利用表面活性剂TritonX-100建立一种提取和测定氧化葡萄糖酸杆菌(Gluconobacter oxydans)L-山梨糖脱氢酶(L-sorbose dehydrogenaseSDH)活性的简便方法,最适提取条件为:TritonX-100浓度ψ(Triton)/%=0.3。提取温度θ=4℃,处理时间t/h=6-10h;用于提取的菌悬液D550nm范围0.090-0.252。该方法提取效率为细胞碎片法的97.83%。图2表2参7  相似文献

采用[日]岛津AA-670型原子吸收分光光度计,四唑氮蓝(Nitro-BT)染色法,及图象分析仪(MIAS-300)分析了交通警察和男性师生两人群精液的精子运动能力参数和血铅水平。结果显示:与对照组比较,交警组精子活动能力降低(P<0.05),精子琥珀酸脱氢酶(SDH)活性明显抑制(P<0.001)。而且SDH活性与血铅水平呈负相关。认为交警精子活动能力下降可能和铅干扰精子线粒体能量代谢有关。  相似文献

Carp fingerlings exposed to a sublethal concentration (0.5?mg?L?1) of sodium cyanide showed a steady decrement over a 7-day period in respiratory rate, rise in lactate dehydrogenase (LDH), and fall in succinate dehydrogenase (SDH) activities followed by variations in lactic and pyruvate levels. Changes in these enzyme activities might be due to impaired oxidative metabolism and severe cellular damage leading to the release of these enzymes. Decline in the activities of SDH and LDH clearly represents a shift from aerobic to anaerobic metabolism as evidenced by elevated lactate and decline in pyruvate levels. The shift to anaerobic metabolism is also reflected by severe drop in the respiratory rate of the fish. This may be a consequence of the blockage of electron transfer from cytochrome c oxidase to molecular oxygen, thus ceasing cellular respiration and it can lead to cellular hypoxia even in the presence of normal hemoglobin oxygenation. Hence, we indirectly reconfirm the inhibition of oxidative metabolism by sodium cyanide. Alterations in behavioral pattern induced by sublethal sodium cyanide exposure may be due to the combination of cytotoxic hypoxia with lactate acidosis, which depresses the central nervous system (CNS); as the brain is the most sensitive site to anoxia, it results in impaired CNS function.  相似文献

Abstract

The effects of low levels of diazinon treatment on four marker enzymes in rat heart and skeletal muscle have been investigated. Adult male Wistar rats were treated twice a week with a dose of 0.5 ml‐kg‐1day‐1 diazinon for 28 weeks. Diazinon treated rats gained significantly less weight than Sham‐treated controls. Typical differences in Succinate dehydrogenase (SDH), Lactate dehydrogenase (LDH), Phosphofructo kinase (PFK) and Hexokinase (HK) activities were observed between heart and skeletal muscles. Diazinon feeding had no effect on heart, soleus, gastrocnemius and plantaris SDH, LDH and PFK enzyme activities after 28 weeks. HK activity was significantly increased in sham‐control soieus and plantaris muscles after 28 weeks. Diazinon feeding inhibited HK activity in plantaris muscle after 28 weeks treatment These results demonstrate that chronic low levels of diazinon have little effect on the glycolytic and oxidative activity in heart and skeletal muscle.  相似文献

运用透射电镜技术、电镜细胞化学技术、酶组织化学技术和JC-1荧光染色,观察不同浓度镉对河南华溪蟹(Sinopotamon henanense)肝胰腺线粒体的损伤.实验设对照组和5个染毒组:3.56、7 12、14.25、28.49和56.98 mg·L-1,处理时间为72 h.透射电镜观察显示,在低浓度组,线粒体轻度肿胀,部分线粒体外膜破裂,随染毒浓度的增加,线粒体水肿增加,嵴缩短甚至消失,直至线粒体破裂.酶组织化学检测显示,琥珀酸脱氢酶(succinic dehydrogenase,SDH)主要在细胞浆中表达,呈蓝紫色颗粒.SDH活性在3.56、7.12和14.25 mg·L-1浓度组中比对照组显著增加(p<0.05),而在28.49和56.98 mg·L-1浓度组中没有明显变化.SDH的亚细胞定位结果与酶组织化学检测的结果一致.JC-1荧光强度在3.56、7.12和14.25 mg·L-1浓度组比对照组显著增加(p<0.05).线粒体膜电位与SDH的活性有正相关性(r=0.736,p<0.05).结果显示,镉引起线粒体结构损伤、功能障碍,并与镉浓度具有剂量-效应关系.线粒体是镉细胞毒性的重要靶位.  相似文献
