Rapid detection of numerical aberrations of chromosomes 13, 18 and 21 in chorionic mesenchymal cells
We have devised and evaluated a rapid screening method for the detection of numerical aberrations of chromosomes13, 18 and 21 in chorionic villus cells. We used non-radioactive in situ hybridization (ISH) with three chromosome-specific probes on overnight-attached mesenchymal cells from chorionic villi. A blind study was performed of 47 karyotypically normal samples, one triploid sample, two samples trisomic for chromosome 21, and two samples from a fetus with putative mosaicism (46/47, +21). All samples were hybridized with the chromosome 18- and 21-specific probes. Thirty samples were additionally hybridized with the chromosome 13-specific probe. The test could be completed within 3-4 days of sampling. In samples disomic with respect to the probed chromosomes, an average of 2 per cent (range 0-9 per cent) had three hybridization signals. By contrast, in the samples trisomic for the probed chromosome(s), 57 per cent (chromosome 13), 51 per cent (chromosome 18), and an average of 74 per cent (55-86 per cent) (chromosome 21) of the nuclei exhibited three signals. In the putative mosaic samples, the number of nuclei with three chromosome 21-specific signals ranged from 41 to 69 per cent. We conclude that this technique rapidly and clearly distinguishes between normal and trisomic/triploid samples, and consequently may be of use in future prenatal diagnosis. 相似文献

Contaminated sites pose a significant threat to groundwater resources worldwide. Due to limited available resources a risk-based prioritisation of the remediation efforts is essential. Existing risk assessment tools are unsuitable for this purpose, because they consider each contaminated site separately and on a local scale, which makes it difficult to compare the impact from different sites. Hence a modelling tool for risk assessment of contaminated sites on the catchment scale has been developed. The CatchRisk screening tool evaluates the risk associated with each site in terms of its ability to contaminate abstracted groundwater in the catchment. The tool considers both the local scale and the catchment scale. At the local scale, a flexible, site specific leaching model that can be adjusted to the actual data availability is used to estimate the mass flux over time from identified sites. At the catchment scale, a transport model that utilises the source flux and a groundwater model covering the catchment is used to estimate the transient impact on the supply well. The CatchRisk model was tested on a groundwater catchment for a waterworks north of Copenhagen, Denmark. Even though data scarcity limited the application of the model, the sites that most likely caused the observed contamination at the waterworks were identified. The method was found to be valuable as a basis for prioritising point sources according to their impact on groundwater quality. Acid sulfate soils have been described as the “nastiest soils on earth” because of their strong acidity, increased mobility of potentially toxic elements and limited bioavailability of nutrients. They only cover a small area of the world's total problem soils, but often have significant adverse effects on agriculture, aquaculture and the environment on a local scale. Their location often coincides with high population density areas along the coasts of many developing countries. As a result, their negative impacts on ecosystems can have serious implications to those least equipped for coping with the low crop yields and reduced water quality that can result from acid sulfate soil disturbance. The Millennium Ecosystem Assessment called on by the United Nations in 2000 emphasised the importance of ecosystems for human health and well-being. These include the service they provide as sources of food and water, through the control of pollution and disease, as well as for the cultural services ecosystems provide. While the problems related to agriculture, aquaculture and the environment have been the focus of many acid sulfate soil management efforts, the connection to human health has largely been ignored. This paper presents the potential health issues of acid sulfate soils, in relation to the ecosystem services identified in the Millennium Ecosystem Assessment. It is recognised that significant implications on food security and livelihood can result, as well as on community cohesiveness and the spread of vector-borne disease. However, the connection between these outcomes and acid sulfate soils is often not obvious and it is therefore argued that the impact of such soils on human well-being needs to be recognised in order to raise awareness among the public and decision makers, to in turn facilitate proper management and avoid potential human ill-health. 相似文献

An Assessment of Stakeholder Perceptions and Management of Noxious Alien Plants in Spain 总被引:1,自引:1,他引:0
Despite biological invasions being a worldwide phenomenon causing significant ecological, economic, and human welfare impacts,
there is limited understanding regarding how environmental managers perceive the problem and subsequently manage alien species.
Spanish environmental managers were surveyed using questionnaires to (1) analyze the extent to which they perceive plant invasions
as a problem; (2) identify the status, occurrence, and impacts of noxious alien plant species; (3) assess current effort and
expenditure targeting alien plant management; and, finally, (4) identify the criteria they use to set priorities for management.
In comparison to other environmental concerns, plant invasions are perceived as only moderately problematic and mechanical
control is the most valued and frequently used strategy to cope with plant invasions in Spain. Based on 70 questionnaires
received, 193 species are considered noxious, 109 of which have been the subject of management activities. More than 90% of
species are found in at least one protected area. According to respondents, the most frequently managed species are the most
widespread across administrative regions and the ones perceived as causing the highest impacts. The perception of impact seems
to be independent of their invasion status, since only half of the species identified as noxious are believed to be invasive
in Spain, while 43% of species thought to only be casual aliens are causing a high impact. Records of management costs are
poor and the few data indicate that the total actual expenditure amounted to 50,492,437 € in the last decade. The majority
of respondents stated that management measures are insufficient to control alien plants due to limited economic resources,
lack of public awareness and support, and an absence of coordination among different public administrations. Managers also
expressed their concern about the fact that much scientific research is concerned with the ecology of alien plants rather

We evaluated the in vitro activity of citrus oils against Mycobacterium tuberculosis and other non-tuberculous Mycobacterium species. Citrus essential oils were tested against a variety of Mycobacterium species and strains using the BACTEC radiometric growth system. Cold pressed terpeneless Valencia oil (CPT) was further tested using the Wayne model of in vitro latency. Exposure of M. tuberculosis and M. bovis BCG to 0.025 % cold pressed terpeneless Valencia orange oil (CPT) resulted in a 3-log decrease in viable counts versus corresponding controls. Inhibition of various clinical isolates of the M. avium complex and M. abscessus ranged from 2.5 to 5.2-logs. Some species/strains were completely inhibited in the presence of CPT including one isolate each of the following: the M. avium complex, M. chelonae and M. avium subsp. paratuberculosis. CPT also inhibited the growth of BCG more than 99 % in an in vitro model of latency which mimics anaerobic dormancy thought to occur in vivo. The activity of CPT against drug-resistant strains of the M. avium complex and M. abscessus suggest that the mechanism of action for CPT is different than that of currently available drugs. Inhibition of latently adapted bacilli offers promise for treatment of latent infections of MTB. These results suggest that the antimycobacterial properties of CPT warrant further study to elucidate the specific mechanism of action and clarify the spectrum of activity. 相似文献

Abstract This study comprehensively characterizes hourly fine particulate matter (PM2.5) concentrations measured via a tapered element oscillating microbalance (TEOM), β-gauge, and nephelometer from four different monitoring sites in U.S. Environment Protection Agency (EPA) Region 5 (in U.S. states Illinois, Michigan, and Wisconsin) and compares them to the Federal Reference Method (FRM). Hourly characterization uses time series and autocorrelation. Hourly data are compared with FRM by averaging across 24-hr sampling periods and modeling against respective daily FRM concentrations. Modeling uses traditional two-variable linear least-squares regression as well as innovative nonlinear regression involving additional meteorological variables such as temperature and humidity. The TEOM shows a relationship with season and temperature, linear correlation as low as 0.7924 and nonlinear model correlation as high as 0.9370 when modeled with temperature. The β-gauge shows no relationship with season or meteorological variables. It exhibits a linear correlation as low as 0.8505 with the FRM and a nonlinear model correlation as high as 0.9339 when modeled with humidity. The nephelometer shows no relationship with season or temperature but a strong relationship with humidity is observed. A linear correlation as low as 0.3050 and a nonlinear model correlation as high as 0.9508 is observed when modeled with humidity. Nonlinear models have higher correlation than linear models applied to the same dataset. This correlation difference is not always substantial, which may introduce a tradeoff between simplicity of model and degree of statistical association. This project shows that continuous monitor technology produces valid PM2.5 characterization, with at least partial accounting for variations in concentration from gravimetric reference monitors once appropriate nonlinear adjustments are applied. Although only one regression technically meets new EPA National Ambient Air Quality Standards (NAAQS) Federal Equivalent Method (FEM) correlation coefficient criteria, several others are extremely close, showing optimistic potential for use of this nonlinear adjustment model in garnering EPA NAAQS FEM approval for continuous PM2.5 sampling methods. 相似文献
