Few studies on the genotoxicity of mercury compounds have been carried out in Drosophila melanogaster, most of them focused in the effects on germinal cells, whereas studies in somatic cells are scarce. In the present study we have analyzed for the first time the genotoxic activity of mercury (II) chloride (MC) and methyl mercury (II) chloride (MMC) in the in vivo wing somatic mutation and recombination test in Drosophila, also known as the wing spot assay. This test is based on the principle that loss of heterozygosis and the corresponding expression of the suitable recessive markers, multiple wing hairs (mwh) and flare-3 (flr(3)), can lead to the formation of mutant clones in larval cells, which are then expressed as spots on the wings of adult flies. The mercury compounds were supplied to third instar larvae (72+/-2h old) at concentrations ranging from 1 to 50 microM for mercury chloride (MC) and from 0.5 to 5 microM for methyl mercury chloride (MMC). Both mercury compounds showed high toxicity; however, MMC was more toxic than MC. The results showed that none of the three categories of mutant spots recorded (small, large, and twin) increased significantly by the treatments, independently of the dose supplied, indicating that the mercury compounds tested exhibit a lack of genotoxic activity in the wing spot assay of D. melanogaster. A forensic approach was used to evaluate sediments from Portão Stream, including analysis of metals, carbon (C) and nitrogen (N) stable isotopes, and C:N ratios. Samples collected at various points located along the stream were tested in order to investigate a possible illegal leachate input. The studied stream is heavily impacted by sewage and industrial discharges from two cities along its course. Among the metals analyzed, chromium (Cr) was noticeably the main pollutant, showing the highest levels, above regulatory limits, downstream from some potential sources of effluents enriched with this metal. Isotope analyses revealed a general trend of depletion in the heavier isotope along the stream for C and N. The exception was one point near a hazardous waste landfill, where relatively more enriched δ13C and δ15N values were found. The isotope and metal analysis results indicated that this site was affected by a particular source, demonstrating the combination of these parameters could be used for the discrimination of sources in a heavily polluted stream. Nevertheless, further investigations are necessary to provide a comprehensive evaluation of the biogeochemical processes involved in the incorporation of leachate in sediments to use this analysis as evidence for the illegal leachate discharge. 相似文献
To distinguish between pyrogenic and biological sources of PAHs in a tropical rain forest near Manaus, Brazil, we determined the concentrations of 21 PAHs in leaves, bark, twigs, and stem wood of forest trees, dead wood, mineral topsoil, litter layer, air, and Nasutitermes termite nest compartments. Naphthalene (NAPH) was the most abundant PAH with concentrations of 35 ng m(-3) in air (>85% of the sum of 21PAHs concentration), up to 1000 microg kg(-1) in plants (>90%), 477 microg kg(-1) in litter (>90%), 32 microg kg(-1) in topsoil (>90%), and 160 microg kg(-1) (>55%) in termite nests. In plants, the concentrations of PAHs in general decreased in the order leaves > bark > twigs > stem wood. The concentrations of most low-molecular weight PAHs in leaves and bark were near equilibrium with air, but those of NAPH were up to 50 times higher. Thus, the atmosphere seemed to be the major source of all PAHs in plants except for NAPH. Additionally, phenanthrene (PHEN) had elevated concentrations in bark and twigs of Vismia cayennensis trees (12-60 microg kg(-1)), which might have produced PHEN. In the mineral soil, perylene (PERY) was more abundant than in the litter layer, probably because of in situ biological production. Nasutitermes nests had the highest concentrations of most PAHs in exterior compartments (on average 8 and 15 microg kg(-1) compared to <3 microg kg(-1) in interior parts) and high PERY concentrations in all compartments (12-86 microg kg(-1)), indicating an in situ production of PERY in the nests. Our results demonstrate that the deposition of pyrolytic PAHs from the atmosphere controls the concentrations of most PAHs. However, the occurrence of NAPH, PHEN, and PERY in plants, termite nests, and soils at elevated concentrations supports the assumption of their biological origin. 相似文献
The presence of potentially hazardous elements (PHEs) in playground soils is generally associated with anthropogenic sources such as vehicle traffic, industries, construction sites, and biomass burning. Studies indicate that PHEs are harmful to human health and may even be carcinogenic. Therefore, the aim of this study was to evaluate the physicochemical, morphological, and mineralogical properties of soil samples from three public playgrounds located in the cities of Bogota, Medellin, and Barranquilla. Besides, the possible impacts caused by the aerodynamics of particles in Colombian cities were verified. The morphology, composition, and structure of the nanoparticles (NPs) (< 100 nm) present in these soils were evaluated by field emission scanning electron microscopy (FE-SEM) equipped with high-precision field emission (FE) and high-resolution transmission electron microscopy (HR-TEM). Soil samples were predominantly feldspar, quartz, and, to a lesser extent, clay minerals, carbonates, and hematites. The average content of PHEs was anthropogenically enriched in relation to the upper continental crust. As and Sn showed a large spatial variation, indicating the influence of local sources, such as vehicle traffic and industries. There is an inverse relationship between the total concentrations of some elements and their leachable fractions. The accumulation of traffic-derived PHEs has a negative impact on human health and the environment, which is alarming, especially for elements such as Pb, Sb, or As. Therefore, the presence of PHEs should receive greater attention from public health professionals, and limits should be set and exposures controlled. This study includes the construction of a baseline that provides basic information on pollution, its sources, and exposure routes for humans in the vicinity of Colombia’s major cities, characterized by their increasing urbanization and industrialization.
Mesotrione is a benzoylcyclohexane-1,3-dione herbicide that inhibits 4-hydroxyphenyl pyruvate dioxygenase in target plants. Although it has been used since 2000, only a limited number of degrading microorganisms have been reported. Mesotrione-degrading bacteria were selected among strains isolated from Brazilian aquatic environments, located near corn fields treated with this herbicide. Pantoea ananatis was found to rapidly and completely degrade mesotrione. Mesotrione did not serve as a sole C, N, or S source for growth of P. ananatis, and mesotrione catabolism required glucose supplementation to minimal media. LC-MS/MS analyses indicated that mesotrione degradation produced intermediates other than 2-amino-4-methylsulfonyl benzoic acid or 4-methylsulfonyl-2-nitrobenzoic acid, two metabolites previously identified in a mesotrione-degrading Bacillus strain. Since P. ananatis rapidly degraded mesotrione, this strain might be useful for bioremediation purposes. 相似文献
The Tubarão River rises in Santa Catarina, Brazil, and has been historically affected by coal mining activities around its springhead. To evaluate its water conditions, an investigation regarding a possible decontamination gradient associated with the increased river flow toward the estuary, as well as the influence of seasonality over this gradient was performed through a series of biomarkers (vitellogenin, comet assay, lipid peroxidation, protein carbonylation, gluthatione, gluthatione S-transferase, acetylcholinesterase, light microscopy in liver, and scanning electron microscopy in gills) and chemical analysis (polycyclic aromatic hydrocarbons (PAHs) in bile and metal analysis in sediment) in the cichlid Geophagus brasiliensis. Two collections (summer and winter) were made in four distinct sites along the river, while sediments were sampled between those seasons. As expected, the contamination linked exclusively to mining activities was not observed, possibly due to punctual inputs of contaminants. The decontamination gradient was not observed, although seasonality seemed to have a critical role in the responses of biomarkers and availability of contaminants. In the summer, the fish presented higher histopathological damages and lower concentrations of PAHs, while in the winter they showed both higher genetic damage and accumulation of PAHs. The effect of a long-term exposure of tenchs to different concentrations (10 and 100 micro g/L) of the pesticide carbofuran has been evaluated. Microsomal hepatic cytochrome P450 subfamily 1A (CYP1A) and 3A (CYP3A) activities, as well as the phase II enzyme uridine diphospho-glucuronosyltransferase (UDPGT) activity were evaluated as adequate biomarkers of fish exposure to environmentally relevant concentrations of the pesticide carbofuran in freshwater ecosystems. A clear time-dependent inhibition of both CYP1A and UDPGT activities was observed in fish exposed to the highest dose of carbofuran with respect to controls, whereas in the case of CYP3A activity, values of exposed animals did not show a clear pattern of alteration during the experiment. The results of the present study demonstrated that hepatic CYP1A and UDPGT activities from tench could be considered as sensitive biomarkers for carbamate pesticides in polluted water, thus allowing future and ecologically relevant biomonitoring studies with this species. 相似文献
In order to evaluate the extraction of pesticide residues that are transferred to the brew during mate drinking process of P.U.1 yerba mate leaves (Ilex paraguariensis), a special device to simulate the way in which mate is drunk in Uruguay was developed. The transfer to the brew of 12 organophosphates, 5 synthethic pyrethroids and one organochlorine pesticide from spiked samples was studied. The relationship between the transfer data thus obtained and physicochemical properties like water solubility (Ws), octanol-water coefficient (Kow) and Henry's constant (H) was evaluated. The extractability of the pesticide residues from yerba mate can be correlated with log Ws and log Kow. These transfer values allowed the calculation of ARLs (acceptable residue level) for the pesticides following Food and Agriculture Organization (FAO), World Health Organizaion (WHO) guidelines. These results can help the future establishment of maximum residue levels (MRLs). 相似文献
Little research has been performed on the impact of pesticides on earthworms under tropical conditions. Taking into consideration the often-limited resources in tropical countries, simple screening tests are needed. Therefore, it was investigated whether three pesticides relevant for the Brazilian Amazon (benomyl, carbendazim, lambda-cyhalothrin) affect the avoidance behavior of the earthworm Eisenia fetida. The tests were performed for two days according to ISO guideline 17512 but were adapted to tropical conditions (i.e. test substrate, test organism and temperature). The results indicate that this test gives reproducible and reliable results. Toxicity values (NOEC, EC50) are lower than those determined in 14 day-acute mortality tests and are approximately in the same range such as those found in 56 day-chronic reproduction tests with the same earthworm species, which were performed in parallel. Therefore, the use of the earthworm avoidance tests is recommended as a screening tool for the risk assessment of pesticides. 相似文献
