Three monoclonal antibodies (MAbs) against trophoblast (GB17, GB21, and GB25) and flow cytometry were used to sort trophoblast-like cells (TLCs) from peripheral blood of pregnant women. Sorted TLCs were processed for electron microscopy and fetal DNA amplification of the Y-specific sequences from mothers carrying male fetuses. At the ultra-structural level, most of the nucleated cells had the morphology of leucocytes, suggesting maternal contaminants, and we did not find the characteristic features of the free inter-villous trophoblast cells. Nevertheless, polymerase chain reaction (PCR) analysis showed an amplification of Y-specific sequences in two out of three samples of sorted TLCs. These results suggest that besides the maternal leucocytes, sufficient trophoblast nucleated fetal cells can be obtained using cell enrichment by sorting. This sensitive method holds promise for non-invasive prenatal diagnosis of fetal sex and if sufficient Y(positive) nuclei are found, for the diagnosis of selected numerical chromosome abnormalities.  相似文献

This study aims to evaluate the influence of start-up on polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans (PCDD/Fs) concentration in the stack flue gas of incinerators and its contributing PCDD/F emission. The PCDD/F emission of the first sample among three consecutive stack flue gas samples of five intermittent incinerators, which sampled at a stable combustion condition after start-up, is 2-3 times higher than the mean of the others. For verifying the PCDD/F characteristics of incinerators during start-up, one continuous MSWI was investigated for two years. The elevated PCDD/F emissions of the MSWI during start-up could reach 96.9 ng I-TEQN m(-3) and still maintained a high PCDD/F emission (40 times higher than the Taiwan emission limit) even 18 h after the injection of activated carbon, indicating the memory effect. Taking the MSWI for example, which consists of four incinerators, the estimated annual PCDD/F emission from normal operational conditions was 0.112 g I-TEQ. However, one start-up procedure can generate approximately 60% of the PCDD/F emissions for one whole year of normal operations. And the PCDD/F emission, which is the result of the start-ups of four incinerators, was at least two times larger than that of a whole year's normal operations, without consideration for the PCDD/F emission contributed by the long lasting memory effect.  相似文献

Activated carbons with diverse physical and chemical properties were produced from four agriculture residues, including raw barley husk, biotreated barley husk, rice husk, and pistachio shell. Results showed that with adequate steam activation (30-90 min, 50% H2O(g),/50% N2), activated carbons with surface areas between 360 and 950 m2 g(-1) were developed. Further increases in the activation time destroyed the pore structure of activated carbons, which resulted in a decrease in the surface area and pore volume. Biotreated agricultural residues were found to be suitable precursors for producing mesoporous activated carbons. The oxygen content of activated carbons increased with increasing activation time. Results from X-ray photoelectron spectroscopy examination further suggested that H2O molecules react with the carbon surface, enhancing the deconvoluted peak area of carbonyl and carboxyl groups. Equilibrium adsorption of toluene indicated that the adsorption capacities increased with an increase in the inlet toluene concentration and a decrease in temperature. The adsorption isotherms were successfully fitted with Freundlich, Langmuir, and Dubinin-Radushkevich equations. Activated carbons derived from agricultural residues appear to be more applicable to adsorb volatile organic compounds at a low concentration and high-temperature environment.  相似文献

Quasi-dynamic leaching characteristics of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) from raw and solidified air pollution control (APC) residues were examined via a nine-time multiple leaching test. The effect of injected activated carbon in the APC residues on the PCDD/F leachability was also evaluated. When humic acid solution was used as a leachant, the leaching concentrations of PCDD/Fs fluctuated between the first and the fifth leaching, followed by a gradual increase and then suddenly reached maximum values at the leaching sequences around seventh and eighth. This significant enhancement in PCDD/F leachability was mainly due to an increase in the release of highly chlorinated PCDD/Fs. Leaching of PCDD/Fs with n-hexane was, in contrast, primarily caused by the partitioning of hydrophobic PCDD/Fs between the APC residue surface and the liquid phase of n-hexane. Consequently, the largest leaching concentrations for n-hexane tests achieved at the first leaching, followed by a decrease and reached plateaus. Solidification/stabilization (S/S) decreased the PCDD/F leachability up to the fifth leaching by the use of humic acid solution. However, S/S increased the PCDD/F leaching concentrations and rates with n-hexane. The activated carbon in APC residues significantly inhibited the release of PCDD/F with n-hexane. The inhibiting effect provided by activated carbon was, however, less significant by the use of humic acid solution.  相似文献

ABSTRACT: To comprehend the distributions of salinity, temperature, and suspended sediment in the Danshuei River estuary in Taiwan, monthly field surveys were conducted in 2003. These included several high and low slackwater surveys and intensive surveys. The results show that the Danshuei River estuary is predominately a partially mixed estuary. The highest concentration of suspended sediment is typically observed at the Chung‐Hsin Bridge, the most upstream sampling station. The suspended sediment concentration exhibits a general decreasing trend in the downstream direction. It may be concluded that the sediments mostly come from the upstream reach. A locally high concentration of suspended sediment is found at the Kuan‐Du station because of the local deep channel bathymetry and two‐layered estuarine circulation. A vertical two‐dimensional hydrodynamic and sediment transport model is applied to investigate the tidally averaged salinity distribution, residual circulation, and suspended sediment concentration. The modeling results reveal that, under the Q75 flow condition (i.e., low flow), a turbidity maximum occurs at the Kuan‐Du station due to the strong estuarine circulation. The model simulation with a much higher river flow condition results in a weaker residual circulation and weaker turbidity maximum.  相似文献

The accumulation of As in rice due to groundwater irrigation in paddy fields represents a serious health hazard in South and Southeast Asia. In Taiwan, the fate of As in long-term irrigated paddy fields is poorly understood. Groundwater, surface soil, and rice samples were collected from a paddy field that was irrigated with As-containing groundwater in southwestern Taiwan. The purpose of this study is to elucidate the source and sink of As in the paddy field by comparing the As fractions in the soils that were obtained by a sequential extraction procedure (SEP) with the As uptake of rice. The risks associated with eating rice from the field can thus be better understood. The concentration of As in groundwater varied with time throughout the growing seasons of rice, but always exceeded the permitted maximum (10 μg L−1) for drinking water by the WHO. The As concentration increased with the concentration of Fe in the groundwater, supporting the claim that a large amount of As was concentrated in the Fe flocs collected from the internal wall of the groundwater pump. The results of the SEP revealed that As bound with amorphous and crystalline hydrous oxides exhibited high availability in the soils. The root of rice accumulated the largest amount of As, followed by the straw, husk, and grain. Although the As concentration in the rice grain was less than 1.0 mg kg−1, the estimated intake level was close to the maximum tolerable daily intake of As, as specified by the WHO.  相似文献

Leachability of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) from raw and solidified air pollution control (APC) residues with selected solvents, including acetic acid, simulated acid rain, humic acid, linear alkylbenzene sulfonate (LAS) and n-hexane was investigated. High-chlorinated PCDD/F congeners were observed in all leachates of raw APC residue samples, with the largest total leaching concentration (61.60 ngm(-3); 0.30 ngI-TEQm(-3)) from treatment with humic acid. Low-chlorinated congeners were mainly leached with LAS and n-hexane. Solidification and stabilization (S/S) processes with cement and sulfur-containing chelating agent decreased the leachability of PCDD/Fs by up to 98% with humic acid and LAS as solvents. However, S/S processes enhanced the leachability of both high- and low-chlorinated PCDD/F congeners with n-hexane as the solvent, which largely increased the toxic equivalent quantity of leachates. These results suggest that conventional S/S processes may effectively restrain the release of PCDD/Fs when APC residues are leached with rain water or natural organic compounds (e.g., humic acid), but may have a deteriorated effect when APC residues are leached with nonpolar organic solvents (e.g., n-hexane) coexisting in the landfill sites.  相似文献

ABSTRACT: A distributed watershed model combining kinematic wave routing, 1‐D dynamic channel‐flow routing, and 2‐D diffusive overland‐flow routing has been developed to simulate flooding and inundation levels of large watersheds. The study watershed was linked to a GIS database and was divided into an upstream mountainous area and a downstream alluvial plain. A kinematic wave routing was adopted at the mountainous area to compute the discharge flowing into the alluvial plain. A 1‐D dynamic channel routing solving the St. Venant equations by the Preissmann method was performed for the main channel of the alluvial plain, whereas a 2‐D overland‐flow routing solving the diffusion wave equation with the Alternating Direction Explicit scheme was used for floodplains. The above two routings were connected by weir‐link discharge formula. The parameters in the model were calibrated and independently verified by single‐event storms. An example application of flooding/inundation analysis was conducted for the Taichung station and the Woozi depot (Taiwan High Speed Rail). Suggested inundation‐proofing measures ‐ including raising ground surface elevation of the station and depot and building a waterproofing exterior wall and their combination ‐ were investigated. It was concluded that building the waterproofing exterior wall had a strong tendency to decrease peak inundation depth.  相似文献

Thermal treatment is a useful tool to remove Hg from contaminated soils. However, thermal treatment may greatly alter the soil properties and cause the coexisting contaminants, especially trace metals, to transform and repartition. The metal repartitioning may increase the difficulty in the subsequent process of a treatment train approach. In this study, three Hg-contaminated soils were thermally treated to evaluate the effects of treating temperature and duration on Hg removal. Thermogravimetric analysis was performed to project the suitable heating parameters for subsequent bench-scale fixed-bed operation. Results showed that thermal decontamination at temperature >400 °C successfully lowered the Hg content to <20 mg kg−1. The organic carbon content decreased by 0.06-0.11% and the change in soil particle size was less significant, even when the soils were thermally treated to 550 °C. Soil clay minerals such as kaolinite were shown to be decomposed. Aggregates were observed on the surface of soil particles after the treatment. The heavy metals tended to transform into acid-extractable, organic-matter bound, and residual forms from the Fe/Mn oxide bound form. These results suggest that thermal treatment may markedly influence the effectiveness of subsequent decontamination methods, such as acid washing or solvent extraction.  相似文献

Abstract

In order to develop a prefabricated treatment and reuse plant for diluted pig wastewater, an entrapped‐mixed‐microbial‐cell (EMMC) process was evaluated for its process performance and economic analysis. At the hydraulic retention time (HRT) of 30 hrs (loading rate of 1.0 g TCOD/L/d) and intermittent aeration of 1 hr of aeration and 1 hr of non‐aeration, it was found that, by using the pretreatment of the ammonium crystallization, both the medium and large carriers were able to reduce TCOD, SCOD, and T‐N by 83.51, 84.11, and 95.10%, respectively. The EMMC unit and lime post‐treatment followed by ammonium crystallization can reduce BOD5, TCOD, SCOD, TSS, T‐N, and T‐P, respectively by 99.22, 93.85, 92.67, 97.73, 96.43, and 97.27%. The treated wastewater, after disinfection, is able to meet the requirements of the standards issued by the USEPA for reuse of food crops. The economic analysis indicates that based on the process performance of the EMMC unit, a prefabricated wastewater treatment plant for 2000 pigs has comparable net present worth (NPW) comparing intermittent aerated biological systems and less operation and maintenance and land requirement than conventional biological processes for removal carbon and nitrogen. A farm operation of more than 2000 pigs meets the unit cost of US$4.91/pig/yr. This will minimize the problems pertaining to technical factors or considerations that heavily influence planning, construction and operation of a pig wastewater treatment system.  相似文献
