Limits on the application of biosolids (anaerobically processed sludges from wastewater treatment plants) as fertilizers for the amendment of soil are becoming greater because of the accumulation of recalcitrant substances, making necessary the use of techniques that bring the concentration of xenobiotics to lower concentrations than those permitted. In general, the biosolids composting process is sufficient to reduce the usual concentration of linear alkylbenzene sulfonates (LAS) to low levels. In this work, an assessment is made on the effect of temperature in the capacity of enriched bacterial populations to biodegrade LAS, together with the influence that the available nutrients may have in the biodegradation of these compounds. The results show that the microbial metabolism of LAS was not observed in the thermophilic range. The optimum temperature for the biodegradation of LAS appears to be around 40 degrees C, this is, the lowest assayed here, and at this temperature the differences in the biodegradation of LAS among the nutritionally supplemented cultures are small.  相似文献

The goal of this work was to study the mechanical behavior of concrete with recycled Polyethylene Therephtalate (PET), varying the water/cement ratio (0.50 and 0.60), PET content (10 and 20 vol%) and the particle size. Also, the influence of the thermal degradation of PET in the concrete was studied, when the blends were exposed to different temperatures (200, 400, 600 °C). Results indicate that PET-filled concrete, when volume proportion and particle size of PET increased, showed a decrease in compressive strength, splitting tensile strength, modulus of elasticity and ultrasonic pulse velocity; however, the water absorption increased. On the other hand, the flexural strength of concrete-PET when exposed to a heat source was strongly dependent on the temperature, water/cement ratio, as well as on the PET content and particle size. Moreover, the activation energy was affected by the temperature, PET particles location on the slabs and water/cement ratio.  相似文献

Analysis and residue levels of forchlorfenuron (CPPU) in watermelons   总被引:2,自引:0,他引:2  
This paper describes the application of liquid chromatography-tandem mass spectrometry (LC/MS-MS) for analysis of residues of forchlorfenuron (CPPU), a new plant growth regulator, in watermelons, after a sample preparation step based on the buffered Quick, Easy, Cheap, Effective, Rugged and Safe extraction method. Analytical determinations were carried out in a triple quadrupole system fitted with an electrospray interphase operating in the positive ionisation mode (ESI+). Three simultaneous MS-MS transitions of the quasi-molecular ion m/z 248 (precursor ion) were monitored for data acquisition (248 > 129, 248 > 155, and 248 > 248), using the transition 248 > 129 for quantitation. Recovery studies on watermelons at levels of 1-200 microg/kg, performing five replicates at each level and using bracketing single-level calibration with matrix-matched standards for quantitation, gave forchlorfenuron mean recoveries ranging from 82 to 106% with relative standard deviations (RSD) lower than 18%. The limit of determination was established at 1 microg/kg. The method was applied to evaluate the persistence of forchlorfenuron residues in watermelons grown in plastic greenhouses, after applying an individual spray treatment to the flower ovary at the anthesis stage (45 mu g/flower and 60 mu g/flower for cv "Extazy" and cv "reina de corazones" watermelons, respectively). One month after treatment, 20 "Extazy" watermelon units (1.5-2 kg/unit) and 20 "Reina de corazones" watermelon units (4-5 kg/unit) were collected and analyzed individually. None of the samples contained forchlorfenuron residues higher than 1 microg/kg.  相似文献

A study is presented on the distribution of 234U, 238U, 235U isotopes in surface water of the Llobregat river basin (Northeast Spain), from 2001 to 2006. Sixty-six superficial water samples were collected at 16 points distributed throughout the Llobregat river basin. Uranium isotopes were measured by alpha spectrometry (PIPS detectors). The test procedure was validated according to the quality requirements of the ISO17025 standard. The activity concentration for the total dissolved uranium ranges from 20 to 261 mBq L−1. The highest concentrations of uranium were detected in an area with formations of sedimentary rock, limestone and lignite. A high degree of radioactive disequilibrium was noted among the uranium isotopes. The 234U/238U activity ratio varied between 1.1 and 1.9 and the waters with the lowest uranium activity registered the highest level of 234U/238U activity ratio. Correlations between uranium activity in the tested water and chemical and physical characteristics of the aquifer were found.  相似文献

We evaluate and compare the performance of Bayesian Monte Carlo (BMC), Markov chain Monte Carlo (MCMC), and the Generalized Likelihood Uncertainty Estimation (GLUE) for uncertainty analysis in hydraulic and hydrodynamic modeling (HHM) studies. The methods are evaluated in a synthetic 1D wave routing exercise based on the diffusion wave model, and in a multidimensional hydrodynamic study based on the Environmental Fluid Dynamics Code to simulate estuarine circulation processes in Weeks Bay, Alabama. Results show that BMC and MCMC provide similar estimates of uncertainty. The posterior parameter densities computed by both methods are highly consistent, as well as the calibrated parameter estimates and uncertainty bounds. Although some studies suggest that MCMC is more efficient than BMC, our results did not show a clear difference between the performance of the two methods. This seems to be due to the low number of model parameters typically involved in HHM studies, and the use of the same likelihood function. In fact, for these studies, the implementation of BMC results simpler and provides similar results to MCMC. The results of GLUE are, on the other hand, less consistent to the results of BMC and MCMC in both applications. The posterior probability densities tend to be flat and similar to the uniform priors, which can result in calibrated parameter estimates centered in the parametric space.  相似文献

The aim of this study was to develop a specific and highly sensitive method able to detect very low concentrations of Francisella tularensis in soil samples by real-time PCR (qPCR) with SYBR Green I. tul4 gene, which encodes the 17-kDa protein (TUL4) in F. tularensis strains, was amplified using a LightCycler (LC) device. We achieved a detection limit of 0.69 fg of genomic DNA from F. tularensis subp. holarctica live vaccine strain (LVS), corresponding to a value less than 3.4 genome equivalents per reaction. The qPCR was shown to be specific, highly sensitive and reproducible. In addition, we evaluated 2 new methods for recovering bacteria from soil based on 1-step filtration using glass fiber filters and PVDF filters. These filtration methods enabled us to recover F. tularensis efficiently from soil samples. As few as 50 CFU per 0.5 g of soil were detected by qPCR. Capture enzyme-linked immunosorbent assay (cELISA) allowed us to detect and quantify the amount of bacteria recovered from soil by an immunological method. Although qPCR was more sensitive than cELISA, we did not observe substantial differences in the amount of bacteria quantified by both methods.  相似文献

Joostella strains are emerging candidates for biosurfactant production. Here such ability was analyzed for Joostella strain A8 in comparison with Alcanivorax strain A53 and Pseudomonas strain A6, all previously isolated from hydrocarbon enrichment cultures made of polychaete homogenates. In pure cultures Joostella sp. A8 showed the highest stable emulsion percentage(78.33%), hydrophobicity rate(62.67%), and an optimal surface tension reduction during growth in mineral medium supplemented with diesel oil(reduction of about 12 mN/m), thus proving to be highly competitive with Alcanivorax and Pseudomonas strains. During growth in pure culture different level of biodegradation were detected for Alcanivorax strain A53(52.7%),Pseudomonas strain A6(38.2%) and Joostella strain A8(26.8%). When growing in consortia,isolates achieved similar abundance values, with the best efficiency that was observed for the Joostella-Pseudomonas co-culture. Gas-chromatographic analysis revealed an increase in the biodegradation efficiency in co-cultures(about 90%), suggesting that the contemporary action of different bacterial species could improve the process. Results were useful to compare the efficiencies of well-known biosurfactant producers(i.e. Pseudomonas and Alcanivorax representatives) with a still unknown biosurfactant producer, i.e. Joostella, and to confirm them as optimal biosurfactant-producing candidates.  相似文献

Feeding, respiration and growth rates of oyster (Ostrea edulis L.) larvae reared at five food levels were measured throughout the entire larval period. Energy budgets were derived as a function of alga (Isochrysis galbana Parke) food concentration. Ingestion rate (IR, cells h-1) and oxygen consumption rate ( , nl h-1) were almost isometric functions of larval size [ash-free dry weight, (AFDW, g)], characterized by the equations: IR=803.9 AFDW1.13 and =4.85 AFDW1.09. Ingested ration was directly correlated to cell concentration up to a maximum at 200 cells l-1, with further increases failing to support higher ingestion rates. Likewise, growth rate linearly increased with food ration up to 100 cells l-1 (max. growth efficiency,K 1=25%) and reached a maximum at 200 cells l-1 (growth rate=5.6 m d-1), with further increases in food not supporting significantly faster growth. Maintenance ration was 2 to 3% daily dry weight (DW); optimum ration increased during larval development from 5 to 20% DW; maximum ration was 20% DW. During larval rearing, an increasing feeding schedule of 50, 100 and 200 cells l-1 from Days 0, 5 and 10, respectively, is recommended.  相似文献
