Impact of Bioaugmentation with a Consortium of Bacteria on the Remediation of Wastewater-Containing Hydrocarbons (5 pp) 总被引:1,自引:0,他引:1

Goals, Scope and Background It has been observed that hydrocarbon treated wastewaters still contain high COD and a number of intermediates. This suggests
that the required catabolic gene pool for further degradation might be absent in the system or, that its titer value is not
significant enough. By providing the desired catabolic potential, the overall efficiency of the treatment system can be improved.
This study aims to demonstrate this concept by bioaugmentation of a lab-scale reactor treating refinery wastewater with a
consortium having the capacity to complement the alkB genotype to the available microbial population.
Methods Two reactors were set up using activated biomass collected from a refinery treatment plant and operated at a continuous mode
for a period of 8 weeks. The feed to both reactors was kept constant. Crude oil was spiked regularly. One reactor was bioaugmented
with a consortium previously described for crude oil spill remediation. The efficiency of the bioaugmented reactor was demonstrated
by reduced COD. The changes in the microbial population over a period of time were analyzed by RAPD. Catabolic activity of
the biomass in both reactors was monitored by PCR. The presence of the catabolic loci was confirmed by Southern Hybridization.
Results and Discussion 52.2% removal of COD was observed in the bioaugmented reactor while only 15.1% reduction of COD was observed in the reactor
without bioaugmentation. The change in microbial population can be seen from the 4th week, which also corresponds to improved
catabolic activity. The presence of the bedA locus was seen in all samples, which indicates the presence of aromatic degraders,
but the appearance of the alkB locus, from the 6th week onwards, which was observed only in the samples from the bioaugmented
reactor. The results suggest that the gene pool of the bioaugmented reactor has catabolic loci that can degrade accumulated
intermediates, thus improving the efficiency of the system.
Conclusions In this study, improvement of efficiency of bioremediation was demonstrated by addition of catabolic loci that are responsible
for degradation. Bioaugmentation was carried out in biomass that was collected from an ETP (effluent treatment plant) treating
hydrocarbon containing wastewater to study the strategies for improvement of the treatment system. Biostimulation, only marginally
improved the efficiency, when compared to bioaugmentation. The improved efficiency was demonstrated by COD removal. The presence
of the alkB locus suggests the importance of a catabolic gene pool that acts on accumulated intermediates. It is well documented
that straight chain aliphatics and intermediates of aromatic compounds after ring cleavage, accumulate in refinery wastewater
systems, thereby hindering further degradation of the wastewater. Supplementation of a catabolic gene pool that treats the
lower pathway compounds and alkanes will improve the overall efficiency. In this study, results suggest that the alkB locus
can also be used to monitor the degradative mode of the activated biomass.
Recommendations and Perspective . Pollution from petroleum and petroleum products around the globe are known to have grave consequences on the environment.
Bioremediation, using activated sludge, is one option for the treatment of such wastes. Effluent treatment plants are usually
unable to completely degrade the wastewater being treated in the biological unit (the aerator chambers). The efficiency of
degradation can be improved by biostimulation and bioaugmentation. This study demonstrates the improved efficiency of a treatment
system for wastewater containing hydrocarbons by bioaugmentation of a consortium that supports degradation. Further experiments
on a pilot scale are recommended to assess the use of bioaugmentation on a large scale. The use of molecular tools, like DNA
probes for alkB, to monitor the system also needs to be explored. 相似文献

针对修复焦化厂高浓度多环芳烃污染土壤高成本的现实,采用以非食用性植物油、生物柴油、表面活性剂及其乳化合成的微乳液为淋洗剂,比较不同淋洗剂的淋洗效果。结果表明乳化合成的微乳液对焦化厂土壤中多环芳烃的总去除率高于单独使用表面活性剂为淋洗剂对土壤中多环芳烃的总去除率,说明生物柴油及植物油与表面活性剂乳化形成的微乳液对原污染土壤中的多环芳烃具有显著的增溶作用。1%TW-80和2.5%TW-80对土壤中多环芳烃总去除率分别为11%和14%;以2.5%TW-80为原料乳化合成的微乳液的淋洗去除率较以1%TW-80为原料乳化合成的微乳液高,总去除率分别为15%~30%和11%~18%;以生物柴油为原料乳化合成的微乳液的淋洗去除率较以植物油为原料乳化合成的微乳液高,分别为17%~30%和15%~23%,且对多环芳烃的去除率与其辛醇水分配系数(logKow)呈线性相关关系。 相似文献

Three beehive matrices, sampled in six different apiaries from West France, were analyzed for the presence of four polycyclic aromatic hydrocarbons (PAH4: benzo[a]pyrene, benzo[a]anthracene, benzo[b]fluoranthene and chrysene). Samples were collected during four different periods in both 2008 and 2009. Honey samples showed the lowest levels of PAH4 contamination (min = 0.03 μg kg−1; max = 5.80 μg kg−1; mean = 0.82 μg kg−1; Sd = 1.17). Bee samples exhibited higher levels of PAH4 contamination (min = 0.32 μg kg−1; max = 73.83 μg kg−1; mean = 7.03 μg kg−1; Sd = 17.83) with a great dispersion of the concentrations due to four main events of high concentrations. Pollen samples showed only one major episode with the highest PAH4 concentration found (min = 0.33 μg kg−1; max = 129.41 μg kg−1; mean = 7.10 μg kg−1; Sd = 22.28). This study investigated environmental distributions and production mechanisms of chlorinated polycyclic aromatic hydrocarbons (Cl-PAHs) in the sediments from some tidal flats located in Asia. Cl-PAHs were found in sediments taken from Arao tidal flat, Kikuchigawa River and Shirakawa River. The range of ∑Cl-PAHs was from 25.5 to 483 pg g−1 for Kikuchigawa River and Arao tidal flat, respectively.Concentrations of PAHs and Cl-PAHs showed no significant correlations (r = 0.134). This result suggests that the origins of these compounds differ. In the identified Cl-PAH isomers, the most abundant Cl-PAH isomer was 9,10-dichloroanthracene (9,10-di-Cl-ANT) in the three sites. In general, concentrations of Cl-ANTs in the coastal environment are about 3-5 orders of magnitude lower than those of anthracene (ANT). However, concentration ratios between Cl-ANTs and ANT (Cl-ANTs/ANT) in the sediments ranged from 4.1% to 24.6%. This result indicated that Cl-PAHs were not generated under industrial processes but the high concentration ratios have resulted from the contribution of photochemical production of Cl-ANTs in the sediments because ANT is known to have high photochemical reactivity.For examining this phenomenon, ANT adsorbed onto glass beads was irradiated with UV under the mimicked field conditions of tidal flats. As a result, it was noticed that, while chlorinated derivatives were negligible in a light-controlled group, production of 2-Cl-ANT, 9-Cl-ANT and 9,10-diCl-ANT on the irradiated surface were found in this study. These results suggest that photochemical reaction of PAHs can be a potential source of the occurrence of Cl-PAHs in the coastal environment. 相似文献

The real-time detection of the mixing states of polycyclic aromatic hydrocarbons (PAHs) and nitro-PAHs in ambient particles is of great significance for analyzing the source, aging process, and health effects of PAHs and nitro-PAHs; yet there is still few effective technology to achieve this type of detection. In this study, 11 types of PAH and nitro-PAH standard samples were analyzed using a high performance-single particle aerosol mass spectrometer (HP-SPAMS) in lab studies. The identification principles ‘parent ions’ and ‘mass-to-charge (m/z) = 77’ of each compound were obtained in this study. It was found that different laser energies did not affect the identification of the parent ions. The comparative experiments of ambient atmospheric particles, cooking and biomass burning emitted particles with and without the addition of PAHs were conducted and ruled out the interferences from primary and secondary organics on the identification of PAHs. Besides, the reliability of the characteristic ions extraction method was evaluated through the comparative study of similarity algorithm and deep learning algorithm. In addition, the real PAH-containing particles from vehicle exhaust emissions and ambient particles were also analyzed. This study improves the ability of single particle mass spectrometry technology to detect PAHs and nitro-PAHs, and HP-SPAMS was superior to SPAMS for detecting single particles containing PAHs and nitro-PAHs. 以南京市城郊不同土地利用类型的农业土壤(水田、菜地和林地)为研究对象,测定了16种PAHs的含量.结果表明,苊烯(Acy)在所有土壤样本中均未被检出,南京城郊农业土壤15种ω(PAHs)的范围在24.49~925.54μg·kg-1之间,平均值为259.88μg·kg-1.PAHs含量由高到低依次为:林地>水田>菜地,总体上以高环PAHs(HMW)含量为主.不同土壤理化性质对PAHs的影响表明:土壤有机碳(TOC)和黏粒(clay)含量与PAHs存在一定的相关性,pH和全氮(TN)与PAHs无明显相关性.毒性当量法和CSI指数法表明,南京城郊农业土壤中PAHs生态风险较小,但是林地中应当给予一定的重视.增量终身癌症风险(ILCR)进行健康风险评价表明,儿童健康的威胁风险略大于成人,林地的总的致癌风险(CR)明显高于菜地和水田,仍处于可接受的范围内.对成人进行了蒙特卡洛模拟表明,确定性健康风险的风险分析低估了PAHs的健康风险.敏感性分析结果表明,对CR总方差影响最大的输入参数是暴露频率EF(占50.7%). 相似文献

The dynamics of total and active microbial communities were studied in seawater microcosms amended with crude or diesel oil at different temperatures (25, 10 and 4 °C) in the presence/absence of organic fertilization (Inipol EAP 22). Total and hydrocarbon-degrading microbes were enumerated by fluorescence microscopy and Most Probable Number (MPN) method, respectively. Total (16S rDNA-based) vs. active (16S rRNA) bacterial community structure was monitored by Capillary-Electrophoresis Single Strand Conformation Polymorphism (CE-SSCP) fingerprinting. Hydrocarbons were analyzed after 12 weeks of incubation by gas chromatography-mass spectrometry. Total and hydrocarbon-degrading microbial counts were highly influenced by fertilization while no important differences were observed between temperatures. Higher biodegradation levels were observed in fertilized microcosms. Temperature and fertilization induced changes in structure of total bacterial communities. However, fertilization showed a more important effect on active bacterial structure. The calculation of Simpson's diversity index showed similar trends among temperatures whereas fertilization reduced diversity index of both total and active bacterial communities. 相似文献

The distribution of polycyclic aromatic hydrocarbons was determined in surface sediments collected at 36 stations along the Spanish Northern continental shelf in March and September 2003, and February 2005. Concentrations of PAHs (Σ13 parent components) were in the range of 22-47528 μg/kg dw, the highest values corresponding to coastal urban-industrial hotspots and decreasing offshore. Sediment quality guidelines (SQGs) showed that concentrations of total PAHs were below the threshold effect level (TEC) in 27 stations (81%) and above in 7, two of which (Gijon and Bilbao) were above the probable effect concentration (PEC). The detailed study of diagnostic ratios suggested a rather uniform mixture of petrogenic and pyrolytic PAH sources along the continental shelf, with a slight decrease of the latter moving westwards and offshore. In order to assess the incidence of sediment sampling on the variability of the results, selected stations were also monitored in February and September 2004 and September 2005. The average field variance of the values obtained for each station was 31% that decreased to 23% when the values were normalized to TOC. 相似文献
