Trophoblast deportation is known to occur in normal human pregnancy, but it is not yet clear whether these cells routinely enter the maternal peripheral circulation and are available as a source of fetal DNA for non-invasive prenatal diagnosis of genetic disorders. To resolve this issue requires an efficient method of enriching trophoblast from maternal blood combined with a means to confirm its identity. Five different techniques were tested on ten retroplacental blood samples to determine the most sensitive and operator-efficient method. Lysis of red cells alone gave the best recovery of trophoblast but had to be discounted, together with Ficoll density gradient centrifugation, due to the very low purity and the excessive time required. Fluorescence-activated cell sorting (FACS) of pre-enriched trophoblast resulted in the lowest recovery rate (8 per cent) despite a 3250-fold enrichment and a very high purity. Immunomagnetic beads (Dynabeads) coated with anti-CD 16 antibody proved to be the best method for the subsequent immunocytochemical characterization of deported trophoblast. However, IO beads coated with anti-CD45 antibody may be more useful for isolating trophoblast for prenatal diagnosis due to the high purity, enrichment (32-fold), and recovery rate (78 per cent) obtained with this method. 相似文献
Sandy beach habitat where sea turtles nest will be affected by multiple climate change impacts. Before these impacts occur, knowledge of how nest site selection and hatching success vary with beach microhabitats is needed to inform managers on how to protect suitable habitats and prepare for scientifically valid mitigation measures at beaches around the world. At a highly successful green turtle (Chelonia mydas) rookery at Akumal, Quintana Roo, Mexico, we measured microhabitat characteristics along the beach crawl (rejected sites) and related nest site conditions (selected sites) to subsequent hatching success rates for 64 nesting events. To our knowledge, this is the first study to report environmental data along the nesting crawl for a green turtle population and the first to use natural breaks in the data to describe their preferred habitat ranges. Our results indicate that turtles were likely using a combination of cues to find nest sites, mainly higher elevations and lower sand surface temperatures (Kruskal-Wallis test, H?=?19.84, p?<?0.001; H?=?10.78, p?<?0.001). Hatching success was significantly and negatively correlated to sand temperature at cloaca depth (Spearman’s ρ?=??0.27, p?=?0.04). Indeed, the preferred range for cloaca sand temperatures at the nest site (26.3–27.5 °C) had significantly higher hatching success rates compared to the highest temperature range (Tukey HSD?=?0.47, p?=?0.05). Sand temperatures at various depths were intercorrelated, and surface and cloaca depth sand temperatures were correlated to air temperature (ρ?=?0.70, p?=?0.00; ρ?=?0.26, p?=?0.04). Therefore, rising air temperatures could alter sand temperature cues for suitable nest sites, preferred nest site ranges, and produce uneven sex ratios or lethal incubating temperatures. The 137Cs activity of salt marsh and tidal flat sediments of the northern part of the European Wadden Sea was studied based on a comprehensive dataset of 210Pb dated cores. The 137Cs inventory of salt marsh sediments shows a major peak corresponding to the Chernobyl accident in 1986, and a minor peak located in the late 1960s interpreted as the combined effect of atmospheric testing of nuclear weapons. Emissions from the nuclear reprocessing plant Sellafield are not reflected as peaks in 137Cs activity, but may contribute to the rising 137Cs activity in the years prior to 1986. The 137Cs activity of tidal flat sediments differs from salt marsh sediment in two respects. First, the activity is much lower and, second, the major peak in the 1980s is located in the beginning instead of in the middle of the decade. The differences in 137Cs inventory between the two environments are interpreted to result from repeated cycles of deposition/resuspension and mixing on tidal flats. A simple model illustrating the consequence of mixings returns an apparent shift of major peaks in 137Cs activities backwards in time corresponding to the mixing depth divided by the deposition rate. 相似文献
The sorption and desorption behavior of benzidine in eight solvent-sediment systems were studied using a batch method. The solvents tested included deionized water (DI), calcium chloride solution (CaCl2), sodium hydroxide solution (NaOH), acetonitrile (ACN), a mixture of acetonitrile and ammonium acetate solution (ACN-NH4OAc), methanol (MeOH), ammonium acetate solution (NH4OAc) and hydrochloric acid solution (HCl). Three sets of sorption isotherm experiments were conducted separately in these eight solvents with seven days, three weeks, and two months of contact times, respectively. The results demonstrated nonlinear benzidine sorption phenomena in all eight solvents with higher sorption affinities for sediment sites in the aqueous solvents than in the organic solvents. The results from the desorption experiments revealed that the benzidine desorption efficiencies in the solvents decreased in an order, which was approximately the reverse order of its sorption affinity. Results also suggested that hydrophobic partitioning and covalent binding processes dominated in the desorption experiments, while cation exchange process had little effect on desorption of benzidine. A three-stage model was subsequently applied to simulate the desorption data in the selected solvents of ACN, ACN-NH4OAc and NaOH, respectively. The rapidly desorbing initial fractions were about 0.13-0.20, 0.15-0.26, and 0.18-0.25 for ACN, ACN-NH4OAc and NaOH, respectively. Finally, the sorbed concentrations of benzidine in slowly and very slowly desorbing domains in the selected solvents were correlated with the maximum sorption capacities obtained from the Langmuir sorption isotherm model. The maximum sorption capacities of benzidine were found to be comparable to the amount of benzidine residing in the slowly and very slowly desorbing domains. 相似文献
The influence of the presence of the so-called seed particles on the emission rate of Tris (1-chloroisopropyl) phosphate (TCIPP) from polyisocyanurate (PIR) insulation boards was investigated in this study. Two Field and Laboratory Emission Test cells (FLEC) were placed on the surface of the same PIR board and respectively supplied with clean air (reference FLEC) and air containing laboratory-generated soot particles (test FLEC). The behavior of the area-specific emission rates (SERA) over a time period of 10 days was studied by measuring the total (gas?+?particles) concentrations of TCIPP at the exhaust of each FLEC. The estimated SERA of TCIPP from the PIR board at the quasi-static equilibrium were found to be 0.82 μg m?2 h?1 in the absence of seed particles, while the addition of soot particles led to SERA of 2.16 μg m?2 h?1. This indicates an increase of the SERA of TCIPP from the PIR board with a factor of 3 in the presence of soot particles. The TCIPP partition coefficient to soot particles at the quasi-static equilibrium was 0.022?±?0.012 m3 μg?1. In the next step, the influence of real-life particles on TCIPP emission rates was investigated by supplying the test FLEC with air from a professional kitchen where mainly frying and baking activities took place. Similar to the reference FLEC outcomes, SERA was also found to increase in this real-life experiment over a time period of 20 days by a factor 3 in the presence of particles generated during cooking activities.
