Present study describes the treatment of molasses spentwash and its use as a potential low cost substrate for production of biopolymer polyhydroxybutyrate (PHB) by waste activated sludge. Fluorescence microscopy revealed the presence of PHB granules in sludge biomass which was further confirmed by fourier transform-infra-red spectroscopy (FT-IR) and 13C nuclear magnetic resonance (NMR). The processing of molasses spentwash was carried out for attaining different ratios of carbon and nitrogen (C:N). Highest chemical oxygen demand (COD) removal and PHB accumulation of 60% and 31% respectively was achieved with raw molasses spentwash containing inorganic nitrogen (C:N ratio = 28) followed by COD removal of 52% and PHB accumulation of 28% for filtered molasses containing inorganic nitrogen (C:N ratio = 29). PHB production yield (Yp/s) was highest (0.184 g g?1 COD consumed) for deproteinized spentwash supplemented with nitrogen. In contrast, the substrate consumption and product formation were higher in case of raw spentwash. Though COD removal was lowest from deproteinized spentwash, evaluation of kinetic parameters suggested higher rates of conversion of available carbon to biomass and PHB. Thus the process provided dual benefit of conversion of two wastes viz. waste activated sludge and molasses spentwash into value-added product-PHB.  相似文献

The present study describes the production and characterization of a feather hydrolyzing enzyme by Serratia sp. HPC 1383 isolated from tannery sludge, which was identified by the ability to form clear zones around colonies on milk agar plates. The proteolytic activity was expressed in terms of the micromoles of tyrosine released from substrate casein per ml per min (U/mL min). Induction of the inoculum with protein was essential to stimulate higher activity of the enzyme, with 0.03% feathermeal in the inoculum resulting in increased enzyme activity (45 U/mL) that further increased to 90 U/mL when 3 d old inoculum was used. The highest enzyme activity, 130 U/mL, was observed in the presence of 0.2% yeast extract. The optimum assay temperature and pH for the enzyme were found to be 60 °C and 10.0, respectively. The enzyme had a half-life of 10 min at 60 °C, which improved slightly to 18 min in presence of 1 mM Ca2+. Inhibition of the enzyme by phenylmethyl sulfonyl fluoride (PMSF) indicated that the enzyme was a serine protease. The enzyme was also partially inhibited (39%) by the reducing agent β-mercaptoethanol and by divalent metal ions such as Zn2+ (41% inhibition). However, Ca2+ and Fe2+ resulted in increases in enzyme activity of 15% and 26%, respectively. The kinetic constants of the keratinase were found to be 3.84 μM (Km) and 108.7 μM/mL min (Vmax). These results suggest that this extracellular keratinase may be a useful alternative and eco-friendly route for handling the abundant amount of waste feathers or for applications in other industrial processes.  相似文献

Fly ash is a solid waste generated in huge quantities from coal fired thermal power stations during the combustion of coal. In India, less than half of this is used as a raw material for concrete manufacturing and construction; the remaining is directly dumped on land side as land fill or simply piled up. Only a small fraction of it is used in development of high valued product. Due to environmental regulations, new ways of utilizing fly ash are being explored in order to safeguard the environment and provide useful ways for its utilization and disposal. With its richness in various metal oxides, it has tremendous potential to be utilized as a filler material in polymer composites. These days glass reinforced polyester composites find widespread application in erosive environment due to several advantages like high wear resistance, strength-to-weight ratio, and low cost. The cost of the composites can be further brought down using cheaper filler materials. To this end, this work uses fly ash in composite making and thereby suggests a new way of better utility of this industrial waste. It includes the processing, characterization and study of the erosion behavior of a class of such fly ash filled polyester-glass fiber composites. The engineering application of composites demands that it should have high wear resistance, low density and high tensile strength. In order to assess the behavior of composites satisfying multiple performance measures, a grey-based Taguchi approach has been adopted. After thorough analysis of factors, optimal factor settings have been suggested to improve multiple responses viz., erosive wear rate, density, flexural strength and tensile strength. This technique eliminates the need for repeated experiments; thus saves time and material. The systematic experimentation leads to determination of significant process parameters and material variables that predominantly influence the multiple responses.  相似文献

This paper presents the results of wireless data acquisition experiments from embedded micro thin film sensors in cutting inserts for machining. A bluetooth module is used to acquire data and establish communication with a receiver PC over the serial port profile (SPP). A signal conditioning circuit is designed and developed to increase the signal-to-noise ratio of the embedded micro thin film sensors. Moreover, an averaging filter algorithm is implemented as a software interface. To characterize the wireless data acquisition (DAQ) system, laser heating and turning tests are conducted. Both tests show that the wireless DAQ system is able to provide desirable capabilities as well as the wired one for the embedded thin film sensors in cutting inserts.  相似文献

This study reports on the biocultural dimensions of the tasat (Arenga obtusifolia Griff.) tree and its conservation by various informal institutions of the Adi tribe of Arunachal Pradesh, India. The study included 240 Adi community members (120 men and 120 women) residing in 12 villages of the East Siang and Upper Siang districts of Arunachal Pradesh. The study was conducted using personal interviews and participatory methods. Results indicated that the stem of tasat is used as a traditional food during droughts and in preparing an ethnic beverage. Tasat is being conserved by the Adi tribe in various habitats. Plant populations of tasat were observed relatively higher in Upper Siang than the East Siang district. The study found differences in the status of biocultural knowledge between genders and different age groups. Kebang and reglep are two indigenous institutions of men and women, respectively, and play leading roles in sustaining tasat plants in various habitats. Women of the Upper Siang district still maintain a knowledge network for diffusing biocultural knowledge related to tasat, among others, through a barter system. Changing land use patterns in agriculture affects tasat conservation. The value addition for tasat based products, and rewarding women who conserve tasat, can enhance the sustainability of tasat based on biocultural knowledge and institutions.  相似文献

In this study, we analysed the indigenous practices of agro-biodiversity, changing patterns of cropping systems and conservation of agriculturally important natural resources of rice–wheat-based agroecosystems. The research was carried out in three villages of Azamgarh district, eastern Uttar Pradesh, India. Participatory approaches and personal interviews were combined to collect the data from 180 farmers. The results indicated that resource-poor (RP) farmers, by applying traditional practices of natural resource conservation, outperform their rich (RR) counterparts in conserving agro-biodiversity. Increase in irrigation facilities, adoption of improved crop varieties, commercialisation of farming and socio-economic changes have threatened the agro-biodiversity and related natural resources of these agroecosystems. For most of the crops, the RP farmers had better production stability as compared to the RR ones because the former tend to apply more sustainable and ecologically sound indigenous practices. A negative growth rate was observed in the area/numbers and production of legumes, livestock, pasture lands and community forestry. Women, who are the local custodians of conserving indigenous biodiversity (uncultivated species) and crop varieties through localised knowledge and sociocultural institutions, had significantly higher knowledge on agro-biodiversity than men. We concluded that integration of farmers’ eco-friendly practices with formal science and policy-making in a participatory mode may enhance sustainability of functions of agroecosystem leading to the strengthening of adaptive practices in changing climate and socio-economic scenario.  相似文献
