We report here the first integrated investigation of both ancient DNA and proteins in archaeobotanical samples: medieval grape (Vitis vinifera L.) seeds, preserved by anoxic waterlogging, from an early medieval (seventh–eighth century A.D.) Byzantine rural settlement in the Salento area (Lecce, Italy) and a late (fourteenth–fifteenth century A.D.) medieval site in York (England). Pyrolysis gas chromatography mass spectrometry documented good carbohydrate preservation, whilst amino acid analysis revealed approximately 90% loss of the original protein content. In the York sample, mass spectrometry-based sequencing identified several degraded ancient peptides. Nuclear microsatellite locus (VVS2, VVMD5, VVMD7, ZAG62 and ZAG79) analysis permitted a tentative comparison of the genetic profiles of both the ancient samples with the modern varieties. The ability to recover microsatellite DNA has potential to improve biomolecular analysis on ancient grape seeds from archaeological contexts. Although the investigation of five microsatellite loci cannot assign the ancient samples to any geographic region or modern cultivar, the results allow speculation that the material from York was not grown locally, whilst the remains from Supersano could represent a trace of contacts with the eastern Mediterranean.  相似文献

To detect threats and reduce predation risk prey animals need to be alert. Early predator detection and rapid anti-predatory action increase the likelihood of survival. We investigated how foraging affects predator detection and time to take-off in blue tits (Parus caeruleus) by subjecting them to a simulated raptor attack. To investigate the impact of body posture we compared birds feeding head-down with birds feeding head-up, but could not find any effect of posture on either time to detection or time to take-off. To investigate the impact of orientation we compared birds having their side towards the attacking predator with birds having their back towards it. Predator detection, but not time to take-off, was delayed when the back was oriented towards the predator. We also investigated the impact of foraging task by comparing birds that were either not foraging, foraging on chopped mealworms, or foraging on whole ones. Foraging on chopped mealworms did not delay detection compared to nonforaging showing that foraging does not always restrict vigilance. However, detection was delayed more than 150% when the birds were foraging on whole, live mealworms, which apparently demanded much attention and handling skill. Time to take-off was affected by foraging task in the same way as detection was. We show that when studying foraging and vigilance one must include the difficulty of the foraging task and prey orientation.Communicated by P.A. Bednekoff  相似文献

Weathering rates of base cations are crucial in critical load calculations and assessments of sustainable forestry. The weathering rate on a single site with detailed geological data can be modelled using the PROFILE model. For environmental assessments on a regional scale, the weathering rates for sites are scaled into regional maps. The step from sites to regional level requires focus on the spatial variation of weathering rates. In this paper, a method is presented by which weathering rates are calculated for 25589 Swedish sites with total elemental analysis for the soil. Based on a part of the results, a methodology for creating area covering maps by geostatistical analysis and kriging is described. A normative reconstruction model was used to transform total elemental analysis to mineralogy. Information from the Swedish Forest Inventory database and other databases were used to derive texture and other important information for the sites, e.g. climate, deposition and vegetation data. The calculated weathering rates show a regional pattern that indicates possibilities for interpolation of data in large parts of Sweden. Geostatistical analysis of an area in southern Sweden shows different properties for different base cations. Kriging was performed for potassium to demonstrate the method. It was concluded that different base cations and different regions have to be analysed separately, in order to optimise the kriging method.  相似文献

Many South African populations of the brown mussel Perna perna have been depleted through over-exploitation by subsistence harvesters. This is problematic because recovery after disturbance is very slow, partly because recruits are largely associated with adult mussels. However, unlike large recruits of 3.5–10 mm that exhibit spatial structure related to that of adults, a very high proportion of settlers and small recruits (0–3.5 mm) occur on foliose algae. We tested the hypothesis that recruits on algae move to adult mussel beds after a period of growth, with the null hypothesis that they die at a certain size. We conducted an indirect field study comparing the ratios of large to small recruits in 100% cover mussel patches at locations with high and low algal cover. A second laboratory experiment analysed whether the size of recruits on algae affects their active movement behaviour in response to nearby mussel patches. Large/small recruit ratios were slightly, but not significantly greater in high than low algal cover locations. Both small (2–2.5 mm) and medium (4.5–5.5 mm) recruits remained on algae and moved very short distances throughout the laboratory experiment, while larger recruits (9–10 mm) moved significantly further distances and more often into mussel patches. The results suggest that very large recruits are able to migrate actively to nearby mussel patches, indicating ontogenetic shifts in this behaviour. However, the absence of a significant difference in ratios between field locations with high and low algal cover suggests many large recruits are accidentally dislodged from the algae and presumably die. Thus settlement of P. perna onto algae is likely to be wasted, with consequences for sustainable management of the mussel resource.  相似文献

An improved on-site characterization of humic-rich hydrocolloids and their metal species in aquatic environments was the goal of the present approach. Both ligand exchange with extreme chelators (diethylenetetraaminepentaacetic acid (DTPA), ethylendiaminetetraacetic acid (EDTA)) and metal exchange with strongly competitive cations (Cu(II) were used on-site to characterize the conditional stability and availability of colloidal metal species in a humic-rich German bogwater lake (Venner Moor, Münsterland). A mobile time-controlled tangential-flow ultrafiltration technique (cut-off: 1 kDa) was applied to differentiate operationally between colloidal metal species and free metal ions, respectively. DOC (dissolved organic carbon) and metal determinations were carried out off-site using a home-built carbon analyzer and conventional ICP-OES (inductively-coupled plasma-optical emission spectrometry), respectively. From the metal exchange equilibria obtained on-site the kinetic and thermodynamic stability of the original metal species (Fe, Mn, Zn) could be characterized. Conditional exchange constants Kex obtained from aquatic metal species and competitive Cu(II) ions follow the order Mn > Zn > > Fe. Obviously, Mn and Zn bound to humic-rich hydrocolloids are very strongly competed by Cu(II) ions, in contrast to Fe which is scarcely exchangeable. The exchange of aquatic metal species (e.g. Fe) by DTPA/EDTA exhibited relatively slow kinetics but rather high metal availabilities, in contrast to their Cu(II) exchange.  相似文献

Using tracking radars, we investigated the variability of flight directions of long-distance nocturnal passerine migrants across seasons (spring versus autumn migration) and sites at the southern (56° N) and northern (68° N) ends of the Scandinavian Peninsula (Lund versus Abisko). Whilst most migrants at Lund are on passage to and from breeding sites in Fennoscandia, the majority of the migrants at Abisko are close to their breeding sites, and migration at Abisko thus to a large degree reflects initial departure from breeding sites (autumn) or final approach to breeding destinations (spring). The radar data were used to test predictions about differences in orientation and wind drift effects between adult and juvenile birds (a large proportion of autumn migrants consists of juvenile birds on their first journey), between situations far away from or near the goals and between different phases of migration (initial departure, en route passage, final approach to goal). The concentrations (both total and within-night concentrations) of flight directions differed significantly between seasons as well as sites, with the highest concentration at Lund in spring (mean vector length of track directions, r = 0.79) and lowest at Abisko during spring (r = 0.35). Partial wind drift and partial compensation were recorded at Lund, with a similar effect size in spring and autumn, whilst possible wind drift effects at Abisko were obscured by the large directional scatter at this site. The results from Lund support the prediction that the high proportion of juveniles in autumn contributes to increase the directional scatter during this season, whilst there was no support for predictions of differential wind drift effects between seasons and situations with different goal distances. The most striking and surprising result was the exceedingly large scatter of flight directions at Abisko, particularly in spring. We suggest that such an exaggerated scatter may be associated with final approach orientation, where migrants reach their specific goals from all various directions by final navigation within a more wide-ranging goal region. The larger scatter of autumn flight directions at Abisko compared to Lund may be due to exploratory flights in variable directions being more common at initial departure from breeding sites than later during migratory passage. These surprising results highlight the importance of studying and analysing orientation during final approach to (and initial departure from) migratory goals for understanding the orientation systems of migratory birds.  相似文献

In a case of hydrops fetalis, serological examination showed a recent maternal human parvovirus B19 infection. Amniocentesis revealed a unique unbalanced translocation between chromosomes 3 and 11 of the fetus. The mother proved to have a balanced reciprocal translocation between chromosomes 3 and 11. A grossly macerated hydropic male fetus was delivered with a flat nose and low implanted deformed ears. Histopathological examination revealed nuclear inclusion bodies in fetal erythroid cells, confirming human parvovirus B19 infection. Parvovirus B19 DNA was demonstrated by in situ hybridization in the nuclei of heart muscle cells. Our finding of two different disorders in one case illustrates the importance of a complete evaluation of every case of hydrops fetalis, especially concerning counselling on the outcome of future pregnancies. The human parvovirus B19 infection will not recur due to the acquired immunity of the mother, whereas the balanced reciprocal translocation will endanger future pregnancies.  相似文献

Conservation and management of marine biodiversity depends on biomonitoring of marine habitats, but current approaches are resource-intensive and require different approaches for different organisms. Environmental DNA (eDNA) extracted from water samples is an efficient and versatile approach to detecting aquatic animals. In the ocean, eDNA composition reflects local fauna at fine spatial scales, but little is known about the effectiveness of eDNA-based monitoring of marine communities at larger scales. We investigated the potential of eDNA to characterize and distinguish marine communities at large spatial scales by comparing vertebrate species composition among marine habitats in Qatar, the Arabian Gulf (also known as the Persian Gulf), based on eDNA metabarcoding of seawater samples. We conducted species accumulation analyses to estimate how much of the vertebrate diversity we detected. We obtained eDNA sequences from a diverse assemblage of marine vertebrates, spanning 191 taxa in 73 families. These included rare and endangered species and covered 36% of the bony fish genera previously recorded in the Gulf. Sites of similar habitat type were also similar in eDNA composition. The species accumulation analyses showed that the number of sample replicates was insufficient for some sampling sites but suggested that a few hundred eDNA samples could potentially capture >90% of the marine vertebrate diversity in the study area. Our results confirm that seawater samples contain habitat-characteristic molecular signatures and that eDNA monitoring can efficiently cover vertebrate diversity at scales relevant to national and regional conservation and management.  相似文献

This study was conducted in the Swedish sub-Arctic, near Abisko, in order to assess the direction and scale of possible vegetation changes in the alpine–birch forest ecotone. We have re-surveyed shrub, tree and vegetation data at 549 plots grouped into 61 clusters. The plots were originally surveyed in 1997 and re-surveyed in 2010. Our study is unique for the area as we have quantitatively estimated a 19% increase in tree biomass mainly within the existing birch forest. We also found significant increases in the cover of two vegetation types—“birch forest-heath with mosses” and “meadow with low herbs”, while the cover of snowbed vegetation decreased significantly. The vegetation changes might be caused by climate, herbivory and past human impact but irrespective of the causes, the observed transition of the vegetation will have substantial effects on the mountain ecosystems.  相似文献
