Phosphor imager autoradiography is a technique for rapid, sensitive analysis of the localization of xenobiotics in plant tissues. Use of this technique is relatively new to research in the field of plant science, and the potential for enhancing visualization and understanding of plant uptake and transport of xenobiotics remains largely untapped. Phosphor imager autoradiography is used to investigate the uptake and translocation of the explosives 1,3,5-trinitro-1,3,5-triazine (RDX) and 2,4,6-trinitrotoluene within Populus deltoides × nigra DN34 (poplar) and Panicum vigratum Alamo (switchgrass). In both plant types, TNT and/or TNT-metabolites remain predominantly in root tissues while RDX and/or RDX-metabolites are readily translocated to leaf tissues. Phosphor imager autoradiography is further investigated for use in semi-quantitative analysis of uptake of TNT by switchgrass. 相似文献

This study was carried out to explore the lowest achievable dinitrotoluene (DNT) isomer concentrations that would support sustained growth of DNT degrading microorganisms under an aerobic condition. Studies were conducted using suspended (chemostat) and attached growth (column) systems. The biodegradation limits for 2,4-dinitrotoluene chemostat and column system were 0.054 ± 0.005 and 0.057 ± 0.008 μM, respectively, and for 2,6-dinitrotoluene, the limits for chemostat and column system were 0.039 ± 0.005 and 0.026 ± 0.013 μM, respectively. The biodegradation limits determined in this study are much lower than the regulatory requirements, inferring that bacterial ability to metabolize DNT does not preclude applications of bioremediation (including natural attenuation) for DNT contaminated media. 相似文献

Aluminium toxicity is a major deterrent for plant growth in acid soils below pH 5.0. This study deals with effect of aluminium toxicity on growth of mungbean (Vigna radiata L. Wilczek) seedlings. Seed germination (in %) declined with increased content of Al2(SO4)3, while promotive effect was observed at very low dosage. Different concentrations of aluminum sulphate salt were applied to mungbean seeds. Measurement of aluminium content in mungbean leaves was done through atomic absorption spectrophotometer. Root length (root and hypocotyl length) and shoot length (shoot and epicotyl length) was measured at seven days old seedling stage. Different concentrations of Al2(SO4)3 were found to have significant effect both on shoot and root length. Leaf area, fresh and dry weight was significantly reduced. Increased stomatal frequency and trichome density with an increase in concentrations of Al2(S04)3 was observed through scanning electron microscope. 相似文献

Season-dependent mineral accumulation was recorded in fruits of Okra and Tomato. The highest concentration was in summer in Okra and in winter in Tomato. Lowest concentrations were in winter in Okra and rainy in Tomato. Both crop plants indicated that the mineral contents were in the order of P > K > Ca > Mg > Na > Fe which also signify their relative functional importance in growth and metabolism. 相似文献

Degradation of flubendiamide as affected by microbial population count in two Indian soils (red and alluvial) varying in physicochemical properties was studied under sterile and non-sterile conditions. Recovery of flubendiamide in soil was in the range of 94.7–95.9 % at 0.5 and 1.0 μg g?1, respectively. The DT50 of flubendiamide at the level of 10 μg g?1 in red soil under sterile and non-sterile conditions was found to be 140.3 and 93.7 days, respectively, and in alluvial soil under sterile and non-sterile condition was 181.1 and 158.4 days, respectively. Residues of flubendiamide dissipated faster in red soil (non-sterile followed by sterile) as compared to alluvial (non-sterile soil followed by sterile soil). A wide difference in half-life of red and alluvial soil under sterile and non-sterile conditions indicated that the variation in physicochemical properties of red and alluvial soil as well as the presence of microbes play a great role for degradation of flubendiamide. The results revealed that slower-degrading alluvial soil possessed microbes with degradative capacity. The degradation rate in this soil was significantly reduced by some of its physicochemical characteristics, despite sterile and non-sterile conditions, which was faster in red soil. 相似文献

Sodium arsenate (Na2HAsO4.7H2O) is a potent inhibitor of mungbean seed germination and seedling growth. Germination is totally stopped at or above 50 microM Na2HAsO4.7H2O. Inhibition of seedling elongation started at a lower concentration of 5 microM As(V) and was drastically reduced at 20 microM As(V). Nutrients like salts of macroelements viz., NaH2PO4.2H2O, KH2PO4, K2SO4, MgSO4.7H2O, CaCl2.2H2O, (NH4)2SO4 NH4NO3 solutions at a concentration of 10mM and microelements viz., ZnSO4, CuSO4.5H2O, Na2MoO4.2H2O, MnCl2.4H2O, CoCl2.6H2O, FeSO4.7H2O solutions at a concentration of 1mM could help to ameliorate the toxic effects of As(V) to different degrees. Amelioration of As(V) toxicity was possible only when the mungbean seeds were pretreated with the above mentioned nutrients for 24 hr and then transferred to sodium arsenate. Simultaneous treatment of nutrients with As(V) or using nutrient solutions following As(V) treatment were of no help to reverse the toxic effects of sodium arsenate. 相似文献

Studies were conducted to analyze the residue of commonly used pesticides viz. methyl parathion, chloropyrifos, endosulfan, cypermethrin, fenvalerate, carbendazim, imidacloprid and carbaryl in mango, Dashehari variety, integrated pest management (IPM) and non-IPM samples were collected from the IPM and non-IPM orchards, Lucknow, India. We also present a method for the simultaneous determination of these pesticides in mango samples. Residues of methyl parathion, chloropyriphos, endosulfan, cypermethrin, fenvalerate were extracted from the samples with acetone: cyclohexane: ethyl acetate in the ratio 2:1:1 followed by cleanup using neutral alumina. Analysis was performed by gas chromatography-electron capture detector (GC-ECD) with a megabore column (OV-1). Residues of carbendazim, imidacloprid and carbaryl were extracted with acetone and after cleanup, analysis was performed by high performance liquid chromatography (HPLC) using photo diode array (PDA) detector. Recoveries of all the pesticides ranged between 72.7 – 110.6%, at 0.1 and 1.0 μg g? 1 level of fortification. The residues detected in non-IPM samples of mango were found to be below the prescribed limits of maximum residue limit (MRL) while IPM samples were free from pesticide residues. 相似文献

Phosphorus (P) is the second most important macronutrient for plant growth. Plants exhibit numerous physiological and metabolic adaptations in response to seasonal variations in phosphorus content. Activities of acid and alkaline phosphatases, ATPase and ATP content were studied in summer, rainy and winter seasons at two different developmental stages (28 and 58 days after sowing) in Okra. Activities of both acid and alkaline phosphatases increased manifold in winter to cope up with low phosphorus content. ATP content and ATPase activity were high in summer signifying an active metabolic period. Phosphorus deficiency is characterized by low ATP content and ATPase activity (which are in turn partly responsible for a drastic reduction in growth and yield) and enhanced activities of acid and alkaline phosphatases which increase the availability of P in P-deficient seasons. 相似文献
