Phenol, like many other organic solvents, is toxic to micro-organisms even at low concentrations. However, some micro-organisms can withstand this toxicity to a certain concentration. To observe the uptake mechanism of phenol, bacteria were isolatedfrom a petroleum refinery effluent and identified. Study was carried out to understand the effect of varying sub-lethal concentrations of phenol, on all the isolated individual bacterial cultures. Out of the bacteria isolated, Serratia liquefaciens was found to tolerate phenol concentration up to 1500 mg l-1. A microbial consortium of the isolated bacteriawas formulated and immobilized. Individual cultures were also immobilized and uptake of phenol by the immobilized micro-organisms was observed in a nutrient-rich and nutrient-stressed medium containing phenol as a sole source of carbon. A time-dependent uptake of phenol was exhibited by the micro-organismsin nutrient-stressed medium, after which a sudden increase in phenol concentration occurred in the extracellular medium, till it reached back to the initial concentration. This was attributedto an active efflux mechanism adopted by the micro-organisms to withstand the toxic shock.  相似文献

This paper reports results of an exploratory study of prenatal diagnosis patients who experienced voluntary terminations of pregnancy following the detection of an abnormality or spontaneous miscarriages. The 121 participants were part of the national collaborative Chorionic Villus Sampling and Amniocentesis Study. They completed semi-structured telephone interviews and mailed questionnaires at 1 month and 6 months after the pregnancy losses. Scores on the Profile of Mood States showed that mood levels improved significantly over time. However, there were some declines in loss-related support from partners and others. The persisting distress and difficulties of a minority highlight the variability in women's responses to pregnancy losses. Women who lost pregnancies later in gestation, showed the greatest mood disturbances at initial assessments, used professional mental health assistance after the loss, or reported less satisfactory loss-related support from significant others showed the greatest levels of mood disturbance at the six-month assessment. Follow-up contacts with patients who lose pregnancies should be used to inform women about the variation in possible grief reactions, to assess the extent of support the women are receiving from their partners and significant others, and to provide additional follow-up or referral of those experiencing the greatest distress.  相似文献

An apparently balanced t(2q;21q) translocation was discovered in fetal blood and amniocytes of a 22-week fetus, monitored because of ultrasonographic evidence of a heart disease. FISH (fluorescence in situ hybridization) analysis disclosed a complex translocation between chromosomes 2q, 18q, and 21q, which was inherited from the healthy mother. This observation corroborates the usefulness of molecular cytogenetic techniques in raising the quality of prenatal diagnosis and detecting subtle rearrangements not resolved by standard cytogenetics.  相似文献

Due to the amounts of chromium in the leachate resulting from leather leaching tests, chromium sulfate tanned leather wastes are very often considered hazardous wastes. To overcome this problem, one option could be recovering the chromium and, consequently, lowering its content in the leather scrap. With this objective, chromium leather scrap was leached with sulfuric acid solutions at low temperature also aiming at maximizing chromium removal with minimum attack of the leather matrix. The effects of leather scrap dimension, sulfuric acid and sodium sulfate concentration in the solutions, as well as extraction time and temperature on chromium recovery were studied, and, additionally, organic matrix degradation was evaluated. The best conditions found for chromium recovery were leather scrap conditioning using 25 mL of concentrated H2SO4/L solution at 293 or 313 K during 3 or 6 days. Under such conditions, 30–60 ± 5% of chromium was recovered and as low as 3–6 ± 1% of the leather total organic carbon (TOC) was dissolved. Using such treatment, the leather scrap area and volume are reduced and the residue is a more brittle material showing enhanced anaerobic biodegradability. Although good recovery results were achieved, due to the fact that the amount of chromium in eluate exceeded the threshold value this waste was still hazardous. Sandy beach habitat where sea turtles nest will be affected by multiple climate change impacts. Before these impacts occur, knowledge of how nest site selection and hatching success vary with beach microhabitats is needed to inform managers on how to protect suitable habitats and prepare for scientifically valid mitigation measures at beaches around the world. At a highly successful green turtle (Chelonia mydas) rookery at Akumal, Quintana Roo, Mexico, we measured microhabitat characteristics along the beach crawl (rejected sites) and related nest site conditions (selected sites) to subsequent hatching success rates for 64 nesting events. To our knowledge, this is the first study to report environmental data along the nesting crawl for a green turtle population and the first to use natural breaks in the data to describe their preferred habitat ranges. Our results indicate that turtles were likely using a combination of cues to find nest sites, mainly higher elevations and lower sand surface temperatures (Kruskal-Wallis test, H?=?19.84, p?<?0.001; H?=?10.78, p?<?0.001). Hatching success was significantly and negatively correlated to sand temperature at cloaca depth (Spearman’s ρ?=??0.27, p?=?0.04). Indeed, the preferred range for cloaca sand temperatures at the nest site (26.3–27.5 °C) had significantly higher hatching success rates compared to the highest temperature range (Tukey HSD?=?0.47, p?=?0.05). Sand temperatures at various depths were intercorrelated, and surface and cloaca depth sand temperatures were correlated to air temperature (ρ?=?0.70, p?=?0.00; ρ?=?0.26, p?=?0.04). Therefore, rising air temperatures could alter sand temperature cues for suitable nest sites, preferred nest site ranges, and produce uneven sex ratios or lethal incubating temperatures. Norovirus (NoV) is recognized as the most common cause of foodborne outbreaks. In 2014, an outbreak of acute gastroenteritis occurred on a cruise ship in Brazil, and NoV became the suspected etiology. Here we present the molecular identification of the NoV strains and the use of sequence analysis to determine modes of virus transmission. Food (cream cheese, tuna salad, grilled fish, orange mousse, and vegetables soup) and clinical samples were analyzed by ELISA, conventional RT-PCR, qRT-PCR, and sequencing. Genogroup GII NoV was identified by ELISA and conventional RT-PCR in fecal samples from 5 of 12 patients tested (41.7%), and in the orange mousse food sample by conventional RT-PCR and qRT-PCR. Two fecal GII NoV samples and the orange mousse GII NoV sample were successfully genotyped as GII.Pe (ORF 1), revealed 98.0–98.8% identities among them, and shared phylogenetically distinct cluster. Establishing the source of a NoV outbreak can be a challenging task. In this report, the molecular analysis of the partial RdRp NoV gene provided a powerful tool for genotyping (GII.Pe) and tracking of outbreak-related samples. In addition, the same fast and simple extraction methods applied to clinical samples could be successfully used for complex food matrices, and have the potential to be introduced in routine laboratories for screening foods for presence of NoV.  相似文献

Torque teno virus (TTV) was surveyed in tap water collected in schools from three municipalities located in the south of Brazil. TTV genomes were found in 11.7 % (4/34) of the samples. TTV DNA was detected in 10.5 % (2/19) of the samples collected at the city of Caxias do Sul and in 25 % (2/8) of the samples from Pelotas. Those cities have a low rate of sewage treatment. All samples from Santa Cruz do Sul, which has nearly 92 % of its sewage treated, were negative. These results suggest that the amount of sewage treated may have an effect on the detection rates of TTV DNA in drinking water in a given urban area, showing a mild negative correlation (r = ?0.76), when comparing the percentage of sewage treatment to the detection of TTV genomes. The detection rate of TTV was also compared with Escherichia coli, showing a strong correlation (r = 0.97), indicating that TTV may be a suitable marker of fecal contamination.  相似文献
