The continuing existence of zoos and their good purposes such as conservation, science, education, and recreation, can be ethically justified only if zoos guarantee the welfare of their animals. The usual criteria for measuring animal welfare in zoos are physical health, long life, and reproduction. This paper looks at these criteria and finds them insufficient. Additional criteria are submitted to expand the range of welfare considerations: natural and abnormal behavior; freedom and choice; and dignity. All these criteria should play a role in analyzing zoo animal welfare and interests but dignity has the overriding part because it impacts on both animal and human interests.  相似文献

Objective

Recent studies have integrated copy number variant (CNV) and gene analysis using target enrichment. Here, we transferred this concept to our routine genetics laboratory, which is not linked to centralized non-invasive prenatal testing (NIPT) facilities.

Method

From a cohort of 100 pregnant women, 22 were selected for the analysis of maternal genomic DNA (gDNA) along with fetal cell-free DNA. Using targeted enrichment, 135 genes were analyzed, combined with aberrations of chromosomes 21, 18, 13, X, and Y. The data were subjected to specificity and sensitivity analyses, and correlated with the results from invasive testing methods.

Results

The sensitivity/specificity was determined for the CNV analysis of chromosomes: 21 (80%/75%), 18 (-/82%), 13 (100%/67%), and Y (100%/100%). The gene detection was valid for maternal gDNA. However, for cell-free fetal DNA, it was not possible to determine the boundary between an artifact and a real sequence variant.

Conclusion

The target enrichment method combining CNV and gene detection seems feasible in a regular laboratory. However, this method can only be responsibly optimized with a sufficient number of controls and further validation on a strong bioinformatic background. The present results showed that NIPT should be performed in specialized centers, and that its introduction to isolated laboratories may not provide valid data.  相似文献

Sex allocation theory for sequential hermaphrodites predicts the size at which an individual should change sex, given the different relationships between individual size and reproductive success in the two sexes. We studied a host-parasite system where the myxozoan Kudoa ovivora infects the ovaries of the reef fish Thalassoma bifasciatum, a protogynous sequential hermaphrodite. The parasite sporulates in the hosts eggs and renders them infertile. It is thus expected to reduce the females reproductive success, and could thereby influence host sex change. We present data from marked fish we observed in the field over 4 months. The data suggest that females infected with Kudoa ovivora have a lower reproductive success, change sex earlier and at a smaller size than uninfected females. These results are in agreement with predictions from sex allocation theory, and provide the first example of a possible parasitic influence on the sex allocation of its host.Communicated by P. The Labyrinthulomycota are a relatively poorly studied group of heterotrophic unicellular eukaryotes. They comprise three lineages, labyrinthulids, thraustochytrids, and aplanochytrids, which are all primarily marine organisms and considered to be important components of marine microbial communities. Recently a number of Labyrinthulomycota have been implicated as parasites of marine (but also terrestrial) plants and marine molluscs. Here we describe a new species of thraustochytrid, Thraustochytrium caudivorum sp. nov. that we have isolated from laboratory cultures of Macrostomum lignano (Rhabditophora, Macrostomorpha), a marine free-living flatworm. In these worms T. caudivorum can cause lesions, which start at the tip of the tail plate and which can lead to the dissolution of the posterior part of the animal. Although the worms can frequently cure these lesions and regenerate the lost parts, the lesions can also result in the complete dissolution of the animal. We describe this thraustochytrid based on pure agar cultures and infestations in the worm cultures. Moreover, we describe its pathological effects on the worms and its morphology using both light and electron microscopy. In addition, we report a phylogenetic analysis using a partial 18S rDNA sequence that allows us to place this new species within the thraustochytrids. Finally, we outline a protocol that allows to permanently remove the parasites from infested worm cultures. We conclude that thraustochytrids represent a novel group of parasites of free-living flatworms.
