Sanfilippo D syndrome: Correction of glucosamine-6-sulphatase deficiency following fibroblast culture in chang's media

The de-O-sulphation of α-linked glucosamine-6-sulphate residues in heparan sulphate requires a specific sulphatase, glucosamine-6-sulphatase, which has been shown to be deficient in tissues of Sanfilippo D, or mucopolysaccharidosis type IIID (MPS IIID), patients. MPS IIID fibroblasts cultured in Basal Eagle's medium supplemented with either fetal calf serum or heat-inactivated fetal calf serum, MDCB or Ultraserg media had residual glucosarnine-6-sulphatase activities towards a heparin-derived trisaccharide substrate, O-(α-N-acetylglucosamine-6-sulphate)-(1→4)-L -O-(α-iduronic acid-2-sulphate)-(1→4)-D -O-2,5-anhydro [1-³H]mannitol-6-sulphate, GlcNAc6S-IdoA2S-anM6S, which were less than 1 per cent of the normal range for fibroblasts cultured in Basal Eagle's medium supplemented with fetal calf serum. However, the glucosamine-6-sulphatase activities of MPS IIID fibroblasts grown in Chang's medium were similar to the activities in normal control fibroblasts which were cultured in Basal Eagle's medium. These results indicate that caution is required for prenatal diagnosis of MPS IIID patients using chorionic villi or amniotic cells cultured in Chang's medium.

Methods for collecting time/activity pattern information related to exposure to combustion products

Focus groups, surveys and questionnaires, diaries and observations can be used to gather information about people's exposure to a wide range of combustion products. Information about locations and durations of exposure, and sources of exposure can be obtained with these instruments. The types of instruments used must be fine tuned to meet the design characteristics of the community in which the study will be conducted.

Treatment Technologies for Hazardous Wastes: Part 1 Treatment Alternatives for Dioxin Wastes

The 1984 Amendments to the Resource Conservation and Recovery Act (RCRA), called the Hazardous and Solid Waste Amendments, (HSWA) direct the EPA to determine if dioxin bearing wastes should be prohibited from landfills. This determination is to made by November 1986. Barring a determination by the Agency that the land disposal of dioxin wastes does not represent a threat to the environment, ot that appropriate alternative treatment technology is not available, a land disposal prohibition would go into effect at that time.¹ The purpose of this paper is to discuss the state of the art for several treatment processes for dioxin wastes that have been proposed to be used instead of land disposal for disposing of solid and liquid wastes containing dioxins.     

Environmental logistics engineering: A new approach to industrial ecology

Industrial Ecology is, thus far, the most plausible model for realizing the ideal of sustainability. It dispels the notion that the flow of resources can continue along a one-way street from nature to mankind. Today, it is fairly well-understood that economies and ecosystems are structured and operate in strikingly similar ways. Healthy economies and healthy ecosystems depend on elemental diversity and unencumbered interchanges. Yet, maintaining and sustaining both healthy economies and healthy ecosystems continue to be viewed as disparate goals. How can this unfortunate mind-set be changed? The authors show how logistics engineering could be the best approach to implementing a two-part solution to this environmental business conundrum.     

A Bayesian assessment of the effect of highway bypasses in Iowa on crashes and crash rate

Introduction

A common contention is that the construction of highway bypasses negatively impacts the economy of local communities by reducing pass-by traffic for businesses. However, as access to specific business' account records is limited, this impact is difficult to quantify. Another common contention is that bypasses contribute to a reduction in overall crashes in the community and in the surrounding areas. Even though a large number of bypasses have been constructed in the State of Iowa over the past several years, their actual impact in terms of traffic safety has not been quantified.

Objectives

This study seeks answers to the following questions: (a) Are bypasses in Iowa associated with a reduction in crash frequencies and crash rates on the bypassed highway? (b) Do bypasses in Iowa introduce a reduction of overall crash frequencies and rates or do they merely shift crashes from the highways through the communities to the bypasses with no significant overall reduction?

Method

We obtained crash information from the Iowa DOT at 19 sites on which a bypass was constructed sometime during the past 23 years. We also obtained the same information at six sites used as comparison sites on which no bypasses were constructed at least until 2005. We them employed a Bayesian approach to estimating the association between the construction of the bypass and crash rates, while also accounting for other factors.

Results

The construction of bypasses in Iowa is associated with a significant increase in traffic safety both on the main road through town and on the combined main road and bypass roadway.
