印楝素对亚洲玉米螟的生物活性及蛋白质合成的影响   总被引:2,自引:0,他引:2  
室内生物测定表明,印楝素对玉米螟幼虫具有明显的毒杀作用和抑制生长发育的活性.浓度3×10-6 ~15×10-6印楝素处理幼虫,均出现活动性降低、化蛹延迟、蛹出现畸形的现象,甚至形成永久性幼虫,且与处理浓度有一定的相关性.同时发现,经印楝素处理后,亚洲玉米螟48h蛹蛋白质的含量和组分与对照相比出现明显的差异,含量与处理浓度有较好的相关性.SDS-PAGE结果显示, 经3×10-6 ~15×10-6印楝素处理,昆虫的蛹均出现分子量Mr55×103、50×103两种新蛋白组分,减少了21. 6×103、20×103两种蛋白组分;另外10×10-6和15×10-6印楝素处理还减少了22×103蛋白组分, 15×10-6处理还出现9×103新蛋白组分; 这6种差异蛋白可能是印楝素分子作用的相关蛋白. 图2表1参20  相似文献

In order to achieve a more substantial appraisal of lake water quality, the assessment must not be based only on chemical measurements and analyses of the water itself, but even more so on the impact of existing conditions on aquatic biota. This is possible with the use of biotests or biomarkers, e.g. investigations of the developmental parameters (96-h embryotoxicity evaluate) or of the induction of heat shock proteins (proteotoxicity evaluate). To evaluate the suitability of these tests for environmental screening, fertilized zebrafish eggs were exposed to water samples collected from five sites of varying levels of stress from Laguna Lake, Philippines. Reconstituted water was used as laboratory control while water samples from a highly polluted freshwater source was used as positive control. Developmental parameters were noted and described within 48 and 96 h of exposure. Dilution experiments of the positive control were also done to further assess and compare toxicity potentials of Laguna Lake waters with those originating from a polluted freshwater. After the 96-h exposure, the levels of stress proteins (hsp 70) were determined in embryos from all exposure groups. Results showed 100% mortality in embryos exposed to undiluted positive control (PC) within only 12 h. Increasing dilution levels, however, resulted in lower mortality and lower abnormality rates. No detectable developmental differences were noted among embryos exposed to either the laboratory control or Laguna Lake waters at the end of 96 h, regardless of the source. Very high survival rates and high hatching success rates were observed in embryos exposed to lake waters as well as laboratory control, and the data did not differ significantly among the groups. Likewise, no significant malformations were noted among all developing embryos throughout the exposure period. However, the levels of heat shock proteins in the two sites located closest to Manila, the Philippine capital (Northern West Bay and Central West Bay), showed a pronounced elevation relative to the control, indicating that these stress proteins protect the embryos from the detrimental effects of pollutants in the water. Based on the 96-h early life stage (ELS) test, the water quality of the lake is good for fishery propagation despite the current levels of pollutants in the water. This finding is in accordance with the Class C status (i.e. suitable for fish growth and propagation) as given to the lake by the local environmental agency. On the other hand, data on proteotoxicity showed that the fish are under stress, presumably deriving from pollutants. This calls for a continuous monitoring and improvement of the lake water. The present study indicates that the two biomarker methods are very easy to use, practical, rapid, and sensitive for assessing water quality in a tropical lake and recommends for their incorporation into the future monitoring program of Laguna Lake.  相似文献

Soy protein isolate (SPI) was modified using sodium dodecyl sulfate (SDS) and guanidine hydrochloride (GuHCl). Adhesion performance of the modified SPI on fiberboard was studied. The Water-soluble mass of the modified SPI adhesives was examined following modified ASTM D5570. The SDS-modified SPI containing 91% protein had a water-soluble mass of 1.7%. To be considered a water-resistant adhesive, the water-soluble mass of adhesive should be less than 2%. The wet shear strength test showed 100% cohesive failure within fiberboard, indicating that the modified SPI has good water resistance. The effect of drying treatment on adhesion performance of the SDS-modified SPI on fiberboard was then investigated. Drying treatment significantly affected the final adhesion performance. Shear strength did not change much, but the percentage of cohesive failure within fiberboard increased markedly as drying temperature increased. All the unsoaked, soaked, and wet specimens glued by the adhesives treated at 70° or 90°C had 100% cohesive failure within fiberboard. Viscosity also increased greatly with an increase in drying temperature. This information will be useful in developing low-cost adhesive processing system in the future.  相似文献

应用利用弹性理论、温度应力场理论、热胀冷缩原理 ,对立井井壁破坏应力进行了较为精确的受力分析 ,给出了立井井壁应力分布规律的解析式。在整个理论的分析过程中 ,笔者着重分析了井壁应力中负摩擦力的影响 ,对负摩擦力引起的井壁应力进行了较为细致的计算 ,并通过具体算例进行了说明。笔者研究的结果 ,为井壁设计的理论提供了新的依据 ,同时对煤矿设计及煤矿工程应用有重要实用价值。  相似文献

This article describes a conductometric bi-layer based bienzyme biosensor for the detection of proteins as a marker of organic matter in rivers. Proteins were chosen to be used as indicators of urban pollution. The working mechanism of the bienzyme biosensor is based on the enzymatic hydrolysis of proteins into several fractions (peptides and amino acids), which results in a local conductivity change depending of the concentration of proteins. In this work, we began with the optimization of biosensor response using bovine serum albumin (BSA) as standard protein. For this objective seven enzymatic biosensors were prepared: four enzymatic sensors with only one layer of enzyme (proteinase K, trypsin, pronase or protease X) and three other enzymatic sensors with two layers (first layer: membrane containing proteinase K; second layer: one of the three other enzymes: trypsin, pronase or protease X). The biosensors were obtained through the deposition of enzymatic layers and the cross-linking process between enzymes and BSA in saturated glutaraldehyde vapour. The response of the various biosensors, described previously, were compared with the values of total organic carbon (TOC), and those of organic nitrogen (Norg), as determined by the laboratory accredits (CEMAGREF of Lyon) using the traditional method of analysis (NF EN 1484, infrared spectroscopy) and (NF EN 25663, mineralization/colorimetry assay) respectively for each water sample obtained from di erent sites in Lyon (France). The linear correlations obtained with the response of the seven biosensors showed the most important indices of correlations for the biosensor with two enzymatic layers: proteinase K + pronase (pkp). The optimum conditions for the preparation of the pkp biosensor increased the sensitivity and gave a limit of quantification of 0.583 g/L for TOC and 0.218 g/L for Norg in water samples. This sensor shows good reproducibility (2.28%), a capacity to be used at temperatures range 10– 30°C (depending on the season) and moreover a long lifetime (5 weeks).  相似文献

为揭示SigL及其增强子结合蛋白(EBPs)在苏云金芽胞杆菌(Bt)中的调控功能,在全基因组测序的基础上,采用生物信息学方法,对YBT-1520菌株的SigL及其EBPs的结构和功能进行深入分析.结果表明,YBT-1520基因组中存在1个SigL和6个EBPs,而且EBPs在结构上具有丰富的多样性,包含了EBPs的所有可能的结构域组织类型.SigL所调控的基因涉及11个假定的COG代谢途径,其中包括能量代谢、氨基酸代谢、翻译与细胞周期等.根据EBPs在基因组的位置推测,YBT-1520的EBPs参与γ-氨基丁酸代谢途径、精氨酸代谢途径、支链脂肪酸降解途径、多糖分解代谢等代谢途径的调控.本研究将为揭示Bt杀虫晶体蛋白大量表达的调控机制提供新的思路.  相似文献

本文介绍了超临界直流电站锅炉检验中发现的水冷壁开裂、变形,调风器鳍片烧损变形,集箱管接头角焊缝裂纹,刚性梁固定耳板处拉裂、变形等问题。对产生这些问题的原因进行了分析和研究,认为各水冷壁管子内水汽温度不均匀是造成拉裂、变形的主要原因。针对产生问题的原因,提出要预防此类问题发生就必须控制水冷壁超温和降低水冷壁各管子间温度差。并详细地论述了控制水冷壁超温和降低水冷壁各管子间温度差的八项具体改进措施。  相似文献

多溴联苯醚(PBDEs)和全氟烷基酸(PFAAs)是两类使用量大、环境污染广泛、人体暴露严重的新型有机污染物,2009年已纳入《斯德哥尔摩公约》持久性有机污染物(POPs)名单,但其毒性效应及作用机制并不明确.本文综述了本课题组近几年针对多溴联苯醚PBDEs和全氟烷基酸PFAAs的分子毒理机制研究工作,主要集中在这两类污染物对甲状腺系统、雌激素系统和肝脏脂肪酸代谢系统干扰效应的分子机制研究.本文分别从分子、细胞和活体三个层面,研究了污染物与核受体的直接结合作用、结合后受体的构象变化、细胞内受体的转录活性、以及活体暴露后受体调控基因的表达变化,由此阐明了污染物通过与受体直接作用导致细胞和活体生物功能改变的分子机制.同时结合计算模拟,探讨了污染物生物效应与其化学结构之间的关系,发现污染物的受体活性取决于它们与受体结合的空间构型,而其活性强度基本与二者的结合能力一致,主要受疏水作用和氢键的影响.此外,还通过研究污染物与天然配体转运蛋白的相互作用,明确了各个污染物与转运蛋白的结合能力,探讨了其构效关系,并评估了污染物对天然配体在体内转运过程的潜在干扰效应.通过上述研究工作,提出了多层面、多靶点研究环境污染物分子毒理机制的新思路,建立和引进了研究污染物与生物靶分子相互作用的新方法,发现了PBDEs、PFAAs与TR、ER、PPARγ核受体结合的新模式,为深入了解这些污染物的分子毒理机制提供了有用的信息和有效的研究手段.  相似文献

为探明妊娠早期胚胎的丢失是否与卵巢、输卵管、子宫组织受到2,3,7,8-四氯苯并二噁英(TCDD)直接毒害有关,检测了NIH小鼠胚胎着床前期和后期TCDD暴露对胚胎毒性影响的敏感性,并利用免疫组化方法分析了模型动物肝脏、子宫、输卵管和卵巢组织中TCDD所引起的AhR、ARNT以及Cyp1a2分子标记物的变化.检测发现:妊娠第9d,100ng·kg-1·d-1剂量TCDD经口染毒,造成胚胎着床数量减少,且着床前期暴露的影响大于着床后期;子宫蜕膜反应受到明显抑制;胚胎迁移率没有明显变化,但胚胎数量减少.免疫组织化学分析发现正常组小鼠的肝脏、子宫、输卵管和卵巢组织中有AhR和Cyp1a2弱阳性信号表达,ARNT有细胞核的强阳性信号表达;妊娠第1~8d、第1~3d和第4~8d处理组小鼠肝脏、子宫、输卵管和卵巢组织中的AhR、Cyp1a2的阳性面积和光密度值均高于正常组;随处理时间和组织蓄积量的增加,ARNT在组织中的变化由胞核(妊娠第1~3d组)表达到胞浆(妊娠第4~8d组)表达,然后完全无表达(妊娠第1~8d组).以上研究结果表明:TCDD对早期妊娠小鼠子宫、输卵管和卵巢组织中的AhR、ARNT和Cyp1a2的激活和代谢方式与肝脏相同,说明雌性生殖系统中的组织有TCDD蓄积和代谢活性,这可能是导致早期胚胎迁移、着床等过程改变,造成胚胎丢失的重要原因.  相似文献

污泥有机质(特别是胞外聚合物,EPS)高值产品回收将会推动污水/污泥资源化.然而,EPS典型提取方法效率较低、回收产物成分复杂,限制了EPS目标物质的高效提取、降低了回收物的潜在高值应用.为此,探究高值、高效EPS提取/回收技术极为重要.这就需要改变目前典型的"笼统"EPS提取方法,将"胞外多糖"和"胞外蛋白"这样的模糊提取物(实为"有机物混合物")转变为单目标物质(如类藻酸盐物质、酸溶性EPS、硫酸盐多糖、淀粉样蛋白质以及透明质酸等)或多目标物质(依靠物理、化学性质差异分离、提纯),实现产物的定向提取并保证其纯度,从而使回收产物获得高值利用.另一方面,把握进水水质、营养物负荷以及运行环境对污泥EPS形成的影响亦十分重要;同时,需要鉴定易形成EPS的优势菌并掌握其生理、生化特征,这对通过污水处理运行而"原位"增加EPS污泥本底含量具有非常重要的意义.此外,还需要针对高值组分进一步发展优化提取方法(如添加表面活性剂等),以实现回收物高效提取与高值回收利用.  相似文献
