Soil metal pollution can trigger evolutionary adaptation in soil-borne organisms. An in vitro screening test showed cadmium adaptation in populations of Suillus luteus (L.: Fr.) Roussel, an ectomycorrhizal fungus of pine trees. Cadmium stress was subsequently investigated in Scots pine (Pinus sylvestris L.) seedlings inoculated with a Cd-tolerant S. luteus, isolated from a heavy metal contaminated site, and compared to plants inoculated with a Cd-sensitive isolate from a non-polluted area. A dose-response experiment with mycorrhizal pines showed better plant protection by a Cd-adapted fungus: more fungal biomass and a higher nutrient uptake at high Cd exposure. In addition, less Cd was transferred to aboveground plant parts. Because of the key role of the ectomycorrhizal symbiosis for tree fitness, the evolution of Cd tolerance in an ectomycorrhizal partner such as S. luteus can be of major importance for the establishment of pine forests on Cd-contaminated soils.  相似文献

Gentle remediation options (GRO) are risk management strategies/technologies that result in a net gain (or at least no gross reduction) in soil function as well as risk management. They encompass a number of technologies, including the use of plant (phyto‐), fungi (myco‐), and/or bacteria‐based methods, with or without chemical soil additives or amendments, for reducing contaminant transfer to local receptors by in situ stabilization, or extraction, transformation, or degradation of contaminants. Despite offering strong benefits in terms of risk management, deployment costs, and sustainability for a range of site problems, the application of GRO as practical on‐site remedial solutions is still in its relative infancy, particularly for metal(loid)‐contaminated sites. A key barrier to wider adoption of GRO relates to general uncertainties and lack of stakeholder confidence in (and indeed knowledge of) the feasibility or reliability of GRO as practical risk management solutions. The GREENLAND project has therefore developed a simple and transparent decision support framework for promoting the appropriate use of gentle remediation options and encouraging participation of stakeholders, supplemented by a set of specific design aids for use when GRO appear to be a viable option. The framework is presented as a three phased model or Decision Support Tool (DST), in the form of a Microsoft Excel‐based workbook, designed to inform decision‐making and options appraisal during the selection of remedial approaches for contaminated sites. The DST acts as a simple decision support and stakeholder engagement tool for the application of GRO, providing a context for GRO application (particularly where soft end‐use of remediated land is envisaged), quick reference tables (including an economic cost calculator), and supporting information and technical guidance drawing on practical examples of effective GRO application at trace metal(loid) contaminated sites across Europe. WhenPseudomonas oleovorans (GPo1) is grown on sodium octanoate under ammonium limiting conditions, it is able to accumulate a copolyester consisting of medium chain length 3-hydroxyalkanoic acids (PHAm). 3-Hydroxybutyrate is only incorporated in trace amounts. WhenP. oleovorans is equipped with the PHB biosynthetic genes ofAlcaligenes eutrophus (GPo1[pVK101::PP1]), it forms a polyester containing major amounts of 3-hydroxybutyrate. The resulting polymer however is a blend of PHAm and PHB, rather than a copolymer of 3-hydroxybutyrate and medium chain length 3-hydroxyalkanoic acids [11]. To establish whether PHAm and PHB molecules are stored in the same or separate granules by this recombinantP. oleovorans strain, we studied polymer forming cells by freeze-fracture electron microscopy. This approach is possible because previous freeze-fracture electron microscopy studies on PHAm and PHB accumulating strains have shown that PHAm and PHB granules can be distinguished from each other: PHAm granules from mushroom-like structures, whereas PHB granules from needle structures during freeze-fracturing. In this paper we show that stationary phase cells of GPo1[pVK101::PP1] contained both mushroom and needle-like structures, indicating that PHAm and PHB chains were stored in separate granules. To be able to determine whether the separation of PHAm and PHB is complete, the respective granules were separated on sucrose gradients. A total cell extract of GPo1[pVK101::PP1] which was subjected to sucrose gradient centrifugation revealed two white bands of different densities: the upper band with a density of 1.05 g/mL consisted exclusively of PHAm granules, while the lower band with a density of 1.19 g/mL consisted of PHB granules only. Thus, when bacteria synthesize both PHAm and PHB, the resulting polymer chains are segregated completely and stored in separate granules.  相似文献

Biosensors for detection of mercury in contaminated soils   总被引:1,自引:0,他引:1  
Biosensors based on whole bacterial cells and on bacterial heavy metal binding protein were used to determine the mercury concentration in soil. The soil samples were collected in a vegetable garden accidentally contaminated with elemental mercury 25 years earlier. Bioavailable mercury was measured using different sensors: a protein-based biosensor, a whole bacterial cell based biosensor, and a plant sensor, i.e. morphological and biochemical responses in primary leaves and roots of bean seedlings grown in the mercury-contaminated soil. For comparison the total mercury concentration of the soil samples was determined by AAS. Whole bacterial cell and protein-based biosensors gave accurate responses proportional to the total amount of mercury in the soil samples. On the contrary, plant sensors were found to be less useful indicators of soil mercury contamination, as determined by plant biomass, mercury content of primary leaves and enzyme activities.  相似文献

32 Dutch human milk samples were analyzed for PCBs with either HRGC-ECD or HRGC-LRMS in the NCI mode. Samples were collected from three different locations in The Netherlands: Amsterdam, Rotterdam and Groningen. Quantitatively, no differences could be observed between the three localities, while in addition the congener specific pattern showed a striking similarity for all individual samples. Only principal component analysis revealed slight individual differences. Based on similarities in the PCB profiles, linear relationships were calculated between 2,3′4,4′,5-PnCB (#118) or 2,2′4,4′5,5′HxCB (#153) and the most relevantnon andmonoortho PCBs exhibiting dioxinlike activity. These PCBs included 2,3,3′,4,4′-PnCB (#105), 3,3′,4,4′5-PnCB (#126) 2,3,3′,4,4′,5-HxCB (#156), 2,3,3′,4,4′,5′-HxCB (#157), 2,3′,4,4′,5,5′-HxCB (#167) and 3,3′,4,4′,5′5-HxCB (#169). Good linear relationships were observed between individual PCBs. Based on the results of this study, PCB #118 can be used to predict concentrations of the PCBs #105 and #126. PCB #153 can be used as a predictor for the PCBs #156, #157, #167 and #169, but also for the total toxic equivalencies (TEQs) ofnon andmonoortho PCBs present in human milk. This method using certain PCBs as predictors for other toxicological relevant congeners, can be useful and cost effective, e.g. for epidemiological studies. However, before applied a number of conditions should be met. These are:
  1. A stable composition of the PCB matrix should be established.
  2. A possible time dependent change in composition of the matrix should first be excluded when used over different time periods.

An international inter-laboratory research program investigated the effectiveness of in situ remediation of soils contaminated by cadmium, lead and zinc, measuring changes in soil and soil solution chemistry, plants and soil microbiota. A common soil, from mine wastes in Jasper County MO, was used. The soil was pH 5.9, had low organic matter (1.2 g kg(-1) C) and total Cd, Pb, and Zn concentrations of 92, 5022, and 18 532 mg kg(-1), respectively. Amendments included lime, phosphorus (P), red mud (RM), cyclonic ashes (CA), biosolids (BIO), and water treatment residuals (WTR). Both soil solution and NH4NO3 extractable metals were decreased by all treatments. Phytotoxicity of metals was reduced, with plants grown in P treatments having the highest yields and lowest metal concentration (0.5, 7.2 and 406 mg kg(-1) Cd, Pb, and Zn). Response of soil micro-organisms was similar to plant responses. Phosphorus addition reduced the physiologically based extraction test Pb from 84% of total Pb extracted in the untreated soil to 34.1%.  相似文献

Flat lizards (Platysaurus broadleyi) at Augrabies Falls National Park, South Africa, are restricted to rocky terrain where the predominant tree is the Namaqua fig (Ficus cordata cordata). P. broadleyi readily feeds on Namaqua figs when they are available, and the lizards sometimes form large (maximum recorded=134) congregations under fruiting fig trees. The distance lizards travel to fruiting trees also exceed normal daily foraging distances. Location of fruiting fig trees by a lizard can have a high pay-off because figs are energetically rich and trees fruit irregularly and asynchronously, resulting in a resource that is available, but unpredictable in time and space. The prediction that bird activity in fig trees provides a cue to the presence of ripe figs was tested experimentally. By placing cages containing birds and empty control cages in trees devoid of fruit, we demonstrated that P. broadleyi are drawn to fig trees with high bird congregations. We also tested if the presence of a fig tree was necessary to draw lizards to bird congregations by placing cages containing birds and empty control cages in a matched-pairs design on rock away from trees. Humpback whales (Megaptera novaeangliae) in the Southeastern Atlantic Ocean (International Whaling Commission ‘Breeding Stock B’—BSB) are distributed from the Gulf of Guinea to Western South Africa. Genetic data suggest that this stock may be sub-structured, but it remains unknown if this is due to reproductive segregation. This paper evaluates the spatial and temporal population structure of BSB humpback whales using a combination of maternally and bi-parentally inherited markers. The genetic differentiation that we identify in this study could be due to a combination of (1) spatial and/or temporal segregation on breeding grounds in the greater Gulf of Guinea, (2) the possibility of maternally inherited site fidelity to specific feeding grounds and (3) the use of two generalized but exclusive migratory routes (coastal and offshore) between feeding and breeding areas. Further, photo-identification and genetic sampling efforts in other areas of the Sub-Saharan Western Africa winter range and targeted deployment of satellite tags would help to clarify some of the apparent complexity in the population structure of animals biopsied in this region.  相似文献

BACKGROUND AND AIM: Following to the 2006 climate summit, the European Union formally set the goal of limiting global warming to 2 degrees Celsius. But even today, climate change is already affecting people and ecosystems. Examples are melting glaciers and polar ice, reports about thawing permafrost areas, dying coral reefs, rising sea levels, changing ecosystems and fatal heat periods. Within the last 150 years, CO2 levels rose from 280 ppm to currently over 400 ppm. If we continue on our present course, CO2 equivalent levels could approach 600 ppm by 2035. However, if CO2 levels are not stabilized at the 450-550 ppm level, the consequences could be quite severe. Hence, if we do not act now, the opportunity to stabilise at even 550 ppm is likely to slip away. Long-term stabilisation will require that CO2 emissions ultimately be reduced to more than 80% below current levels. This will require major changes in how we operate. RESULTS: Reducing greenhouse gases from burning fossil fuels seems to be the most promising approach to counterbalance the dramatic climate changes we would face in the near future. It is clear since the Kyoto protocol that the availability of fossil carbon resources will not match our future requirements. Furthermore, the distribution of fossil carbon sources around the globe makes them an even less reliable source in the future. We propose to screen crop and non-crop species for high biomass production and good survival on marginal soils as well as to produce mutants from the same species by chemical mutagenesis or related methods. These plants, when grown in adequate crop rotation, will provide local farming communities with biomass for the fermentation in decentralized biogas reactors, and the resulting nitrogen rich manure can be distributed on the fields to improve the soil. DISCUSSION: Such an approach will open new economic perspectives to small farmers, and provide a clever way to self sufficient and sustainable rural development. Together with the present economic reality, where energy and raw material prices have drastically increased over the last decade, they necessitate the development and the establishment of alternative concepts. CONCLUSIONS: Biotechnology is available to apply fast breeding to promising energy plant species. It is important that our valuable arable land is preserved for agriculture. The opportunity to switch from low-income agriculture to biogas production may convince small farmers to adhere to their business and by that preserve the identity of rural communities. PERSPECTIVES: Overall, biogas is a promising alternative for the future, because its resource base is widely available, and single farms or small local cooperatives might start biogas plant operation.  相似文献

A bloom of the diatom Rhizosolenia chunii Karsten sensu Sundström occurred in Port Phillip Bay (38°S; 145°E), southeastern Australia, between late August and mid-October 1987. Coincident with this bloom, mussels (Mytilus edulis planulatus), scallops (Pecten alba) and flat oysters (Ostrea angasi) throughout the Bay developed a bitter taste that was so unpleasant and persistent that mussels became unmarketable for 7 mo. The bitter taste was concentrated in the digestive gland, which also showed extensive inflammation and degeneration. Digestive gland lesions were evident in mussels during September 1987, but became progressively more severe and, in the chronic end-stage were associated with high shellfish mortality 3 to 8 mo after the bloom had ceased. This study provides strong indirect evidence that the bitterness and the toxicity were caused by a bloom of R. chunii. R. chunii is the first species of diatom to be associated with shellfish mortality and, as the mortality occurred many months after the bloom had ceased, we suggest that the toxicity of certain algal blooms may be difficult to detect rather than uncommon.  相似文献
