SSCP技术解析硫酸盐还原反应器中微生物群落结构   总被引:17,自引:5,他引:12  
采用改进的单链构象多态性(SSCP)技术,以16SrRNA基因的V3区为靶对象,分析完全混合式硫酸盐还原反应器中微生物的群落结构以及硫酸盐还原菌(SRBs)与产酸菌(ABs)的种间关系.共得到13个可辨晰的SSCP条带,对其中的6条带(A1,A3,A4,A5,A9,A10)进行了测序分析,分别同嗜柠檬酸明串球菌(GenBank登录号:AY453065,下同)、未培养细菌(AJ318147,AF227834,AJ576427)、产乙醇杆菌(AY434722)、梭杆菌(AB084627)等相似性较大.为检测系统中的SRBs,以SRBs富集培养基,对反应器中的活性污泥进行选择性富集,培养的混合菌群同样采用SSCP分析,带型与前者相差较大,其中有2条带与Bacteroidetes(AB074606)和脱硫弧菌(Y12254,U42221)最为相似.分析表明,SRBs在总DNA中的含量可能不到1·5%,但是它们与大量的ABs形成很好的种间协作关系,从而维持工艺系统较高的硫酸盐去除率和运行稳定性.  相似文献

Many species of platyrrhine primates are characterised by sex-linked color vision polymorphism. This presents an opportunity to study the biology and ecology of individuals with different phenotypes living in the same group. Several evolutionary processes could maintain polymorphic genes in populations. In this study, we evaluate the hypothesis that foraging niche divergence among monkeys explains the presence of multiple color vision phenotypes. Specifically, we test whether dichromats and trichromats differ in foraging time devoted to cryptic vs brightly colored resources. We did not find any differences in foraging time spent on different food types by dichromatic and trichromatic monkeys in two groups of white-faced capuchins (Cebus capucinus) living in a tropical dry forest. We conclude that in so far as these variables are concerned, niche divergence does not likely explain color vision polymorphism in our study population.  相似文献

Most small-colony termites live confined within a single piece of wood on which they feed and do not possess permanent workers: Tasks are done by developmentally flexible immatures (pseudergates). By contrast, large-colony termites possess a specialized (true) worker caste and forage outside their nest for food. To shed light on possible transitional steps between these contrasting patterns of social organization, we studied an atypical Rhinotermitidae, Prorhinotermes inopinatus. In this species, despite the absence of a true worker caste, soldiers, pseudergates, and neotenic reproductives may leave the nest and explore their surroundings. Although evidence presented in this study indicates that termites recognize unknown areas, there is no directional recruitment toward them. The discovery of a food source, i.e., a piece of wood, is followed by the establishment of a long-lasting trail between the nest and the food source. A large fraction of the colony, including neotenic reproductives, ultimately migrates into the piece of wood. Our results thus demonstrate that multiple features of external foraging behavior can evolve independently of the existence of a true worker caste in termites. We suggest that large colonies with true workers, like those of most Rhinotermitidae, may easily have evolved from a Prorhinotermes-like pattern if submitted to increasing selective pressures for worker efficiency in a stable environment.  相似文献

Reduction in DNA repair capacity is associated with increased rates of birth defects, cancer, and accelerated ageing. Genetic polymorphisms in DNA repair genes might influence the repair activities of the enzymes predisposing individuals to cancer risk. Owing to the presence of these genetic variants, inter-individual and ethnic differences in DNA repair capacity have been observed in various populations. India harbors enormous genetic, cultural and linguistic diversity. The present study was undertaken to determine the allele and genotype frequencies of four non-synonymous SNPs, XRCC1 Arg399Gln (C > T, rs25487), XRCC3 Thr241Met (G > A, rs861539), XPD Lys751Gln (T > G, rs13181), and OGG1 Ser326Cys (C > G, rs1052133) in the Maharashtrian population, residing in the Vidarbha region of central India and to compare them with HapMap and other Indian populations. The variant alleles of these polymorphisms have been found to be positively associated with different forms of cancer in several genetic epidemiological studies. The basic prevalence of these polymorphisms in the general population must be known to evaluate their significance in risk assessment in cancer and other phenotypes. About 215 healthy and unrelated individuals from the Maharashtrian population were genotyped for each of these four polymorphisms using PCR-RFLP. The allele and genotype frequency distribution at the four DNA repair gene loci among Maharashtrians revealed a characteristic pattern. To the best of our knowledge, this is the first report of these DNA repair gene polymorphisms in a central Indian population.  相似文献

The two types of males in the ant genus Cardiocondyla differ remarkably in morphology and behavior. Ergatoid males are wingless fighters whose spermatogenesis continues throughout their entire adult lives and which therefore have an “unlimited” sperm supply. They attempt to kill all eclosing ergatoid rivals and thus to increase their share in copulations with the virgin queens reared in their nests. Winged males, on the other hand, are docile and emigrate from the nests a few days after eclosion, probably to mate with queens from other colonies. By this time, their testes have fully degenerated and all sperm is stored in the seminal vesicles. Before emigration, winged males may mate with virgin queens in their maternal nests, but they are nevertheless rarely attacked by ergatoid males. In the laboratory, the life expectancy of ergatoid males is only slightly higher than that of winged males, but because of the emigration of the latter the difference is likely to be more pronounced in the field. Both male morphs are capable of inseminating more than 35 virgin queens. However, winged males older than 14 days mate less often than ergatoid males of similar age, probably due to sperm depletion in later life. The spermathecae of queens inseminated by ergatoid males contained significantly more sperm than those of queens which mated with winged males. Identifying the factors that mediate covariation between an ornament and other phenotypic attributes is important to determine the signaling function of ornaments. Sign and magnitude of a covariation may vary across environments if the expression of the ornament or of its linked genes regulating correlated phenotypes is condition-dependent. I investigated in the barn owl Tyto alba whether sign and magnitude of covariation between body mass and two heritable melanin-based plumage ornaments change with food supply, along the reproductive cycle and from the morning to the evening. Using a dataset of 1,848 measurements of body mass in 336 breeding females, I found that females displaying large black spots were heavier than conspecifics with smaller spots in the afternoon (i.e., a long time after the last feeding) but not in the morning (i.e., a short time after the last feeding). This is consistent with the recently proposed hypothesis that eumelanin-based ornaments are associated with the ability to maintain energy balance between food intake and energy expenditure. Thus, covariation between melanin-based coloration and body mass can be detected only under specific conditions potentially explaining why it has been reported in only ten out of 28 vertebrate species. The proposition that ornamented individuals achieve a higher fitness than drab conspecifics only in specific environments should be tested for other ornaments.  相似文献
