The contamination of food and feed by mycotoxins as toxic metabolites of fungi is a risk not only for consumers resulting in various embarrassment regarding health status and well-being, but also for producers, companies and export market on the ground of economic losses and ruined stability of economic trade. As it is given in historical evidence, the contamination of food by mycotoxins is a topic as old as a history of mankind, finding some evidence even in the ancient books and records. Nowadays, the mycotoxins are used in modern biotechnological laboratories and are considered an agent for targeting the specific cells (e.g., defected cells to eliminate them). However, this promising procedure is only the beginning. More concern is focused on mycotoxins as abiotic hazard agents. The dealing with them, systematic monitoring, and development of techniques for their elimination from agricultural commodities are worldwide issues concerning all countries. They can be found alone or in co-occurrence with other mycotoxins. Thus, this review aims to provide widened information regarding mycotoxins contamination in environment with the consequences on health of animals and humans. The inevitability for more data that correctly determine the risk points linked to mycotoxins occurrence and their specific reactions in the environment is demonstrated. This review includes various symptoms in animals and humans that result from mycotoxin exposure. For better understanding of mycotoxin's impact on animals, the sensitivities of various animal species to various mycotoxins are listed. Strategies for elimination and preventing the risks of mycotoxins contamination as well as economical approach are discussed. To complete the topic, some data from past as historical evidences are presented.  相似文献

Abstract

Penicillic acid and ochratoxin A are environmentally important toxic fungal metabolites (mycotoxins) that are synergistic in combination. The effects of penicillic acid on the pancreatic enzyme, :arboxypeptidase A were investigated in vitro and in vivo. A broad range of inhibition in vitro of the enzyme by PA was demonstrated with a half‐maximal inhibitory concentration equal to 1.1 x 10‐4M PA. Inhibition of carboxypeptidase A was time and temperature dependent, and resulted in decreased conversion of parent ochratoxin A to the non‐toxic metabolite, ochratoxin alpha. Studies in vivo demonstrated a penicillic acid‐dependent inhibition of pancreatic carboxypeptidase A activity in the mouse and the chicken following multiple oral exposure. It is postulated that the mode of toxic interaction of the two mycotoxins may be due, in part, to impaired detoxification of ochratoxin A through peni‐cillic acid depletion of carboxypeptidase A activity.  相似文献

Abstract

Fu monisin B1 (FB1) is a mycotoxin commonly found in maize and maize-based products. Ingestion of FB1-contaminated causes a myriad of dose- and species-dependent toxic effects to human and animal health. In the present study we evaluated the effects of FB1 (8?mg/kg, i.p. for 4?days) on body weight and oxidative stress parameters in the liver, kidney and lung of C57BL/6 male mice. No changes in the organ-to-body weight ratio, organ-to-adrenal gland weight ratio or organ-to-brain weight ratio were found. On the other hand, FB1 exposure increased NPSH levels in liver and lungs whereas decreased FRAP content in liver and kidneys. Levels of TBARS, ascorbic acid and NOx content were not altered by FB1. In summary, four days of FB1 exposure are sufficient to disrupt antioxidant defenses in liver, kidneys and lungs of C57BL/6 male mice without concomitant changes in organs weight.  相似文献

Mycotoxins are harmful substances produced by fungi in several commodities with a widespread presence in foodstuffs. Human exposure to mycotoxins occurs mainly by contaminated food. The quantitation of mycotoxins in cereal-based food, highly consumed by different age population, is of concern. In this survey, 159 cereal-based samples classified as wheat, maize and rice-based, have been evaluated for the occurrence of patulin, deoxynivalenol, 3-acetyl-deoxynivalenol, fusarenon-X, diacetoxyscirpenol, nivalenol, neosolaniol, HT-2, T-2 and zearalenone by gas chromatography–tandem mass spectrometry. Intakes were calculated for average consumers among adults, children and infants and compared with the tolerable daily intakes (TDI). Data obtained were used to estimate the potential exposure levels. 65.4% of the samples were contaminated with at least one mycotoxin and 15.7% of the analyzed samples showed co-occurrence of mycotoxin. The dietary exposure to HT-2 and T-2 toxins was estimated as 0.010 and 0.086 μg kg−1 bw d−1, amounting to 10% and 86% of the TDI, for adults and infants respectively. These results back up the necessity to take a vigilant attitude in order to minimize human intake of mycotoxins.  相似文献

Abstract

Penicillic Acid (PA) and Ochratoxin A (OA) are toxic fungal metabolites that are synergistic in combination. This interaction was investigated using mice which were dosed orally as follows: control, none; solvent control, 0.2 ml bicarbonate buffer; PA, 40 mg/kg; OA, 10 mg/kg and combination, 40 mg/kg PA + 10 mg/kg OA. The only significant histopathologic change observed was an acute multifocal toxic tubular nephrosis which appeared most severe in the combination‐treated mice killed on day 10. While the combination group had a death rate of 20% (5/25), no deaths occurred in the other treatment groups. The increased death rate and the extensive nephrotoxic findings in the combination group indicate a toxic interaction between OA and PA at sub‐lethal dose levels and is consistent with a renal site of action.  相似文献

ABSTRACT

An analytical method based on liquid chromatography–tandem mass spectrometry (LC–MS/MS) was validated and applied for the analysis of aflatoxin M1 (AFM1), ochratoxin A (OTA) and deoxynivalenol (DON) in infant formula and milk-based products for young children commercialized in Brazil. A total of 38 samples were evaluated, including 12 infant formula, 14 follow-on formula and 12 samples of milk-based products. AFM1 was detected in 12 (32%) samples, and seven (18%) samples contained AFM1 levels above the method limit of quantification in a concentration range between 0.013 and 0.067 ng mL?1 (0.026 ± 0.019). Two samples of milk-based products exceeded the maximum level (ML) fixed by the European Union for AFM1 in baby foods, however, all samples were in agreement with the levels established by the Brazilian regulation. OTA and DON were not detected in any of the analyzed samples.  相似文献

The objective of this study was to evaluate the reduction on the levels of the mycotoxin deoxynivalenol (DON) in whole wheat flour (WWF) with different moisture levels, on the wet milling effluent through ozone (O3) processing, as well as the impact of ozonation on the rheological properties of flour. The results have shown that the reduction of DON was improved with increasing moisture and exposure time of WWF and wet milling effluent to ozone. The maximum reduction was about 80%, proving that ozonation is an effective and promising technology in reducing mycotoxins in different products. However, the process altered the rheological profile of WWF. Therefore, further studies are needed to better understand the process.  相似文献

In this study, a two-dimensional liquid chromatography tandem mass spectrometry method was developed and validated for the determination of pesticide residues and contaminants in whole wheat grains and oats. The samples were extracted with a mixture of acetonitrile and water and were injected into the two-dimensional LC-MS/MS system without any further clean-up or sample preparation. Samples were analyzed with four different matrix matched calibrations. Matrix effects were evaluated by comparing analyte signals in the respective matrix matched standard with the neat solvent standards. The final method was validated according to the current Eurachem validation guide and SANTE document. The number of successfully validated analytes throughout all three validation levels in oats and wheat, respectively, were as follows: 330 and 316 out of 370 pesticides, 6 and 13 out of 18 pyrrolizidine alkaloids and 7 out of 9 regulated mycotoxins. Moreover, both plant growth regulators mepiquat and chlormequat as well as the tropane alkaloids atropine and scopolamine met the validation criteria. The majority of pesticides showed limits of detection below 1?µg kg?1, pyrrolizidine alkaloids below 0.7?µg kg?1, tropane alkaloids below 0.2?µg kg?1, growth regulators below 0.7?µg kg?1 and mycotoxins below 8?µg kg?1 in both matrices.  相似文献
