This note describes a technique for cutting undisturbed brittleblock samples into smaller specimens for further geotechnical testing. This technique revealed very useful in dealing with collapsible soils, where the sampling is recommended to be donewith block soil samples. When females mate with more than one male, the ensuing sperm competition leads to the evolution of male mechanisms that skew
paternity. Males of the yellow mealworm beetle (Tenebrio molitor) transfer a spermatophore to females during copulation, but sperm release and storage occur later. We investigated how the
interval between two matings with different males affects sperm precedence by varying the interval between the copulations
so that the second mating was either: (1) before sperm release from the first spermatophore (<5 min); (2) after sperm release
but before spermatophore ejection (15–20 min); (3) after spermatophore ejection but before sperm storage (4 h), or (4) after
complete sperm storage (24 h). We collected offspring over a period of 2 weeks and determined paternity by protein electrophoresis.
There was second-male sperm precedence in all treatments, but when the interval was <5 min, the second male usually (86% of
cases) had complete sperm precedence (i.e., P
2=1). Investigations into the mechanism of second-male sperm precedence during <5-min mating intervals indicate that sperm
release from the first spermatophore is inhibited, a phenomenon which has not been previously documented.

Iodoacetic acid (IAA) is an unregulated water disinfection byproduct that is an ovarian toxicant. However, the mechanisms of action underlying IAA toxicity in ovarian follicles remain unclear. Thus, we determined whether IAA alters gene expression in ovarian follicles in mice. Adult female mice were dosed with water or IAA (10 or 500 mg/L) in the water for 35-40 days. Antral follicles were collected for RNA-sequencing analysis and sera were collected to measure estradiol. RNA-sequencing analysis identified 1063 differentially expressed genes (DEGs) in the 10 and 500 mg/L IAA groups (false discovery rate FDR < 0.1), respectively, compared to controls. Gene Ontology Enrichment analysis showed that DEGs were involved with RNA processing and regulation of angiogenesis (10 mg/L) and the cell cycle and cell division (500 mg/L). Pathway Enrichment analysis showed that DEGs were involved in the phosphatidylinositol 3-kinase and protein kinase B (PI3K-Akt), gonadotropin-releasing hormone (GnRH), estrogen, and insulin signaling pathways (10 mg/L). Pathway Enrichment analysis showed that DEGs were involved in the oocyte meiosis, GnRH, and oxytocin signaling pathways (500 mg/L). RNA-sequencing analysis identified 809 DEGs when comparing the 500 and 10 mg/L IAA groups (FDR < 0.1). DEGs were related to ribosome, translation, mRNA processing, oxidative phosphorylation, chromosome, cell cycle, cell division, protein folding, and the oxytocin signaling pathway. Moreover, IAA exposure significantly decreased estradiol levels (500 mg/L) compared to control. This study identified key candidate genes and pathways involved in IAA toxicity and can help to further understand the molecular mechanisms of IAA toxicity in ovarian follicles. 相似文献

To evaluate whether poly-beta-hydroxyalkanoate (PHA) production and wastewater treatment could be combined in a single biological process, a bench-scale sequencing batch reactor was operated with sequential anaerobic and aerobic stages and removal of excess sludge at different stages of treatment. The reactor treated synthetic wastewater with a high organic and low nutrient content, simulating industrial wastewater. Chemical oxygen demand removal efficiency was more than 90% in all cases. Poly-beta-hydroxyalkanoate accumulation was significant, although it did not appear to be induced by oxygen limitations during the anaerobic stage. Sphaerotilus natans was apparently the dominant PHA-accumulating organism at the end of each reactor run and corresponded to a PHA accumulation of 16 to 20% of the total dry cell mass. Before S. natans dominated the reactors, PHA accumulation was approximately 17% when biomass was removed at the end of the aerobic stage and 6.6% when sludge removal also occurred during the anaerobic stage. 相似文献
