Forest fragments have biodiversity value that may be enhanced through management such as control of non‐native predators. However, such efforts may be ineffective, and research is needed to ensure that predator control is done strategically. We used Bayesian hierarchical modeling to estimate fragment‐specific effects of experimental rat control on a native species targeted for recovery in a New Zealand pastoral landscape. The experiment was a modified BACI (before‐after‐control‐impact) design conducted over 6 years in 19 forest fragments with low‐density subpopulations of North Island Robins (Petroica longipes). The aim was to identify individual fragments that not only showed clear benefits of rat control, but also would have a high probability of subpopulation growth even if they were the only fragment managed. We collected data on fecundity, adult and juvenile survival, and juvenile emigration, and modeled the data in an integrated framework to estimate the expected annual growth rate (λ) of each subpopulation with and without rat control. Without emigration, subpopulation growth was estimated as marginal (λ = 0.95–1.05) or negative (λ = 0.74–0.90) without rat control, but it was estimated as positive in all fragments (λ = 1.4–2.1) if rats were controlled. This reflected a 150% average increase in fecundity and 45% average increase in adult female survival. The probability of a juvenile remaining in its natal fragment was 0.37 on average, but varied with fragment connectivity. With juvenile emigration added, 6 fragments were estimated to have a high (>0.8) probability of being self‐sustaining (λ > 1) with rat control. The key factors affecting subpopulation growth rates under rat control were low connectivity and stock fencing because these factors were associated with lower juvenile emigration and higher fecundity, respectively. However, there was also substantial random variation in adult survival among fragments, illustrating the importance of hierarchical modeling for fragmentation studies. In the present investigation, the oxidative metabolism of 14C-labeled metamitron was examined in plant cell cultures of tobacco overexpressing human P450 enzymes CYP1A1 or CYP1A2; special interest was in the aromatic hydroxylation of the herbicide. The oxidative metabolites deaminometamitron (DAM) and 4-hydroxydeaminometamitron (4-HDAM) were found in the untransformed control culture as well as in the transgenic culture. The transgenic cultures, however, exhibited higher turnover rates after 48 h of incubation with 20 μg 14C-metamitron per assay (untransformed: 40%, CYP1A1: 80%, CYP1A2: 100%). Primary metabolite 4-HDAM was partially found in glucosylated form in the transgenic cultures. As minor oxidative metabolites, 6-hydroxyphenyl-3-methoxymethyl-1,2,4-triazine-5(4H)-one and 3-hydroxymethyl-6-phenyl-1,2,4-triazine-5(4H)-one were identified in the transgenic cultures by GC-MS, LC-MS. Additionally, it could be demonstrated that both foreign enzymes (CYP1A1, CYP1A2) also catalyzed the deamination of metamitron. In a large-scale study (up to 400 μ g per assay) with the transgenic culture expressing CYP1A2, the high efficiency of this P450 system toward metamitron was demonstrated: turnover of the xenobiotic was almost complete with 400 μ g. Since large portions of unglucosylated 4-H-DAM were found, the activity of foreign CYP1A2 apparently exceeded that of endogenous O-glucosyltransferases of the tobacco cell culture. We concluded that in comparison to the nontransformed cell culture, the extent of metabolism was considerably higher in the transgenic cultures. The transgenic cell cultures expressing human CYP1A1 or CYP1A2 are thus suitable tools for the production of large quantities of primary oxidized metabolites of metamitron.  相似文献

Abstract:  Captive breeding of animals is widely used to manage endangered species, frequently with the ambition of future reintroduction into the wild. Because this conservation measure is very expensive, we need to optimize decisions, such as when to capture wild animals or release captive-bred individuals into the wild. It is unlikely that one particular strategy will always work best; instead, we expect the best decision to depend on the number of individuals in the wild and in captivity. We constructed a first-order Markov-chain population model for two populations, one captive and one wild, and we used stochastic dynamic programming to identify optimal state-dependent strategies. The model recommends unique sequences of optimal management actions over several years. A robust rule of thumb for species that can increase faster in captivity than in the wild is to capture the entire wild population whenever the wild population is below a threshold size of 20 females. This rule applies even if the wild population is growing and under a broad range of different parameter values. Once a captive population is established, it should be maintained as a safety net and animals should be released only if the captive population is close to its carrying capacity. We illustrate the utility of this model by applying it to the Arabian oryx ( Oryx leucoryx ). The threshold for capturing the entire Arabian oryx population in the wild is 36 females, and captive-bred individuals should not be released before the captive facilities are at least 85% full.  相似文献
