Biofilms mediate crucial biochemical processes in aquatic ecosystems. It was hypothesized that eutrophication may promote the growth of biofilms, resulting in larger numbers of functional genes. However, the metabolic activity and the roles of biofilms in N cycling will be affected by ambient inorganic nitrogen availability, not by the abundance of functional genes. Biofilms were cultured either with replete inorganic nitrogen(N-rep) or without exogenous inorganic nitrogen supply(N-def) in a flow incubator, and the N-cycling gene abundances(nifH, N_2 fixation; amoA, ammonia oxidation, archaea and bacteria; nirS and nirK, denitrification) and enzyme activities(nitrogenase and nitrate reductase) were analyzed. The results showed that, comparing the N-def and N-rep biofilms, the former contained lower nifH gene abundance, but higher nitrogenase activity(NA), while the latter contained higher nifH gene abundance, but lower NA. Different patterns of NA diel variations corresponded to the dynamic microbial community composition and different stages of biofilm colonization. Ammonia oxidizing bacteria(AOB), detected only in N-def biofilms, were responsible for nitrification in biofilms. N-rep biofilms contained high nirS and nirK gene abundance and high denitrification enzyme activity, but N-def biofilms contained significantly lower denitrification gene abundance and activity. In general,the strong N_2 fixation in N-def biofilms and strong denitrification in N-rep biofilms assured the balance of aquatic ecosystems. The results suggested that evaluation of the functional processes of N cycling should not only focus on genetic potential, but also on the physiological activity of biofilms. 相似文献

ABSTRACT: The Chowan River system consists of three rivers in southeast Virginia that form two confluences before flowing into Albermarle Sound in North Carolina. A computer program was written to simulate flows through the river system to determine flow rates, velocities, and depths. The output of the flow program was input into a second program that calculated the concentrations of BOD5, COD, DO, and four nitrogen parameters (organic, ammonia, nitrite-nitrate and algal-N). Measured field data were used to calibrate the model. The effect of reducing the concentration of nutrients from overland runoff on algal concentrations at the mouth of the river was studied. The program was also run to simulate the water quality of the watershed in a primitive condition, in which the watershed was assumed to consist only of forests. The results of the computer program indicate that the major changes in the water quality of the river are simulated satisfactorily. The program can be used to assess the impact of any management scheme to improve water quality. 相似文献

The accumulation of organic matter in substratum pores is regarded as an important factor causing clogging in the subsurface flow constructed wetlands.In this study,the developing process of clogging separately caused by biofilm growth and organic particles accumulation instead of total organic matter accumulation was investigated in two groups of lab-scale vertical flow constructed wetlands (VFCWs) fed with glucose (dissolved organic matter) and starch (particulate organic matter) influent.Results showed that the growth of biofilms within the substratum pores certainly caused remarkable reduction of effective porosity,especially for the strong organic wastewater,whereas its influence on infiltration rate was negligible.It was implied that the most important contribution of biofilm growth to clogging is accelerating the occurrence of clogging.In comparison with biofilm growth,particles accumulation within pores could rapidly reduce infiltration rate besides effective porosity and the clogging occurred in the upper 0-15 cm layer.With approximately equal amount of accumulated organic matter,the effective porosity of the clogged layer in starch-fed systems was far less than that of glucose-fed systems,which indicated that composition and accumulation mode of the accumulated organic matter played an important role in causing clogging besides the amount.According to the results,some related methods to prevent and recover the clogging phenomenon were suggested. 相似文献

三峡水库支流大宁河水华特征研究 总被引:19,自引:8,他引:11
以三峡水库支流大宁河水华敏感期(2007年4~6月和2008年4~5月)水质和水华的调查数据为依据,研究了其水质变化、营养盐构成及水华特征.结果表明,水华敏感期大宁河自回水段以下氮磷含量较高,总氮范围为0.84~3.21 mg/L,总磷为0.011~0.531 mg/L,氮营养盐主要以TDN为主(TDN/TN为84%),而磷营养盐以TDP占优(TDP/TP为60%);N/P值均高于16,藻类生长受磷限制.高锰酸盐指数和溶解氧含量低且变化稳定.叶绿素a为1.41~219.04 mg.m-3.相关性分析表明Chl-a与TP、高锰酸盐指数、DO、pH呈显著正相关(rChla-TP=0.453、rChla-高锰酸盐指数=0.641、rChla-DO=0.584、rChla-pH=0.409,p0.01),与SD呈显著负相关(rChla-SD=-0.392,p0.01);pH受多参数影响,但受藻类生长影响较大.通过显微镜对藻类进行鉴定,发现大宁河自回水段以下水华敏感期藻类分布较广,共发现8门82属124种,其中硅藻门和绿藻门分布最多,其次为蓝藻门和甲藻门,其余藻种较少.大宁河在水华敏感期暴发了3次自回水段以下大规模水华,水华期间藻密度最高值是正常值的14~1427倍,水华优势种主要有绿藻门的波吉卵囊藻、小空星球藻、土生绿球藻、实球藻和小球藻;裸藻门的扁裸藻、矩圆囊裸藻;硅藻门的小环藻、冠盘藻和舟形藻;甲藻门的埃尔多甲藻等;发生多藻种同时水华的情况;出河口处藻类较少. 相似文献

As representatives of primary producers, algae and cyanobacteria play an important role in assessing the risk of chemicals. However, the list of standard algal species commonly used for toxicity testing comprises very few species so that there is an urgent demand to identify further non-standard species which can be used for higher-tier risk assessment in general and the analysis of species sensitivity distributions in particular. In this study, four measuring techniques to assess growth inhibition were compared using five non-standard and two standard algal species and 3,5-dichlorophenol as reference substance. Regarding sensitivity and suitability, the measuring techniques were ranked in the following descending order: delayed fluorescence, prompt fluorescence, photometric absorbance, and microscopic cell count. 嘉陵江为三峡库区最大支流,对库区水环境具有重要影响,2004年夏进行了嘉陵江出口段藻类活动规律研究,2005年春首次发现嘉陵江出口段小环藻(Cyclotella)暴发。2008年春嘉陵江出口段小环藻暴发水样经中科院武汉水生所鉴定的水华藻种为极小冠盘藻(Stephanodiscus minutulus),由于极小冠盘藻光学显微特征与小环藻暴发水样中主要小环藻光学显微特征不一致,因此需要弄清春季嘉陵江出口段水华小环藻分类地位。为认知春季嘉陵江出口段水华小环藻种,2009年春季于嘉陵江出口段小环藻发生期间,采集水样,离心浓缩,酸化处理,经电子显微镜扫描得到主要小环藻的电子显微照片,通过中国淡水藻志第四卷硅藻门中心纲相关小环藻特征分析比较,嘉陵江出口段春季水华小环藻种为星肋小环藻(Cyclotella asterocostata Grun.)。 相似文献

近年来太湖流域局部水质状况有所改善,但太湖藻型生境条件还未根本改变,水污染防治任务依然艰巨。确保太湖湖体水质稳定达标,尤其是加强对太湖重点湖区和水源地重点污染物的调查研究十分重要。在此背景下,本文调研了太湖重点湖区和水源地水质概况、藻毒素污染时空分布特征、环境影响因子和迁移转化规律,并总结了藻毒素的环境和健康风险研究的最新进展,指出了太湖西部湖区和饮用水源地的主要环境风险,以及未来太湖藻毒素污染相关研究需解决的关键技术问题,以期为促进太湖流域重点污染物的控制和治理,确保太湖饮用水源地安全提供有益借鉴。 相似文献

人工湿地中抗生素抗性大肠杆菌和抗性基因的去除与分布 总被引:1,自引:0,他引:1
抗生素的滥用导致抗生素抗性菌和抗性基因随生活污水和养殖废水的排放在环境中肆意散播,其去除及环境行为越来越受到关注。采用K-B纸片法测定了9套不同工艺构型模拟人工湿地中大肠杆菌对7种抗生素的抗性率,并应用多重PCR检测磺胺类sul1、2、3与四环素tetA、B、C、D抗性基因,探究人工湿地对抗性菌的去除效率及抗性菌、抗性基因的分布规律。结果显示,人工湿地能有效去除污水中70%左右的抗性大肠杆菌,有效降低了细菌抗性的传播风险;共计分离出535株大肠肝菌中有378株对一种以上抗生素有抗性性,以四环素、磺胺类和氨苄西林抗性率最高,达到25%以上,其他4种抗性率较低,不足20%;2种抗性基因的检出率都在70%以上;对不同采样点大肠杆菌的抗性性及抗性基因的比较发现,各部分大肠杆菌的抗性水平、多重抗性指数(MRI)以及抗性基因(sul、tet)检出率和组合数表现出:基质≥出水>进水,推测抗性菌被湿地基质截留,在基质生物膜上发生抗性基因的重组,并释放抗性菌,提高了出水中抗性水平和抗性基因检出率。 相似文献
