The effects of sewage sludge (SL) application on the soil and olive trees (Olea europaea L., cultivar: cornicabra) were studied. The plants were grown in 8.5L pots and subjected to the following treatments: 0, 3.66, 7.32, 14.65, 29.3, 58.6, and 117.2 g SL kg(-1) soil that corresponded, respectively, to 0, 4, 8, 16, 32, 64 and 128 M g ha(-1) dry weight of sewage sludge. The application of SL at the rates 64 and 128 M g ha(-1) produced leaf tip burning and leaf drop after 120 days, although cumulative metal pollutant loading rates was below USEPA and European regulations. This toxicity symptom could be caused by the high sodium levels in the leaves (over 0.19%), which can damage olive tree development. The Na contents of leaves were well correlated with soil Na content (r2: 0.91). In general, SL rates significantly increased the level of Cr, Ni, Cu, Zn, Cd and Pb in soil and plants, but these concentrations were in the normal ranges, except for the Zn concentration, which was over the critical soil content for the rates of 32, 64, 128 Mg ha(-1) but not in the leaves. Results suggested that regulations about the utilization of sewage sludge on agricultural land should consider the limit values for salt, and not only metals, that may be added to soil, in order to minimize the risk of negative effects to plant health. 相似文献

ABSTRACT The present work was designed to determine the potential genotoxicity at the vicinity of a solid waste incinerator in the metropolitan area of Sâo Paulo, using the Tradescantia stamen-hair bioassay. Experiments were carried out between December 1998 and April 1999 in four regions (40 pots of plants per site) selected on the basis of their pollution levels predicted by theoretical modeling of the dispersion of the incinerator's plume. The exposure sites were defined as follows: highest level (incinerator); a high level (museum) located 1.5 km from the emission point; a moderate level (school, at a distance of 3.5 km from the incinerator); and a control (at Jaguariuna countryside). The difference in genotoxicity among the groups was statistically significant (p < 0.001). The frequency of mutations observed in the countryside was significantly lower [2.25 ± 1.55, mean ± SD (standard deviation)] than that of the sites close to the incinerator. The frequency of mutations measured at the school (3.70 ± 1.36) was significantly lower than that measured at both the museum (4.89 ± 1.12) and the incinerator (5.69 ± 1.34). In conclusion, we found a positive correlation between the spatial distribution of the emissions of the incinerator located in an urban area and the mutagenic events measured by the Tradescantia stamen-hair assay. The in situ approach employed in this study was simple, efficient, and of low cost. No air or chemical extraction of pollutants was necessary for genotoxicity testing as required by other assays. 相似文献

Molecular techniques and modelling are presented as powerful tools required in the performance of efficient soil and water bioremediation systems. An Escherichia coli CC118-D strain was constructed by inserting the Klebsiella pneumoniae hpa B gene, coding for the unstable 3,4-dihydroxyphenylacetate 2,3-dioxygenase, into its chromosome. When the constructed strain was immobilized, both enzyme stability and viability increased along the studied period, in absence of antibiotic. We proposed this strategy as an approach to overcoming plasmid instability and to enhance enzyme activity and stability, avoiding antibiotic utilization. A model was developed to understand and predict the behaviour of bacteria and pollutants in a bioreactor system, considering: fluid dynamics, molecular/cellular scale processes and biofilm formation. 相似文献

The method of single-strand conformational polymorphism (SSCP) was modified in our laboratories for the characterization of baculoviruses, insect viruses with great potential for use as bioinsecticides in biological protection programs. A series of primers were synthesized after the comparison of the polyhedrin gene sequences of over 20 baculoviruses. Polyhedrin is a highly conserved protein which is responsible for the persistence of the virus in the environment. Universal primers were designed which could be used in polymerase chain reactions (PCR) containing genomic DNA from an array of nucleopolyhedrosis viruses (NPVs) including these which are used as biopesticides against important pests of forests and crops, such as Anticarsia gemmatalis, Spodoptera frugiperda, Lymantria dispar, Lymantria monacha and many others. PCR products were denatured and subjected to single-strand DNA electrophoresis at variable temperatures (MSSCP) where, after silver staining, they gave ssDNA band patterns characteristic for each baculovirus species. This technique can be potentially applied to detect baculoviruses in insects collected in the field, as well as to plant tissues and the excrements or bodies of predators without need for sequencing the PCR products. Sometimes MSSCP can be used not only for species determination but also as an indication of genomic variability which can be related to infectivity. 相似文献

We propose a new approach to evaluate the natural attenuation capacity of soil by using fluorescence in situ hybridization (FISH). A specific oligonucleotide probe AtzB1 was designed based on the sequence data of the atzB gene involved in the hydrolytic deamination of s-triazines; this gene, located in a multiple copy plasmid was detected by the optimized FISH protocol. Two agricultural soils (Lodi and Henares) with a history of simazine treatments, and two natural soils (Soto and Monza), without previous exposure to simazine, were studied. AtzB1 probe-target cells were found only in the agricultural soils and, in a greater percentage, in the Lodi soil, compared to the Henares one. Moreover, the greatest percentage of AtzB1 probe-target cells in Lodi was accompanied by a greater mineralization rate, compared to the Henares soil. The FISH method used in this study was suitable for the detection of simazine-degrading bacteria and could be a useful indicator of the potential of soil bioremediation. 相似文献
