Aquatic ecosystems around the world, lake, estuaries and coastal areas are increasingly impacted by anthropogenic pollutants through different sources such as agricultural, industrial and urban discharges, atmospheric deposition and terrestrial drainage. Lake Victoria is the second largest lake in the world and the largest tropical lake. Bordered by Tanzania, Uganda, and Kenya, it provides a livelihood for millions of Africans in the region. However, the lake is under threat from eutrophication, a huge decline in the number of native fish species caused by several factors including loss of biodiversity, over fishing and pollution has been recently documented. Increasing usage of pesticides and insecticides in the adjacent agricultural areas as well as mercury contamination from processing of gold ore on the southern shores are currently considered among the most emergent phenomena of chemical contamination in the lake. By the application of globally consistent and comprehensive geospatial data-sets based on remote sensing integrated with information on heavy metals accumulation and insecticides exposure in native and alien fish populations, the present study aims at assessing the environmental risk associated to the contamination of the Lake Victoria water body on fish health, land cover distribution, biodiversity and the agricultural area surrounding the lake. By the elaboration of Landsat 7 TM data of November 2002 and Landsat 7 TM 1986 we have calculated the agriculture area which borders the Lake Victoria bay, which is an upland plain. This process has greatly enhanced nutrient loading to the soil, which is subsequently transported to the lake by rain or as dry fall. All the data has been insert in the Geographical information System (ARCGIS) to be upgraded and consulted. Heavy metals in fish fillets showed concentrations rather low except for mercury being higher than others as already described in previous investigations. In the same tissue, cholinesterases activity (ChE) as an indicator of insecticides exposure showed significant differences among fish species in both activity and sensitivity of selected inhibitor insecticides. This integrated approach aims at identifying and quantifying selected aquatic environmental issues which integrated with monitoring techniques such as contaminant concentrations and biological responses to insecticides exposure in fish populations will provide a scientific basis for aquatic zones management and assist in policy formulations at the national and international levels. 相似文献

Several ABC transporters have been characterized from many aquatic organisms, but no information is yet available for Antarctic fish. The aim of this work was to identify the expression of genes for ABC proteins in Trematomus bernacchii, a bioindicator species of the Southern Ocean. Partial cDNA sequences of ABCB1, ABCC1, ABCC2, ABCC4 and ABCC9 were cloned from liver. Using RACE technology, 3.5 and 2.2 kb contigs were obtained for ABCB1 and ABCC2. Considering the elevated natural bioavailability of cadmium at Terra Nova Bay, responsiveness of ABCB1 and ABCC2 to this element was investigated under laboratory conditions. ABCB1 and ABCC2 mRNA levels were approximately four-fold higher in Cd-exposed fish compared to the controls. Induction of ABCB1 protein was also found by western blot. This study provides the first identification of five ABC genes in the liver of an Antarctic key species, some of which may be involved in cellular detoxification. 相似文献

The aim of this study was to validate a multi-trial biomarker approach for the evaluation of toxicological risk due to benzo(alpha)pyrene. Carcinus aestuarii, exposed to increasing concentrations of B(alpha)P in the water, was used as the bioindicator organism. A set of biomarkers were tested in order to: identify biological materials for biomarker and residue analysis; determine a group of sensitive techniques for the assessment of PAH contamination; investigate correlation between responses at different levels of biological organisation. The results underlined that BPMO activities in hepatopancreas and gills were a good biomarker of exposure to PAH-type compounds. B esterases activities in hemolymph and porphyrin patterns in excreta could be proposed as a non-destructive approach for evaluating chemical exposure in this species. 相似文献

The induction of liver ethoxyresorufin O-deethylase (EROD) activity was investigated in the European eel, Anguilla anguilla, collected from a Mediterranean brackish environment and experimentally exposed to benzo[a]pyrene (B[a]P) and beta-naphthoflavone (BNF). Eels were injected intraperitoneally at increasing doses (0.1, 1, 10, and 50 mg/kg wet body weight) using corn oil as a carrier and sacrificed after 7 days. The main objectives of the present study are: (1). to assess of the sensitivity of EROD induction as a biomarker to polycyclic aromatic hydrocarbon (PAH) exposure; (2). to determine an EROD dose-response relationship of the contaminants used; and (3). to compare the efficiency of B[a]P and BNF as inducers of EROD activity. Results showed that both chemicals resulted in a dose-dependent EROD induction, but increases were not linear. EROD activity seemed to reach a plateau at the exposure of 10 mg/kg in both treatment groups; B[a]P was a more potent inducer than BNF was at the higher doses (10 and 50 mg/kg), while the opposite result was observed at the lower ones (0.1 and 1 mg/kg). The greatest induction occurred in eels treated with 10 mg/kg B[a]P, in which a 261-fold increase in EROD activity was observed. Results showed that EROD activity in A. anguilla is significantly induced by B[a]P and BNF exposure, responding to a wide range of concentrations of these contaminants. We infer that this tool may be suited as a diagnostic biomarker for biomonitoring PAHs pollution in Mediterranean brackish environments and further field research is suggested. 相似文献

In order to gain preliminary knowledge about the threat to marine ecosystems due to leakage of chemical warfare agents (CWAs)
and other pollutants from rusting bombshells on the seabed, a case study was conducted in a dumping area in the southern Adriatic
Sea (depth 200–300 m). Following electroacoustic and magnetometric surveys of the CWA dumping area, an integrated ecotoxicological
approach was used. This approach was based on analysis of CWA residues and their metabolites, including arsenic, in sediment
and organisms, as well as multimarker methodology including the Health Assessment Index, histological lesion analysis and
enzyme assays. Two sentinel species were selected, the blackbelly rosefish [Helicolenus dactylopterus (Delaroche, 1809)] and the European conger (Conger conger L., 1758). Sediment analysis revealed the presence of CWA degradation products, including 1-4-thioxane and 1-4-dithiane.
Tissues of fish from the CWA dumping site showed higher levels of arsenic than those from the reference site. Neither CWAs
nor their metabolites were detected in fish tissues. Arsenic levels recorded in blackbelly rosefish were well above those
reported for other fish species from the southern Adriatic, much higher than the FDA limit for food (2.6 mg kg−1) and close to the LD50 calculated for mammals (20 mg kg−1 body weight). The presence of pollutants in the CWA dumping site was also confirmed by pathological lesions in both species
and EROD activity, two to three times higher than in fish from the reference site (16.45±8.08 and 8.05±5.87 pmol min−1 mg protein−1 in blackbelly rosefish and 269±24.92 and 78.71 pmol min−1 mg protein−1 in European conger, respectively). Cholinesterase activity seemed unaffected in muscle of both species, whereas in brain
they were one-third of those recorded in fish from the reference site (14.22±10.05 and 72.87 nmol min−1 mg protein−1, respectively). This suggests that acetylcholinesterase is sensitive to CWAs. In conclusion, the agreement of all the chemical
and biological parameters investigated suggests that the integrated ecotoxicological approach used is appropriate to reveal
the presence and biological effects of CWAs in the marine ecosystem.

ABSTRACT Previous research has indicated that residential washing machines are potential sources of pollution due to the associated use of chemicals found in consumer products, for example, ethanol in laundry detergent and chlorine in bleach.1,2 Washing machines may also emit hazardous air pollutants found in contaminated drinking water. To better understand the extent and impact of chemical emissions from tap water, 26 experiments were completed using a residential washing machine and a cocktail of chemical tracers representing a wide range of physicochemical properties. Variable operating conditions for these experiments included water temperature, amount of clothes present in the machine, water volume, and level of washwater agitation. Chemical stripping efficiencies and mass transfer coefficients were determined during each cycle (fill, wash, and rinse) of a normal washing machine event. Headspace ventilation rates were determined using an isobutylene tracer gas. Mass transfer rates were significantly influenced by operating parameters as exhibited by a wide range of chemical stripping efficiencies. Stripping efficiencies ranged from 0.74 to 36% for acetone, 8.2 to 99% for toluene, 10 to 99% for ethylbenzene, and 6.9 to 100% for cyclohexane. 相似文献

Biomonitoring aquatic environmental quality in a marine protected area: a biomarker approach 总被引:1,自引:0,他引:1
The main aims of the present study, conducted in the framework of the MONIQUA-Egadi Scientific Project, were twofold: first, to make the first step in the development and validation of an ecotoxicological approach for the assessment of marine pollution in coastal environments on the basis of a set of biomarker responses in new sentinel species; and second, to obtain preliminary information on environmental quality in an Italian marine protected area, the Egadi Islands (Sicily). Several cytochrome P450-dependent mixed-function oxidase activities were measured in the following sentinel species: rainbow wrasse Coris julis, gastropod limpet Patella caerulea, and sea urchin Paracentrotus lividus. The results suggest that specimens from the Favignana Harbor may be exposed to P450 inducers, whereas most of the other sites seem to share similar environmental quality. The proposed approach has potential for assessment of environmental quality in marine protected areas. 相似文献

Eels are exposed to pollutants due to their unusual life cycle and are vulnerable to contamination associated with sediment
due to their diet, feeding habits and territoriality. Since the 1980s, a decline in American and European eel populations
has been recorded. The causes of this decline still are unknown but pollution from domestic and industrial effluents is known
to be involved. Since little data is available on PCB contamination in eels from Italian waters, PCBs were measured in muscle
of Anguilla anguilla captured in 2001 in two brackish ecosystems of the Mediterranean: the lagoons of Orbetello and Santa Giusta (Italy). PCBs
were higher in samples from Orbetello lagoon than in those detected from Santa Giusta lagoon (P < 0.05). Congener and isomer analyses indicated Aroclor 1260 as a likely source of PCBs in the Orbetello and Santa Giusta

Reproductive and oxidative stress biomarkers have been recommended as tools to assess the health of aquatic organisms. Though validated in the laboratory, there are few studies that tie a change in gene expression to adverse reproductive or population outcomes in the field. This paper looked at 17 streams with varying degrees of urbanization to assess the use of biomarkers associated with reproduction or stress in predicting reproductive success of fathead minnows. In addition, the relationship between biomarkers and water quality measures in streams with varying degrees of urbanization was examined. Liver vitellogenin mRNA was correlated with reproduction within a period of 11 d prior to sampling irrespective of habitat, but its correlation with egg output declined at 12 d and beyond indicating its usefulness as a short-term biomarker but its limits as a biomarker of total reproductive output. Stress biomarkers such as glutathione S-transferase may be better correlated with factors affecting reproduction over a longer term. There was a significant correlation between GST mRNA and a variety of anthropogenic pollutants. There was also an inverse correlation between glutathione S-transferase and the amount of the watershed designated as wetland. Egg production over the 21-d was negatively correlated with the amount of urbanization and positively correlated to wetland habitats. This study supports the development of multiple biomarkers linking oxidative stress and other non-reproductive endpoints to changes in aquatic habitats will be useful for predicting the health of fish populations and identifying the environmental factors that may need mitigation for sustainable population management. 相似文献
