加而升高,随空速的增加而稍有下降.基于漫反射红外光谱,认为反应机理为:NO、丙烯和氧反应,在Ag/SAPO-34催化剂上生成吸附的有机-氮氧化物,再由这些吸附物种分解成N2,催化还原的关键是形成有机-氮氧化物中间体.氧的作用是充分促进丙烯活化以及增加NOx吸附态含量,并且氧的存在是有效产生一系列中间物不可缺少的条件.  相似文献
