Monitoring of Phenol in Wastewater Bioremediation by HPLC   总被引:1,自引:0,他引:1  
Bioremediation emphasizes the detoxification and destruction of toxic substances by microorganisms. Wastewater obtained from an industrial concern was solvent extracted with methyl alcohol and dichloromethane and analysed by GC/MS. Besides phenol, a large variety of organic compounds were detected. Under controlled laboratory conditions, the wastewater was innoculated with a mixed culture of microorganisms specially selected for their abilities to degrade phenol. Samples were collected at regular intervals from the stirred tank bioreactor and analysed for phenol by reverse phase HPLC with a C18 column. Results shows that from an initial phenol concentration of 987 ppm, slightly more than 50% was destroyed within 163 hours. The dry weight of the microorganisms and the plate count (CFU/ml) shows a steady increase from 0.5238 gms to 0.5355 gms and from 1.1E+9 to 1.94E+13 respectively over the same period. This suggested that the phenol was consumed by the microorganisms as the sole carbon source.  相似文献

Denitrification is a critical biogeochemical process that results in the conversion of nitrate to volatile products, and thus is a major route of nitrogen loss from terrestrial environments. Riparian buffers are an important management tool that is widely utilized to protect water from non-point source pollution. However, riparian buffers vary in their nitrate removal effectiveness, and thus there is a need for mechanistic studies to explore nitrate dynamics in buffer soils. The objectives of this study were to examine the influence of specific types of soluble organic matter on nitrate loss and nitrous oxide production rates, and to elucidate the relationships between these rates and the abundances of functional genes in a riparian buffer soil. Continuous-flow soil column experiments were performed to investigate the effect of three types of soluble organic matter (citric acid, alginic acid, and Suwannee River dissolved organic carbon) on rates of nitrate loss and nitrous oxide production. We found that nitrate loss rates increased as citric acid concentrations increased; however, rates of nitrate loss were weakly affected or not affected by the addition of the other types of organic matter. In all experiments, rates of nitrous oxide production mirrored nitrate loss rates. In addition, quantitative polymerase chain reaction (qPCR) was utilized to quantify the number of genes known to encode enzymes that catalyze nitrite reduction (i.e., nirS and nirK) in soil that was collected at the conclusion of column experiments. Nitrate loss and nitrous oxide production rates trended with copy numbers of both nir and 16s rDNA genes. The results suggest that low-molecular mass organic species are more effective at promoting nitrogen transformations than large biopolymers or humic substances, and also help to link genetic potential to chemical reactivity.  相似文献

Previous studies show that conservation actions have prevented extinctions, recovered populations, and reduced declining trends in global biodiversity. However, all studies to date have substantially underestimated the difference conservation action makes because they failed to account fully for what would have happened in the absence thereof. We undertook a scenario‐based thought experiment to better quantify the effect conservation actions have had on the extinction risk of the world's 235 recognized ungulate species. We did so by comparing species’ observed conservation status in 2008 with their estimated status under counterfactual scenarios in which conservation efforts ceased in 1996. We estimated that without conservation at least 148 species would have deteriorated by one International Union for Conservation of Nature (IUCN) Red List category, including 6 species that now would be listed as extinct or extinct in the wild. The overall decline in the conservation status of ungulates would have been nearly 8 times worse than observed. This trend would have been greater still if not for conservation on private lands. While some species have benefited from highly targeted interventions, such as reintroduction, most benefited collaterally from conservation such as habitat protection. We found that the difference conservation action makes to the conservation status of the world's ungulate species is likely to be higher than previously estimated. Increased, and sustained, investment could help achieve further improvements.  相似文献

Recent air pollution legislation, introduced as a Clean Air Act into New South Wales, the most populous and most heavily industrialized Australian State relies mainly upon the establishment of maximum limits of emission of air impurities. Regulations made under the Act set out the first of a series of such limits which cover the more commonly encountered pollutants. Other limits will be included later as knowledge is accumulated but, in the meantime, where standards have not been fixed the "best practicable means" of controlling air pollution must be used. To facilitate administration and to provide financial support for scientific and engineering services in the form of the Air Pollution Control Board, industries of significant air pollution potential have been designated as scheduled premises and pay annual license fees. Further technical support for the legislation is provided by an Air Pollution Advisory Committee.  相似文献

Warmer, more acidic water resulting from greenhouse gas emissions could influence ecosystem processes like bioerosion of calcifying organisms. Based on summer-maxima values (temperature = 26 °C; pH = 8.1) at a collection site in New York (40°56″ N, 72°30″ W), explants of the boring sponge Cliona celata Grant, 1826 were grown for 133 days on scallop shells in seawater ranging from current values to one scenario predicted for the year 2100 (T = 31 °C; pH = 7.8). High water temperature had little effect on sponge growth, survival, or boring rates. Lower pH slightly reduced sponge survival, while greatly influencing shell boring. At pH = 7.8, sponges bored twice the number of papillar holes and removed two times more shell weight than at pH = 8.1. Greater erosion resulted in weaker scallop shells. This study suggests that lower seawater pH may increase boring rates of C. celata in shellfish, with potentially severe implications for wild and farmed shellfish populations.  相似文献

The Dosage-Area Product is an areawide smog index obtained by combining contaminant concentration, duration, and areal extent: DAP = c×f×A (ppm × hr × mi2). The first step in developing a DAP is to plot daily contaminant dosages from an air monitoring network and to draw isopleths showing the dosage pattern. The second step is to determine the areas between isopleths and to multiply these areas by the applicable dosage levels. The final step is to sum the above partial products into a single number and to add a type designator indicating the geographic configuration of the dosage pattern. The DAP smog index is a realistic yardstick for measuring areawide smog experience. Reproducible results are obtained at low cost by using standard techniques. The standard DAP smog index is also useful for: analyzing air pollution episodes, estimating contaminant trends, and informing the public.  相似文献

The practical necessity for continuous air monitoring by an instrument network is explained. Network criteria are listed. A coulometric monitoring system for oxidantis described and evaluated. Operating methods, calibration techniques, costs, and comparative field data are given. The authors conclude the coulometric system is feasible for operation on a widespread basis. Attractive features include: portability; remote recording; easy installation; centralized, coincident calibration of several monitors; linear data output; data consistent with other systems; and a potential for mobile use in horizontal and vertical surveys of the atmosphere.  相似文献
