Extraction and quantification of pesticide residue from the milk matrix at or below the established maximum residue limit (MRL) is a challenging task for both analytical chemists and the regulatory institutions to take corrective actions for the human health and safety. The main aim of the study is to develop a simple rapid and less expensive QuEChERS extraction and cleanup method for simultaneous analysis of 41 multiclass pesticide residue in milk by gas chromatography-electron capture detector (GC-ECD), followed by confirmation of the residues with gas chromatography-mass spectrometer (GC-MS). Effect of sorbent type, temperature, spiking concentration, matrix effect (ME), measurement uncertainty (MU), inter- and intra-assay repeatability, reproducibility of recovery, and trueness of the results were investigated to validate the effectiveness of the method. Limit of determination (LOD) and limit of quantitation (LOQ) for all the analytes ranged within 0.001–0.02 and 0.002–0.05 µg mL?1, respectively. The % recovery of all the pesticides ranged between 91.38 and 117.56% with relative standard deviation (RSD) below 2.79%. The MU for all the analytes was ≤29% of respective LOQs, and except for few pesticides, the ME was largely negative. Persistent organic pollutants (POPs) are ubiquitous residual contaminants in the environment and in the food chain. Milk is an important matrix for monitoring POP contamination. This study determined the concentrations of POPs including polybrominated diphenyl ethers (PBDEs), hexachlorobenzene (HCB), polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs), and dioxin-like polychlorinated biphenyls (DL-PCBs) in raw bovine milk, and the data was used to estimate dietary intakes in South Korea. The average concentrations of PBDEs, HCB, PCDD/Fs, and DL-PCBs in raw milk were 0.29 ng g−1 fat, 0.50 ng g−1 fat, 0.27 pg TEQ g−1 fat, and 0.33 pg TEQ g−1 fat, respectively. No significant relationship was found between the concentrations of analytes and the regions sampled. The dietary intakes of PBDEs from the consumption of milk was calculated to be 0.26, 0.10, and 0.05 ng kg−1 bw d−1 for the group of ages 0–6, 7–12, and 13–19, respectively. The dietary intakes of HCB was calculated to be 0.44, 0.16, and 0.09 ng kg−1 bw d−1 for the group of ages 0–6, 7–12, and 13–19, respectively. The dietary intakes of PCDD/Fs and DL-PCBs was calculated to be 0.78, 0.29, and 0.16 pg TEQ kg−1 bw d−1 for the group of ages 0–6, 7–12, and 13–19, respectively. These results indicated that the residual levels of PBDEs, HCB, PCDD/Fs, and DL-PCBs in raw bovine milk were within safe levels. 相似文献

Very limited information is available on body burdens and environmental levels of dioxins and dioxin-like PCBs (dl-PCBs) in mainland China. In the current studies, human milk samples were collected from 30 breastfeeding mothers in Shijiazhuang city (industrialized) and 11 in the Tanshan countryside (agricultural) of Hebei Province in northern China. An additional 20 samples were obtained from mothers in Tokyo, Japan. PCDDs, PCDFs, and dl-PCBs in human milk were analyzed by high-resolution gas chromatography/high-resolution mass spectrometry. Our results show that arithmetic means for body burdens of PCDDs/Fs and dl-PCBs in Hebei were 3.6 and 1.9 pg TEQ g−1 fat, respectively, which were only about one fourth of the levels in Japan. In addition, no difference was found in the chemical levels except dl-PCBs between the urban and rural areas. Based on the results of an in-person interview of the Chinese mothers using a 59-item questionnaire, freshwater fish consumption was found to correlate with the body burden of dioxins. Principal component analysis of dioxin congeners revealed that the patterns of dioxins in the Hebei urban and rural areas are quite similar; however, they are clearly different from those in Japan. Collectively, our results suggest that the lower body burdens of dioxin in Hebei may be due in part to the relatively slow industrialization and a lower consumption of marine foods. Finally, the results indicate that comprehensive monitoring of dioxins and dl-PCBs in humans as well as in the environment and foods is necessary in China. 相似文献

This study was undertaken to develop and validate direct competitive ELISA for the determination of chloramphenicol residues in bovine milk. Antisera and an enzyme-tracer for chloramphenicol were prepared and used to develop an ELISA with inhibition concentrations, IC20 and IC50, of 0.09 and 0.44 ng mL?1, respectively. Milk samples were spiked with standards equivalent to 0, 0.2, 0.3, 0.5, 1.0 &; 1.5 ng mL?1 and extracted in methanol. The mean recoveries were found to be 73–100% with coefficient of variance 7–11%. The decision limit (CCα) and detection capability (CCβ) were calculated as 0.10 and 0.12 ng mL?1, respectively. The results were found comparable with the commercial ELISA, having recoveries of 87 to 100%, CCα 0.09 ng mL?1 and CCβ 0.12 ng mL?1. As per Commission Decision 2002/657/EC, in-house ELISA was further validated by using LC-MS/MS. Mass spectral acquisition was done by using electrospray ionization in the negative ion mode applying single reaction monitoring of the diagnostic transition reaction for CAP (m/z 152, 194 and 257). The calibration curve showed good linearity in concentrations from 0.025 to 1.6 ng mL?1 with correction coefficient 0.9902. The mean recoveries were found to be 88 to 100%. The CCα was calculated as 0.057 ng mL?1 and CCβ 0.10 ng mL?1. Since CCα and CCβ are less than half of the MRPL (0.15 ng mL?1), the test was found suitable for screening and quantification of CAP residues in bovine milk samples. Results of surveillance studies indicated that out of 31 analyzed milk samples, 12.9% samples were found with CAP residues but only 3.2% samples were declared positive with maximum concentration 0.31 ng mL?1, slightly above the MRPL. 相似文献

The objective of this study was to determine the concentrations of polycyclic aromatic hydrocarbons (PAHs) in cow's milk from industrial farms that are located near an industrial park in Hidalgo, Mexico. It was found that the concentrations of PAHs in the raw milk of cattle from industrial farms have increased in recent years. Composite samples were collected between 2008 and 2010 and analysis carried out according to 8100 EPA procedures and analyzed by gas chromatography with FID detection. The results show that combustion PAHs were mostly Ace, Acy, and Fla (0.25, 0.32, and 0.22 µg g?1, respectively). Diagnostic ratios were used to show that the probable sources were grass and fuel combustion. The sum of concentrations of 16 individual PAHs did not breach permissible levels in milk (25 µg g?1 according to the United States EPA), indicating a limited health risk to animals and humans in the study area. Fertilizer management and straw returning are effective measures to regulate greenhouse gas emissions and increase crop yields, which have attracted wide attention in agricultural production. To clarify the effect of Chinese milk vetch returning with nitrogen fertilizer on rice yield and greenhouse gas emissions in paddy field, field experiments were conducted during 2017–2018 and four treatments were proposed in this study, including the treatments with Chinese milk vetch returning plus different nitrogen fertilizer application amount, namely RA, RB, RC, and control CK (winter fallow without Chinese milk vetch returning). The results showed that treatments RA, RB, and RC significantly increased the early rice yield by 15.35%, 12.94%, and 15.35%, respectively (P<0.05), and treatment RA had the best effect on the annual yield (P<0.05) compared with control CK. Meanwhile, all the treatments with Chinese milk vetch returning plus nitrogen fertilizer increased the global warming potential, but the difference between RA, RB, and control CK was not significant (P>0.05); the greenhouse gas intensity produced by RA was 11.76% lower than control CK. Polybrominated diphenyl ethers(PBDEs) can be transferred to infants through the ingestion of breast milk, resulting in potential health risk. In this study, PBDEs, hydroxylated polybrominated diphenyl ethers(OH-PBDEs) and 2,2′,4,4′,5,6′-hexachlorobiphenyl(CB-153)in human milk from women living adjacent to e-waste recycling sites of Wenling,China, were investigated. The median level of PBDEs in samples from residents living in the e-waste recycling environment 20 years(R_20group, 19.5 ng/g lipid weight(lw))was significantly higher than that for residents living in Wenling 3 years(R_3group,3.88 ng/g lw)(p 0.05), likely ascribable to specific exposure to PBDEs from e-waste recycling activities. In the R_20 group, most congeners(except for BDE-209) were correlated with each other(p 0.05). Moreover, CB-153 showed significant association with most PBDE congeners, rather than BDE-209. The relationship indicated that most BDE congeners other than BDE-209 shared common sources and/or pathways with CB-153, e.g., dietary ingestion. The correlations between BDE-209 and other congeners were different in the two groups, likely suggesting their different exposure sources and/or pathways for PBDEs.Although estimated dietary intake of PBDEs for infants via breast milk was lower than the minimum value affecting human health, the PBDE exposure of infants should be of great concern because of their potential effect on the development of neonates over long-term exposure. OH-PBDEs were not detected in the collected samples, which is in accordance with reports in published literature, likely indicating that they were not apt to be accumulated in human milk. 相似文献

Synthetic musks are used as additives in many household products. After absorption into the human body, they accumulate andtheir concentrations in human milk reflect both the mother and her infant’s exposure level. Concentrations of four synthetic musks,musk xylene (1-tert-butyl-2,6-dimethyl-2,4,6-trinitrobenzene, MX), musk ketone (4-tert-butyl-2,6-dimethyl-3,5-dinitroacetophenone,MK), 1,3,4,6,7,8-hexahydro-4,6,6,7,8,8-hexamethylcyclopenta[]-2-benzopyran (HHCB) and 7-acetyl-1,1,3,4,4,6-hexamethyl-1,2,3,4-tetrahydronaphthalene (AHTN), were determined in human milk samples collected from Shanghai, Wuxi, and Shaoxing in EasternChina. The four synthetic musks were found in most samples analyzed, with HHCB the dominant component followed by MX. Themedian (mean) values for HHCB, AHTN, MX and MK concentrations were 63 (82), 5 (12), 17 (24) and 4 (9) ng/g lipid weight,respectively. These data suggested the total synthetic musk contamination was low, and the distribution percentage was HHCB > MX> AHTN MK. The relative high ratio of nitro to polycylic musk indicated that nitro musks were still widely used. The muskconcentrations in these cities were not significantly different from each other (p > 0.05). Principal components score plots wereobtained, which showed similar exposure sources. The amount of total synthetic musks in human milk were not associated withmother’s age, although HHCB was significantly correlated with AHTN (p < 0.05). Daily ingestion of HHCB, AHTN, MX and MK forinfants from human milk were estimated as (2526 2926), (370 524), (7391 832), and (277 462) ng/day, respectively.
