Three monoclonal antibodies (MAbs) against trophoblast (GB17, GB21, and GB25) and flow cytometry were used to sort trophoblast-like cells (TLCs) from peripheral blood of pregnant women. Sorted TLCs were processed for electron microscopy and fetal DNA amplification of the Y-specific sequences from mothers carrying male fetuses. At the ultra-structural level, most of the nucleated cells had the morphology of leucocytes, suggesting maternal contaminants, and we did not find the characteristic features of the free inter-villous trophoblast cells. Nevertheless, polymerase chain reaction (PCR) analysis showed an amplification of Y-specific sequences in two out of three samples of sorted TLCs. These results suggest that besides the maternal leucocytes, sufficient trophoblast nucleated fetal cells can be obtained using cell enrichment by sorting. This sensitive method holds promise for non-invasive prenatal diagnosis of fetal sex and if sufficient Y(positive) nuclei are found, for the diagnosis of selected numerical chromosome abnormalities.  相似文献

The basal non-mammaliaform cynodonts from the late Permian (Lopingian) and Early Triassic are a major source of information for the understanding of the evolutionary origin of mammals. Detailed knowledge of their anatomy is critical for understanding the phylogenetic transition toward mammalness and the paleobiological reconstruction of mammalian precursors. Using micro-computed tomography (μCT), we describe the internal morphology of the interorbital region that includes the rarely fossilized orbitosphenoid elements in four basal cynodonts. These paired bones, which are positioned relatively dorsally in the skull, contribute to the wall of the anterior part of the braincase and form the floor for the olfactory lobes. Unlike procynosuchids and the more basal therapsids in which the orbitosphenoids are well developed, dense, and bear a ventral keel, the basal epicynodonts Cynosaurus, Galesaurus, and Thrinaxodon display cancellous, reduced, and loosely articulated orbitosphenoids, a condition shared with many eucynodonts. The hemi-cylindrical orbitosphenoid from which the mammalian condition is derived re-evolved convergently in traversodontid and some probainognathian cynodonts.  相似文献

In this paper we address two important aspects of micro-scale urban airflow model evaluation: (a) the identification of key flow features as dictated by the physics of the problem and as captured by the simulations, and (b) the comparison of important model output parameters (mean flows and fluctuations) with experimental data. A series of mesh-adaptive large eddy simulations (LES) was carried out for the study of air flows within two intersecting street canyons with varying building configurations. The novelty of the approach lies in the combination of LES with mesh adaptivity, which allows a variable-filter length and the implementation of an anisotropic eddy-viscosity model. Both coarse and fine-mesh simulations were carried out, using single and parallel-processor systems respectively. The simulations showed clearly that the expected flow patterns such as the street canyon recirculation and the street-mouth vortices, as well as the exchange of air flow at the street intersections, can readily be captured by the mesh-adaptive LES.In addition, the detailed comparisons of mean flows and fluctuations of the resolved velocity field with the measured data showed that the simulation results agreed well with the patterns and trends of the wind tunnel measurements. In most cases the finer-mesh simulations improved considerably the accuracy of the mean flows, especially for the symmetrical configuration. The improvement in the predicted fluctuations was less obvious, with several detector locations underpredicting the measured values, although the overall comparison was also satisfactory. The typical errors for the mean flows for all three building configurations were less than 30%, whilst for the velocity fluctuations less that 40%. Both the simulated means flows and turbulence levels were generally more accurate in the streets parallel to the wind (streamwise direction) than in the streets normal to the wind.  相似文献

Fossil energy depletion and growing environmental concerns have brought up increasing interest in bio-based eco-efficient and high technology materials. Among them, starch nanocrystals (SNC) consist of crystalline nano-platelets produced from the hydrolysis of starch and mainly used as nano-fillers in polymeric matrix. New applications have brought up the need for scaling-up the SNC preparation process. However, for this new bio-based nano-material to be sustainable, its preparation and processing should have limited impacts on the environment. Thus, together with analyzing and making recommendations for the scaling-up of SNC production process, it is worth identifying “environmentally sensitive” steps using life cycle analysis (LCA). To that purpose, different scenarios have been proposed and compared according to different environmental impacts. Also, a comparison to its main competitor, i.e. organically modified nanoclay (OMMT), is proposed. From a LCA point of view, SNC preparation requires less energy than OMMT extraction, but global warming and acidification indicators were higher than for OMMT. However, SNC have the added advantages to be renewable and biodegradable contrary to OMMT which contribute to non-renewable energy and mineral depletion. Thus, used as filler, SNC have a positive impact on the end of life of the filled material. From these observations, recommendations for the scaling-up of the SNC preparation process are made and deal mainly with the use of land and water.  相似文献

The body of information presented in this paper is directed to air pollution engineers who are concerned with the effect of indirect sources on ambient concentrations of carbon monoxide (CO). Data taken under controlled conditions are used to empirically derive and calibrate a model for predicting CO concentrations in the vicinity of roadway intersections and other points of possible vehicular congestion. Since the predicted free flow CO contribution of vehicles traveling at normal road speeds is relatively low, it is concluded that idling vehicles at points of congestion are the major cause of CO violations, and that state and federal programs should place more emphasis on relieving congestion and reducing idling emission rates in new vehicles.  相似文献

A methodology based on social participation through the use of resident diaries was applied to evaluate the odour annoyance in the surroundings of an industrial park in Belgium during one year. The studied area covers about 8 km2 and includes13 potential odour emitting facilities. The network involved 44 residents in the survey, among whom 19 were particularly considered for a detailed analysis. The questionnaire aimed at providing an odour rating twice-daily on a 6-level scale together with an odour type.The fact that the response rate corresponding to “no-odour” was high (79%) is particularly discussed. Some tests are proposed to check the plausibility of the answers, the coherence within clusters of residents and the individual performance of respondents to discriminate among odour ratings. The odour rose is presented as an attractive and visual tool, particularly suited in the case of multi-source areas, to map the different odour emissions, to point out the most worrying ones, to identify others creating less annoyance and possibly new unpredicted ones. The resident diary method has proven to be particularly useful, conjointly to other ones, to the case of multi-sources facilities in large areas, when the purpose is the assessment of the long-term evolution of odour annoyance.  相似文献

Between January 1988 and February 1989, gizzards were collected from 1155 birds from the groups; Anatidae, Charadriidae, Scolopacidae and Rallidae, shot in the Camargue, and examined for the presence of ingested lead shot. Shot was found in nine out of ten Anatidae species, three of 15 Scolopacidae species and all of three Rallidae species. Twenty-seven per cent of Anatidae sampled had ingested shot, 13.8% had one shot present and 6.3% had more than five. Shot ingestion levels ranged from 3-60% of birds examined according to species. The incidence of shot ingestion did not appear to vary seasonally. In Anatidae, a strong interspecific relationship was found between the grit size ingested and susceptibility to ingest shot (r = 0.94). In Scolopacidae, shot ingestion may have been related to grit size ingested, feeding method and bill length. The levels of shot ingestion presented are for most species higher than have been reported elsewhere in the world. This may result from the absence of grit and presence of high shot densities in Camargue marshes.  相似文献

To examine current genetic-based paradigms pertaining to the structure and possible philopatry of red drum populations, we used solution-based inductively coupled plasma-mass spectrometry (ICP-MS) to analyze the otolith chemistry of juvenile red drum (Sciaenops ocellatus) from eight different estuaries in the Gulf of Mexico (Gulf) and the North Atlantic Ocean. One estuary (Tampa Bay, Fla.) was sampled in three different years. Analyses of variance for five elemental ratios (Mg/Ca, Mn/Ca, Zn/Ca, Sr/Ca, Ba/Ca) were all significantly different between estuaries, as was a multi-element signature (MANOVA, Pillais trace F 50, 1020=19.41, P<0.0001). We also found that red drum from the Gulf could be distinguished from those taken from the Atlantic Ocean with 99.5% accuracy, likely due to differences in water chemistry between these water masses. A discriminant function developed using these elemental ratios was more than 80% accurate in assigning juvenile red drum to their natal estuary, or in the case of Tampa Bay, to the correct year of spawning. We also used laser ablation ICP-MS to examine the otolith core chemistry of adult red drum collected from spawning aggregations near Tampa Bay. Using a discriminant function analysis with a calibration data set derived from juvenile signatures, we found that 75% of the adult cores matched the juvenile signal established for Tampa Bay 1982. Although preliminary, the results presented here suggest that red drum may return to their natal estuary to spawn, which has been postulated from genetic data.Communicated by P.W. Multixenobiotic defence mechanism (MXDM) has been recently described as a first line defence system in many aquatic organisms and it has raised attention for its potential as an early biomarker of exposure to environmental stress. In order to investigate the relevance of this biomarker in the freshwater zebra mussel Dreissena polymorpha, we examined its response to exposure and to cessation of exposure. For this purpose, depurated zebra mussels were transplanted to natural rivers or exposed to medium experimentally contaminated with fuel oil 2 in the laboratory. After the exposure period, mussels were submitted to depuration in clean water in the laboratory. The level of MXDM activity was assessed by the efflux method that allows a simple and fast measure of the rate of rhodamine B efflux. MXDM activity was induced following exposure and induction appears earlier after laboratory exposure (3 days) than after in the field exposure (3-14 days). The level of MXDM response could not be directly correlated to exposure dose in the laboratory and to isolated physicochemical parameters or AOX levels in the field. The behaviour of MXDM activity seems to reflect the occurrence of exposure to a global stress rather than to specific compounds. Our results highlighted the possibility to monitor MXDM response in the freshwater zebra mussel. In spite of the difficulty to interpret responses resulting from the lack of specificity of response, multixenobiotic defence mechanism displays qualities to represent a biomarker of general stress in freshwater ecosystems.  相似文献
