苯酚降解菌UW7的鉴定及对苯酚的降解作用   总被引:5,自引:0,他引:5  
从焦化厂污水处理曝气池泥样中分离出具有降解苯酚能力的UW7菌株,根据形态、生理生化性状初步鉴定为不动杆菌属(Acinetobacter).该菌16S rRNA基因序列(在GenBank中的登录号为GU083586)与多株鲁氏不动杆菌(A.lowffii)的相似性在99%以上.结合形态、生理生化特性,鉴定UW7菌株为鲁氏不动杆菌(A.lowffii UW7),该种细菌具有降解苯酚的特性尚未见报道.该菌株降解苯酚的最适温度为30℃,最适生长pH 7.0,对2.5 g/L浓度的苯酚能够有效降解,对3.5~4.0 g/L浓度的苯酚有较强的耐受能力,是处理高浓度苯酚废水的良好菌种资源.  相似文献

从云南哀牢山泥土中分离出一株生长温度范围为7~40℃的兼性嗜冷产甲烷杆菌8-1.该菌株革兰氏染色阳性,直径0.2~0.3μm,长2~5μm,具有弯曲和微弯杆状两种形态,不运动,单生﹑成对﹑多数成聚集状态存在.能够利用H2/CO2和甲酸盐生长,不利用甲醇﹑三甲胺﹑乙酸和二元醇类.最适生长条件:温度为35℃,pH值为7.5~8.5,盐浓度为0 mol/L NaCI.最低生长的温度可达到7℃.菌株8-1与标准株Methanobacterium subterraneum strain A8p(=DSM 11074T)的16S rRNA基因序列相似性为99%.  相似文献

农村污水膜生物反应器系统中微生物群落解析   总被引:5,自引:2,他引:3  
农村污水的随意排放和未加限制的灌溉回用可能会导致水源地环境污染加剧,并且其中潜在的病原微生物对周边淡水资源安全和农村居民的身心健康造成潜在的威胁.为了解农村污水的微生物群落组成,为后续污水灌溉的病原微生物风险评价提供理论依据,采用末端限制性片段长度多态性(T-RFLP)和16S r DNA基因克隆文库技术研究农村污水膜生物反应器(MBR)系统中的微生物群落多样性,并结合实时荧光定量PCR方法监测处理前后典型病原菌——弓形菌(Arcobacter spp.)以及总细菌数量的变化.从未处理的农村污水中获取的73个阳性克隆测序结果表明,其主要细菌类群为变形菌门(91.8%)、厚壁菌门(2.70%)、拟杆菌门(1.40%),以及部分不可培养细菌(4.10%).其中弓形菌属是ε-变形菌门的优势菌,也是农村污水中的优势菌株,占克隆总数的68.5%.TRFLP的结果表明,不同处理工艺阶段的主要细菌类群及丰度明显不同,调节池的物种丰度(S)、Shannon-Wiener(H)和物种均匀度(E)最高,分别为43.0、3.56和0.95.定量PCR结果显示未处理的农村污水中弓形菌数量高达(1.09±0.064 0)×1011copies·L-1,该结果与克隆文库结果均表明弓形菌在污水中确实占较高比例.与未处理的农村污水相比,处理后的出水中所监测的弓形菌及细菌总拷贝数分别减少了2~3个数量级,说明MBR处理系统在一定程度上能够去除微生物.处理后的再生水理化指标及指示菌卫生指标均符合农田灌溉用水标准,但其中残留病原微生物引发的健康风险仍需进一步评价.  相似文献

拟通过筛选出高效还原Cr(Ⅵ)的菌株来修复铬污染土壤,从重庆废弃化工厂铬污染土壤中分离得到一株高效还原菌株,对该菌进行形态和生理生化特征研究,并结合分子生物学进行测序分析,同时对该菌在不同条件下还原Cr(Ⅵ)的能力进行检测,探讨了pH值、接种量、初始Cr(Ⅵ)质量浓度及温度对Cr(Ⅵ)还原的影响.结果表明,该菌株为革兰氏阳性杆菌,命名为NO.1.经形态和生理生化特征及16S rDNA序列比对分析,鉴定为蜡样芽孢杆菌(Bacillus cereus)(相似度为99%).该菌株具有较强的还原Cr(Ⅵ)的能力,在pH=8.0、接种量50%(体积分数)及30℃条件下,100 mg/L的Cr(Ⅵ)在处理3d后可基本被还原.研究表明,该细菌有望用于铬污染土壤的修复.  相似文献

以污泥干化芦苇床中芦苇内生菌的多样性作为主要研究对象,结合16S rDNA高通量测序技术与PCR-DGGE技术,对污泥干化芦苇床和天然湿地中污泥菌群及芦苇内生菌群多样性进行了比较分析。结果表明,污泥干化芦苇床芦苇内生菌的细菌类群包含了20门、30纲、73目、152科、467属、1 167种细菌。其中变形菌门(65.6%)和拟杆菌门(25.5%)占绝对优势,γ-变形菌纲是丰度最高的纲,占总数的43.4%;排前20位的优势菌种相对百分比之和为46.5%,分属于所对应的18个属、12科、10目、6纲、3门。DGGE分析表明,污泥干化芦苇床中的污泥和芦苇内生菌优势菌群的种类与天然湿地相比差异显著;同时由于污泥的影响,芦苇内生菌群种类也发生了较为明显的变化。  相似文献

从绍兴水处理发展有限公司的曝气池污泥中筛选到6株聚乙烯醇( PVA)降解功能菌株,编号为rF1、rF2、rF3、rF4、rF5和rF6。通过16S rDNA测序比对表明其分别为Pseudochrobactrum sp、解淀粉芽孢杆菌(Bacil-lus amyloliquefaciens)、芽孢杆菌(Bacillus sp)、门多萨假单胞菌(Pseudomonas mendocina)、杆菌属(Lysinibacillus sp)、不动杆菌( Acinetobacter sp )。在自配培养基中,有3株菌PVA降解率可达70%~80%,在印染废水中,有5株菌PVA降解率可达到90%以上。其中rF1、rF2和rF5无论是在自配培养基还是印染废水中都具有较好效果,表明具有一定的实际应用价值。  相似文献

Most of the oxalotrophic bacteria are facultative methylotrophs and play important ecological roles in soil fertility and cycling of elements. This study gives a detailed picture of the taxonomy and diversity of these bacteria and provides new information about the taxonomical variability within the genus Methylobacterium. Twelve mesophilic, pink-pigmented, and facultatively methylotrophic oxalate-oxidizing strains were included in this work that had been previously isolated from the soil and some plant tissues by the potassium oxalate enrichment method. The isolates were characterized using biochemical tests, cellular lipid profiles, spectral characteristics of carotenoid pigments, G+C content of the DNA, and 16S rDNA sequencing. The taxonomic similarities among the strains were analyzed using the simple matching (S SM) and Jaccard (S J) coefficients, and the UPGMA clustering algorithm. The phylogenetic position of the strains was inferred by the neighbor-joining method on the basis of the 16S rDNA sequences. All isolates were Gram-negative, facultatively methylotrophic, oxidase and catalase positive, and required no growth factors. Based on the results of numerical taxonomy, the strains formed four closely related clusters sharing ≥85% similarity. Analysis of the 16S rDNA sequences demonstrated that oxalotrophic, pink-pigmented, and facultatively methylotrophic strains could be identified as members of the genus Methylobacterium. Except for M. variabile and M. aquaticum, all of the Methylobacterium type strains tested had the ability of oxalate utilization. Our results indicate that the capability of oxalate utilization seems to be an uncommon trait and could be used as a valuable taxonomic criterion for differentiation of Methylobacterium species.  相似文献

Purification of the total DNA extracted from activated sludge samples was studied. The effects of extraction buffers and lysis treatments (lysozyme, sodium dodecyl sulfate (SDS), sonication, mechanical mill and thermal shock) on yield and purity of the total DNA extracted from activated sludge were investigated. It was found that SDS and mechanical mill were the most effective ways for cell lysis, and both gave the highest DNA yields, while by SDS and thermal shock, the purest DNA extract could be obtained. The combination of SDS with other lysis treatment, such as sonication and thermal shock, could apparently increase the DNA yields but also result in severe shearing. For the purification of the crude DNA extract, polyvinyl polypyrrolidone was used for the removal of humic contaminants. Cetyltrimethyl ammonium bromide, potassium acetate and phenol/chloroform were used to remove proteins and polysaccharides from crude DNA. Crude DNA was further purified by isopropanol precipitation. Thus, a suitable protocol was proposed for DNA extraction, yielding about 49.9 mg (total DNA)/g volatile suspended solids, and the DNA extracts were successfully used in PCR amplifications for 16S rDNA and 16S rDNA V3 region. The PCR products of 16S rDNA V3 region allowed the DGGE analysis (denatured gradient gel electrophoresis) to be possible.  相似文献

Based on the successful performance of a lab-scale upflow anaerobic sludge blanket (UASB) reactor with the capacity of simultaneous methanogenesis and denitrification (SMD), the specific phylogenetic groups and community structure of microbes in the SMD granule in the UASB reactor were investigated by the construction of the Eubacteria and Archaea 16S rDNA clone libraries, fragment length polymorphism, and sequence blast. Real time quantitative-polymerase chain reaction (RTQ-PCR) technique was used to quantify the contents of Eubacteria and Archaea in the SMD granule. The contents of some special predominant methanogens were also investigated. The results indicated that the Methanosaeta and Methanobacteria were the predominant methanogens in all Archaea in the SMD granule, with contents of 71. 59% and 22. 73% in all 88 random Archaea clones, respectively. The diversity of Eubacteria was much more complex than that of Archaea. The low GC positive gram bacteria and Б-Protebacteria were the main predominant Eubacteria species in SMD granule, their contents were 49. 62% and 12. 03% in all 133 random Eubacteria clones respectively. The results of RTQ-PCR indicated that the content of Archaea was less than Eubacteria, the Archaea content in total microorganisms in SMD granule was about 27. 6%.  相似文献

从啤酒废水处理系统SBR池中取得的水样,分离纯化得到菌株35株,分别以进站原水和水解酸化池出水作为污水培养基,进行COD降解试验,复筛后分别得到3株COD降解率高且降解效果稳定的细菌12#,15#,23#与15#,25#,31#。将高效菌株与SBR池的活性污泥混合,考察其对啤酒废水的处理效果,结果表明,23#与活性污泥交互作用时在原水污水培养基中摇床培养16h COD降解率达到75.13%;25#与活性污泥交互作用处理水解酸化池出水120h时降解率达到82.40%。对4株高效菌株15#,23#,25#,31#进行16S rDNA序列测定,将所得序列与GenBank中已登录的相近细菌种群的DNA序列对比,得到高度同源性的细菌菌种。  相似文献
