Uterine lavage affords the potential for non-invasive human blastocyst recovery, with obvious potential for preimplantation genetic diagnosis. In an effort to duplicate in women the multiple blastocyst recovery per cycle that can be achieved in several other species, we initiated a programme in which fertile women underwent superovulation, followed by lavage and embryo collection. We superovulated 15 fertile women, aged 21–40, in 29 cycles using one of four regimens. Insemination was by either intercourse or artificial intracervical donor insemination with cryopreserved sperm from men of proven fertility. In 28 of 29 cycles, the uterus was lavaged daily for 1, 2, or 3 days between 5 and 10 days after human chorionic gonadotropin (hCG) administration or luteinizing hormone (LH) surge. Almost total fluid volume was recovered in every lavage. There were no retained pregnancies and no complications. Surprisingly, only two morulae, one blastocyst, and four unfertilized ova were recovered. Thus, alterations in ovulation induction, insemination timing, or lavage techniques must be contemplated in order to increase the blastocyst yield and thus fulfil the potential of uterine lavage for preimplantation diagnosis.  相似文献

In 19 pregnancies at risk for 21-hydroxylase deficiency (21OHD) in 18 families with at lea one affected child, prenatal diagnosis was performed by RFLP analysis using the enzymi Taq I and EcoRI and the DNA probes specific for the 21 OH genes, the closely linke complement C4 genes and the highly polymorphic HLA class II genes DRB, DQB, and DPI For fetal DNA analysis either chorionic villi or cultivated amniotic cells were used. In all 1 cases, a clear prenatal diagnosis was possible either with the 21OH probe alone or in mo cases, by combining the results of the different closely linked loci.  相似文献

Repeated application may increase rates of pesticide dissipation in soil and reduce persistence. The potential for this to occur was investigated for the fungicide, tebuconazole (alpha-[2-(4-chlorophenyl)ethyl]-alpha-(1,1-dimethylethyl)-1H-1,2,4-triazole-1-ethanol), when used for peanut (Arachis hypogaea L.) production. Soil samples were collected from peanut plots after each of four tebuconazole applications at 2-wk intervals. Soil moisture was adjusted to field capacity as necessary and samples were incubated in the laboratory for 63 d at 30 degrees C. Untreated plot samples spiked with the compound served as controls. Results indicated accelerated dissipation in field-treated samples with the time to fifty percent dissipation (DT50) decreasing from 43 to 5 d after three tebuconazole applications. Corresponding increases in rates of accumulation and decay of degradates were also indicated. Best-fit equations (r2 = 0.84-0.98) to dissipation kinetic data combined with estimates of canopy interception rates were used to predict tebuconazole and degradates concentration in soil after each successive application. Predicted concentrations compared with values measured in surface soil samples were from twofold less to twofold greater. Use of kinetic data will likely enhance assessments of treatment efficacy and human and ecological risks from normal agronomic use of tebuconazole on peanut. However, the study indicated that varying soil conditions (in particular, soil temperature and water content) may have an equal or greater impact on field dissipation rate than development of accelerated dissipation. Results emphasize that extension of laboratory-derived kinetic data to field settings should be done with caution.  相似文献

The technical viability of using inert construction and demolition (C&D) materials for the construction of seawall and breakwater foundations has been established by laboratory testing of the materials, numerical analysis of foundation stability, and a pilot field-scale engineering performance evaluation. However, quality control measures are still required so that only suitable materials are used for seawall and breakwater foundation construction. The development of different quality control measures for different site conditions is presented in this paper. The rationale, practicality, and implementation of these quality control measures are also discussed.  相似文献

This study examines the feasibility of coupling a Catalytic Wet Air Oxidation (CWAO), with activated carbon (AC) as catalyst, and an aerobic biological treatment to treat a high-strength o-cresol wastewater. Two goals are pursued: (a) To determine the effect of the main AC/CWAO intermediates on the activated sludge of a municipal WasteWater Treatment Plant (WWTP) and (b) To demonstrate the feasibility of coupling the AC/CWAO effluent as a part of the influent of a municipal WWTP. In a previous study, a high-strength o-cresol wastewater was treated by AC/CWAO aiming to establish the distribution of intermediates and the biodegradability enhancement. In this work, the biodegradability, toxicity and inhibition of the most relevant intermediates detected in the AC/CWAO effluent were determined by respirometry. Also, the results of a pilot scale municipal WWTP study for an integrated AC/CWAO-aerobic biological treatment of this effluent are presented. The biodegradation parameters (i.e. maximum oxygen uptake rate and oxygen consumption) of main AC/CWAO intermediates allowed the classification of the intermediates into readily biodegradable, inert or toxic/inhibitory compounds. This detailed study, allowed to understand the biodegradability enhancement exhibited by an AC/CWAO effluent and to achieve a successful strategy for coupling the AC/CWAO step with an aerobic biological treatment for a high-strength o-cresol wastewater. Using 30%, as COD, of AC/CWAO effluent in the inlet to the pilot scale WWTP, the integrated AC/CWAO-biological treatment achieved a 98% of total COD removal and, particularly, a 91% of AC/CWAO effluent COD removal without any undesirable effect on the biomass.  相似文献
