Certain hydrothermal vent invertebrates, e.g. Riftia pachyptila and Calyptogena magnifica, are clearly established as harboring dense populations of chemoautotrophic sulfur bacteria in specialized tissues. By contrast, the physiological characteristics of the abundant intracellular gill symbiont of the vent mussel Bathymodiolus thermophilus have been questioned. The low activities of enzymes diagnostic for CO2 fixation (Calvin cycle) and for sulfur-driven energy generation, as measured by other investigators, have been attributed to bacterial contamination of the gill surface. Based on research at the Galápagos Rift hydrothermal vents in 1988 and subsequent laboratory experiments, the current study confirms that the B. thermophilus symbiont is a psychrophile for which thiosulfate and sulfide stimulate CO2 fixation. It strongly indicates that the symbiont is a chemoautotroph by establishing the following: (1) Sulfide and thiosulfate can stimulate CO2 fixation by partially purified symbionts by up to 43-fold and 120-fold, respectively; (2) the ribulose-1,5-bisphosphate carboxylase/oxygenase activity of the symbiont is sufficient to account for its sulfide- or thiosulfate-stimulated CO2 incorporation; (3) the symbiont's molar growth yield on thiosulfate, as judged by CO2 incorporation, is indistinguishable from that of free-living chemoautotrophs. Due to the high protein-degrading activity of B. thermophilus gill lysate, it is also suggested that host lysis of symbionts plays a more important role in the nutrition of the vent mussel than in R. pachyptila or C. magnifica, for which no comparable protein-degrading activity was found. 相似文献
Variability in the level of expression of very long chain fatty acids (VLCFAs) is documented in cultured chorionic villus (CV) cells derived from two fetuses, one at risk for an unusual peroxisomal fatty acid β-oxidation defect, and the other at risk for the X-linked form of adrenoleucodystrophy (ALD). Cells from early subcultures of chorionic cells from both cases gave normal values for VLCFA ratios. The results for the fetus at risk for the β-oxidation defect were interpreted to indicate that the fetus was not affected; however, at birth, the infant was clinically and biochemically affected. In the case of the fetus at risk for X-linked ALD, although VLCFAs were normal in subculture 1, the levels of these fatty acids increased dramatically in subculture 3, suggesting an abnormal fetus. Termination of the pregnancy and subsequent biochemical and morphological follow-up confirmed that the fetus was indeed affected by ALD. 相似文献
In-situ sparging has been accepted as a method to rapidly remediate groundwater at considerably lower costs compared to remedies based on groundwater recovery alone. The success of in-situ sparging depends on effective mass transfer between air and contaminated media in the subsurface. Factors affecting mass transfer include advective airflow, diffusive transport, interphase chemical partitioning, and chemical and biological reaction rates between sparged gases and subsurface contaminants, minerals, and naturally occurring organic compounds. Understanding these factors can increase the design efficiency of in-situ sparging and assist in developing sparging systems that use gases other than air (i.e., oxygen, ozone, and methane). 相似文献
Remotely sensed imagery is becoming a common source of environmental data. Consequently, there is an increasing need for tools to assess the accuracy and information content of such data. Particularly when the spatial resolution of imagery is fine, the accuracy of image processing is determined by comparisons with field data. However, the nature of error is more difficult to assess. In this paper we describe a set of tools intended for such an assessment when tree objects are extracted and field data are available for comparison. These techniques are demonstrated on individual tree locations extracted from an IKONOS image via local maximum filtering. The locations of the extracted trees are compared with field data to determine the number of found and missed trees. Aspatial and spatial (Voronoi) analysis methods are used to examine the nature of errors by searching for trends in characteristics of found and missed trees. Questions around how to conserve nature are increasingly leading to dissonance in conservation planning and action. While science can assist in unraveling the nature of conservation challenges, conservation responses rely heavily on normative positions and constructs to order actions, aid interpretations, and provide motivation. However, problems can arise when norms are mistaken for science or when they stymy scientific rigor. To highlight these potential pitfalls, we used the ethics-based tool of argument analysis to assess a controversial conservation intervention, the Pelorus Island Goat Control Program. The program proponents' argument for restorative justice was unsound because it relied on weak logical construction overly entrenched in normative assumptions. Overreliance on normative constructs, particularly the invocation of tragedy, creates a sense of urgency that can subvert scientific and ethical integrity, obscure values and assumptions, and increase the propensity for flawed logic. This example demonstrates how the same constructs that drive biodiversity conservation can also drive poor decision making, spur public backlash, and justify poor animal welfare outcomes. To provide clarity, a decision-making flowchart we devised demonstrates how values, norms, and ethics influence one another. We recommend practitioners follow 3 key points to improve decision making: be aware of values, as well as normative constructs and ethical theories that those values inform; be mindful of overreliance on either normative constructs or ethics when deciding action is justified; and be logically sound and transparent when building justifications. We also recommend 5 key attributes that practitioners should be attentive to when making conservation decisions: clarity, transparency, scientific integrity, adaptiveness, and compassion. Greater attention to the role of norms in decision making will improve conservation outcomes and garner greater public support for actions. 相似文献

In tropical areas, pioneer occupation fronts steer the rapid expansion of deforestation, contributing to carbon emissions. Up-to-date carbon emission estimates covering the long-term development of such frontiers depend on the availability of high spatial–temporal resolution data. In this paper, we provide a detailed assessment of carbon losses from deforestation and potential forest degradation from fragmentation for one expanding frontier in the Brazilian Amazon. We focused on one of the Amazonia’s hot-spots of forest loss, the BR-163 highway that connects the high productivity agricultural landscapes in Mato Grosso with the exporting harbors of the Amazon. We used multi-decadal (1984–2012) Landsat-based time series on forested and non-forested area in combination with a carbon book-keeping model. We show a 36% reduction in 1984s biomass carbon stocks, which led to the emission of 611.5 TgCO2 between 1985 and 1998 (43.6 TgCO2 year−1) and 959.8 TgCO2 over 1999–2012 (68.5 TgCO2 year−1). Overall, fragmentation-related carbon losses represented 1.88% of total emissions by 2012, with an increasing relevance since 2004. We compared the Brazilian Space Agency deforestation assessment (PRODES) with our data and found that small deforestation polygons not captured by PRODES had increasing importance on estimated deforestation carbon losses since 2000. The comparative analysis improved the understanding of data-source-related uncertainties on carbon estimates and indicated disagreement areas between datasets that could be subject of future research. Furthermore, spatially explicit, annual deforestation and emission estimates like the ones derived from this study are important for setting regional baselines for REDD+ or similar payment for ecosystem services frameworks.

The pH of Salmonella pre-enrichment media can become acidic (pH 4.0–5.0) when feeds/ingredients are incubated for 24?h. Salmonella in feed that have been stressed by heat and desiccation exhibit different pH tolerances than non-stressed cultures. Acidic conditions can result in cell injury/death and affect biochemical pathways. In this study, eight serotypes of Salmonella were grown in sterile meat and bone meal that was subjected to desiccation and heat stress. Cultures of non-stressed and stressed isolates were subsequently exposed to acidic pH from 4.0 to 7.0 in 0.5?pH increments (3 replicates/pH increment) in citrate buffer. At 6 and 24?h, serial dilutions were plated in duplicate on XLT-4 (xylose lysine tergitol-4) agar. Four serotypes showed an impaired ability to decarboxylate lysine on XLT-4. This inability to decarboxylate lysine was dependent on isolate, stress status, and incubation time. When the isolates’ ability to decarboxylate lysine was examined using biochemical tests, cultures were found to be able to decarboxylate lysine with the exception of S. Infantis. This suggests that XLT-4 contains a biochemical stressor(s) which affects the rate of decarboxylation by these Salmonella. These results suggest that acidic conditions may influence the detection and confirmation of Salmonella in feed. 相似文献
