DNA-based population analysis was applied in combination with Raman spectrometry and Environmental Scanning Electron Microscopy for the characterisation of natural biofilms from sand and activated carbon filters operated for a long term at a municipal waterworks. Whereas the molecular biology polymerase chain reaction combined with denaturing gradient gel electrophoresis approach provides a deeper insight into the bacterial biofilm diversities, Raman spectrometry analyses the chemical composition of the extracellular polymer substances (EPS), microorganisms embedded in EPS as well as other substances inside biofilm (inorganic compounds and humic substances). Microscopy images the spatial distribution of biofilms on the two different filter materials. In addition, bacterial bulk water populations were compared with biofilm consortia using the molecular fingerprint technique mentioned.Population analysis demonstrated the presence of more diverse bacterial species embedded in a matrix of EPS (polysaccharides, peptides, and nucleic acids) on the sand filter materials. In contrast to this, activated carbon granules were colonised by reduced numbers of bacterial species in biofilms. Besides α-, β-, and γ-Proteobacteria, a noticeable specific colonisation with Actinobacteria was found on activated carbon particles. Here, the reduced biofilm formation came along with a decreased EPS synthesis. The taxonomy profiles of the different biofilms revealed up to 60% similarity on the same filter materials and 32% similarity of different materials. Similarity of adherent communities from filter materials and bulk water populations from the filter effluent varied between 36% and 58% in sand filters and 6–40% in granular activated carbon filters.The biofilm investigation protocols are most crucial to subsequent acquisition of knowledge on biofilm dynamics and bacterial contributions to transformation or adsorption processes in waterworks facilities.  相似文献

We studied if the levels of copper released from antifouling treated nets used in finfish mariculture could affect the immune defense mechanism and/or induce oxidative stress in Dicentrarchus labrax, after short term exposure in laboratory experiments. Dissolved copper concentration released from the treated nets, copper bioavailability and a set of biomarkers responses were measured. Biomarkers included hemoglobin concentration, activities of lysozyme, total complement, respiratory burst, glutathione S-transferase and acetycholinesterase and concentration of thiobarbituric acid reactive substances. Results indicated elevated copper concentration in seawater (184 μg L−1) but low concentration in muscle (1.5 μg g−1) and liver (117 μg g−1). Copper bioavailability was independent of copper complexes with dissolved organic carbon. However, formation of copper complexes with other matrices could neither be excluded nor justified. The released copper from the treated nets did not induce oxidative stress but affected the immediate immune defense mechanism of the exposed fish making them more easily vulnerable to diseases. Consequently, copper-based antifouling treated nets could be a risk factor for D. labrax health.  相似文献

Consumers influence climate change through their consumption patterns and their support or dismissal of climate mitigation policy measures. Both climate-friendly actions and policy support comprise a broad range of options, which vary in manifold ways and, therefore, might be influenced by different factors. The aims of the study were, therefore, two-fold: first, we intended to find a meaningful way to classify different ways of addressing climate change. Second, we aimed to examine which determinants influence people's willingness to engage in these behaviors. We conducted a large-scale mail survey in Switzerland in which respondents rated, among other items, their willingness to act or support a range of possible actions and mitigations measures. A principal component analysis indicated that a distinction in terms of a behavior's directness as well as a differentiation according to perceived costs seem to be appropriate to classify climate-friendly actions. Multiple regression analyses showed that perceived costs and perceived climate benefit turned out to be the strongest predictors for willingness to act or to support climate policy measures. The strong influence of perceived climate benefit might reflect a strategy of reducing cognitive dissonance. As high-cost behaviors are more difficult to adopt, consumers may reduce dissonance by dismissing high-cost behaviors as not effective in terms of climate mitigation. Political affiliation proved to be another strong determinant of willingness to act or support. Participants on the right wing were less willing to show indirect climate-friendly behaviors, change their mobility behaviors, and to support any type of climate mitigation policy measures. Climate-friendly low-cost behaviors, however, were not influenced by political affiliation.  相似文献

Environmental justice studies that focus on the management of municipal solid waste (MSW) typically examine the unequal distribution of associated health and environmental risks in minority social groups and the political processes that generate these inequalities. With the aim to complement current views on the field, in this work, we explore whether there is an issue of environmental justice in municipal systems' grade of self-sufficiency in treating the MSW that they generate and in their effort to close their material cycles. The methodology used is based on the concept of urban metabolism and is applied to 12 coastal municipalities of Barcelona's Metropolitan Region in Spain. The metabolism of the MSW flows of each system is analysed to examine (i) the system's efficiency to close its MSW cycles, corresponding to an indicator of environmental sustainability, and (ii) the MSW export and import flows, as an indicator of social sustainability. The results demonstrate a positive correlation between socioeconomic status and the externalisation of MSW treatment-related hazards. The proposed indicator proves to be an excellent tool for the evaluation of both the environmental and social performance of a system considering MSW management.  相似文献

Stakeholder analysis means many things to different people. Various methods and approaches have been developed in different fields for different purposes, leading to confusion over the concept and practice of stakeholder analysis. This paper asks how and why stakeholder analysis should be conducted for participatory natural resource management research. This is achieved by reviewing the development of stakeholder analysis in business management, development and natural resource management. The normative and instrumental theoretical basis for stakeholder analysis is discussed, and a stakeholder analysis typology is proposed. This consists of methods for: i) identifying stakeholders; ii) differentiating between and categorising stakeholders; and iii) investigating relationships between stakeholders. The range of methods that can be used to carry out each type of analysis is reviewed. These methods and approaches are then illustrated through a series of case studies funded through the Rural Economy and Land Use (RELU) programme. These case studies show the wide range of participatory and non-participatory methods that can be used, and discuss some of the challenges and limitations of existing methods for stakeholder analysis. The case studies also propose new tools and combinations of methods that can more effectively identify and categorise stakeholders and help understand their inter-relationships.  相似文献

Predator-prey relationships are often altered as a result of human activities. Where prey are legally protected, conservation action may include lethal predator control. In the Columbia River basin (Pacific Northwest, USA and Canada), piscivorous predators have been implicated in contributing to a lack of recovery of several endangered anadromous salmonids (Oncorhynchus spp.), and lethal and nonlethal control programs have been instituted against both piscine and avian species. To determine the consequences of avian predation, we used a bioenergetics approach to estimate the consumption of salmonid smolts by waterbirds (Common Merganser, California and Ring-billed Gull, Caspian Tern, Double-crested Cormorant) found in the mid-Columbia River from April through August, 2002-2004. We used our model to explore several predator-prey scenarios, including the impact of historical bird abundance, and the effect of preserving vs. removing birds, on smolt abundance. Each year, <1% of the estimated available salmonid smolts (interannual range: 44,830-109,209; 95% CI = 38,000-137,000) were consumed, 85-98% away from dams. Current diet data combined with historical gull abundance at dams suggests that past smolt consumption may have been 1.5-3 times current numbers, depending on the assumed distribution of gulls along the reaches. After the majority (80%) of salmonid smolts have left the study area, birds switch their diet to predominantly juvenile northern pikeminnow (Ptychocheilus oregonensis), which as adults are significant native salmonid predators in the Columbia River. Our models suggest that one consequence of removing birds from the system may be increased pikeminnow abundance, which--even assuming 80% compensatory mortality in juvenile pikeminnow survival--would theoretically result in an annual average savings of just over 180,000 smolts, calculated over a decade. Practically, this suggests that smolt survival could be maximized by deterring birds from the river when smolts are present, allowing bird presence after the diet switch to act as a tool for salmonid-predator control, and conducting adult-pikeminnow control throughout. Our analysis demonstrates that identifying the strength of ecosystem interactions represents a top priority when attempting to manage the abundance of a particular ecosystem constituent, and that the consequences of a single-species view may be counterintuitive, and potentially counterproductive.  相似文献

High winter-time PM10, sulfate, nitrate and ammonium levels in Istanbul were investigated using a high resolution WRF/CMAQ mesoscale model system. A model-ready anthropogenic emission inventory on 2 km spatial resolution was developed for the area and the present study is the first attempt to test these emissions. The results suggested that the system was capable of producing the magnitudes. PM10 levels calculated by the model underestimated the observations with an average of 10 per cent at Bogazici University sampling station, whereas an overestimation of 12 per cent is calculated for all stations. High uncertainties, particularly in traffic and coal combustion, led to over estimations around emission hot spots. Base case results together with the sensitivity studies pointed significant contribution of local sources, pointing to the need of control strategies focusing on primary particulate emissions.  相似文献
