Nucleic acids and their analogues are getting more and more attention. Metal-mediated base pairs as a kind of simple and functionalized nucleic acids in special positions have widened the scope of application of functional nucleic acids and their analogues. In this type of base pairs, the representative is the interaction between metal ions and pyrimidine bases, especially the research on thymine-Hg2+-thymine (T-Hg2+-T) and cytosine-Ag+-cytosine (C-Ag+-C) base pairs. This review summarizes the structure and mechanism of metal-mediate pyrimidine base pairs as well as the application in the biochemical analysis. It explores the mode and ratio of coordination between metal ions and base pairs, the effects on the stability of DNA helical structure, the related crystal structure and the three-dimensional configuration information in the DNA helix. The analytical application mainly includes various probes of metal ions, small molecules, protein and the detection of single nucleotide polymorphism. Among them the most widely used metal ions detection, and its combination with spectrum technology, visualization and amplification technology greatly promoted the rapid development of sensitive detection technology. In further studies, it is necessary to reveal the mechanism of interaction between T-Hg2+-T and C-Ag+-C, and more attention should be paid to combining of T-Hg2+-T and C-Ag+-C base pairs with other new technologies. The present research focused on the experimental investigation of phytoreclamation of areas affected by sewage sludge disposal and improving the quality of the sludge. It was conducted in two stages: laboratory and pilot applications. This paper reports on the first stage, which tested, under greenhouse conditions, the possibility of using various species of plants cultivated in the sludge from the Thessaloniki Wastewater Treatment Plant (ThWWTP, Greece) with different amendments. The results of the 30-day experiment showed that the quality of the output sludge material varied significantly in comparison to the input sludge material. These variations were considered as positive, as the output sludge obtained texture and structure, aeration conditions were improved, and the content of some trace metals, excess nutrients (e.g., P), and salts (of Na) were reduced. These were all influenced by both the type of treatment and the plant species used. In conclusion, the best treatments, T1, T3, and T4 (soil/sludge mixture 70%/30%, soil/sludge/gravel mixture 50%/25%/25%, and soil/sludge/gravel mixture 45%/45%/10%, respectively), and the best suited plant species, Helianthus annuus L. and Zea mays L., were proposed to be used in the pilot application (in situ) at the site within the ThWWTP. 相似文献

To better understand the diversity of metal resistance genetic determinant from microbes that survived atmetal tailings in northwest of China, a highly elevated level of heavymetal containing region, genomic analyses was conducted using genome sequence of three native metal-resistant plant growth promoting bacteria (PGPB). It shows that: Mesorhizobium amorphae CCNWGS0123 contains metal transporters from P-type ATPase, CDF (Cation Diffusion Facilitator), HupE/UreJ and CHR (chromate ion transporter) family involved in copper, zinc, nickel as well as chromate resistance and homeostasis. Meanwhile, the putative CopA/CueO system is expected to mediate copper resistance in Sinorhizobium meliloti CCNWSX0020 while ZntA transporter, assisted with putative CzcD, determines zinc tolerance in Agrobacterium tumefaciens CCNWGS0286. The greenhouse experiment provides the consistent evidence of the plant growth promoting effects of these microbes on their hosts by nitrogen fixation and/or indoleacetic acid (IAA) secretion, indicating a potential in-site phytoremediation usage in themining tailing regions of China. 相似文献

Experimental and geochemical modelling studies were carried out to identify mineral and solid phases containing major, minor, and trace elements and the mechanism of the retention of these elements in Flue Gas Desulphurisation (FGD)-gypsum samples from a coal-fired power plant under filtered water recirculation to the scrubber and forced oxidation conditions. The role of the pH and related environmental factors on the mobility of Li, Ni, Zn, As, Se, Mo, and U from FGD-gypsums for a comprehensive assessment of element leaching behaviour were also carried out. Results show that the extraction rate of the studied elements generally increases with decreasing the pH value of the FGD-gypsum leachates. The increase of the mobility of elements such as U, Se, and As in the FGD-gypsum entails the modification of their aqueous speciation in the leachates; UO2SO4, H2Se, and HAsO2 are the aqueous complexes with the highest activities under acidic conditions. The speciation of Zn, Li, and Ni is not affected in spite of pH changes; these elements occur as free cations and associated to SO42 in the FGD-gypsum leachates. The mobility of Cu and Mo decreases by decreasing the pH of the FGD-gypsum leachates, which might be associated to the precipitation of CuSe2 and MoSe2, respectively. In this paper, laser additive manufacturing (LAM) of Fe–TiC composite coating on AISI 1030 carbon steel is investigated using a numerical and experimental method. In order to have a desired result using LAM, it is crucial to understand the effects of the process parameters’ values on the TiC morphology and microstructure. For this purpose, the LAM process is numerically simulated in order to calculate cooling rate and peak temperature. Experimental data and numerical results are in good agreement in terms of the phase development. Results show that cooling rate plays a crucial role in phase transformation in the clad, however, final microstructure strongly depends on the cooling rate and powder's chemical composition. Two main carbide morphologies (i.e. dendritic and particulate) are studied and relevant cooling rates are detected. Based on this paper and developed map, it is possible to control the cooling rate in order to achieve specific carbide morphologies in the clad. In this study, scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS) are used to characterize clads’ microstructure. 相似文献
