Fetal nucleated erythrocytes (NRBC) in maternal blood are a non-invasive source of fetal DNA for prenatal genetic screening. We compared the effectiveness of three monoclonal antibodies for the separation of fetal cells from maternal blood by flow sorting. Mononuclear blood cells from 49 healthy pregnant women were incubated with antibody to CD 71, CD 36, and/or glycophorin A (GPA), employed singly or in combination with each other. These monoclonal antibodies recognize surface antigens on haematopoietic precursor cells. Successful isolation of fetal cells was defined as detection of Y chromosomal sequences in maternal blood from women carrying male fetuses, with absence of Y sequences when female fetuses were carried. Thus, gender prediction accuracy was used as a measure of fetal cell separation. Using anti-CD 71 to isolate fetal cells, gender prediction was 57 per cent correct; with anti-CD 36, it was 88 per cent correct. Anti-GPA, an erythrocyte-specific antigen, used alone or in combination with anti-CD 71 or 36, improved gender prediction to 100 per cent. We conclude that antibody to GPA improves the retrieval of fetal NRBC from maternal blood, permitting genetic analysis by the polymerase chain reaction.  相似文献

研究了10种常用稻田除草剂对中华大蟾蜍(Bufobufogargarizans)蝌蚪红细胞微核的诱突变效应。蝌蚪在各除草剂试验液中染毒7d,采心脏血制片。结果表明,毒草胺、杀草丹等8种除草剂能诱发蝌蚪红细胞的微核细胞率显著增高,最高达11.5‰(毒草胺、0.5mg/L),与对照组(4.2‰)比较,差异极显著(P<0.01),其中酰胺类,氨基甲酸酯类诱突变效应最强,其0.8mg/L浓度组平均微核细胞率分别为8.2‰和6.5‰。研究中还发现,在一定范围内微核细胞率与除草剂浓度呈正相关,使用浓度下杀草丹等6种除草剂具有明显诱突变效应。  相似文献

将稀有鮈鲫(Gobiocypris rarus)半静态暴露于重铬酸钾溶液中,研究发现稀有鮈鲫的本底微核率处于较低的水平,重铬酸钾在不同浓度和时间暴露后能明显观察到外周血红细胞微核增加。在一定条件下存在剂量-效应关系和时间-效应关系,表明稀有鮈鲫可用于鱼类外周血红细胞微核试验。试验中每尾鱼观察15000个细胞,能有效地减小试验偏差,保证试验结果的可靠性。暴露浓度大于等于0.01mg/L时,染毒组与空白组的外周血红细胞微核率有显著性差异,其微核率随染毒时间的延长呈先升高后下降的趋势,均在24h时出现所有测定时间微核率的峰值。与其他鱼类比较显示,稀有鮈鲫具有较高的敏感性,可用于遗传毒物诱发微核的监测。  相似文献

We have determined whether derivated fetal haemoglobin (dHbF, consisting of glycated and acetylated HbF) can be used as a cell age marker for fetal red blood cells (RBCs). Cord blood was obtained between 19 and 39 weeks of gestation from 28 alloimmunised anaemic fetuses (23 RhD+ and 5 Kell) and from 20 non-anaemic fetuses and newborns (controls). Density gradient centrifugation was applied to 36 samples (20 RhD+, 15 controls and 1 Kell) to obtain fractions of increasing cell age. Blood samples were used for measurements of mean cellular volume (MCV), mean cell haemoglobin (MCH), mean corpuscular haemoglobin concentration (MCHC), pyruvate kinase activity (PK) and derivated fetal haemoglobin (dHbF) by cation-exchange HPLC. Reticulocytes were counted only in the whole blood samples. In all density gradient separated RBC fractions, the values for MCV, MCH and PK activity decreased and those of MCHC and dHbF increased with increasing density (equivalent to increasing cell age). The mean density was lower for RBCs of the anaemic RHD group (1.072±0.007 g/ml) than for the non-anaemic controls (1.077±0.005 g/ml) (p<0.05) The RBC density of the Kell sensitised fetus did not differ from those of the controls. In the control group, the values of the cell age markers in whole blood changed significantly with the gestational age, showing an increase of mean age of the erythrocyte population. The best linear relationship was found for dHbF (y=6.28+0.17*weeks; r=0.84; p<0.001). In the anaemic RhD+ fetuses, the RBC age markers did not change with gestational age; the dHbF percentages were lower, and the MCV, MCH, PK values and the reticulocyte counts were higher than in the controls (0.05<p<0.001). The dHbF values of the Kell sensitised fetuses were above (p<0.01) and the reticulocyte counts were below normal (p<0.05) for gestational age. For the anaemic fetuses, a significant number of the dHbF values (86%) and of the reticulocyte counts (78%) differed from the values of the controls (p<0.01). The dHbF percentages in RhD+ fetuses showed the best correlation with the Hb deficit, which is a measure for anaemia (r=−0.81, p<0.0001). We conclude that the percentage derivated HbF may indicate whether the RBC production is normal for gestational age. It may in that sense reflect stimulated or impaired erythropoiesis in alloimmunised haemolytic anaemia. Copyright © 2001 John Wiley & Sons, Ltd.  相似文献

Thymol, the main phenolic compound in Thymus vulgaris, has been shown to have various biological effects. The main objective of this study was to investigate the efficacy of thymol on counteracting hexavalent chromium-induced oxidative damage in rat erythrocytes in vitro. The radical scavenging activity of thymol was examined using the 1, 1-diphenyl-2-picrylhydrazyl assay. Erythrocytes resistance to oxidative damage, lipid peroxidation, osmotic pressure, hemolysis as well as morphological alterations were evaluated in the presence of 2.5 µg thymol mL?1 with or without 5 µmol hexavalent chromium mL?1 of the incubation media. Results from the 1,1-diphenyl-2-picrylhydrazyl assay denoted good radical scavenging activity of thymol. Thymol caused a significant increase in superoxide dismutase and catalase activities and reduced glutathione content in erythrocytes intoxicated with hexavalent chromium. In contrast, the presence of thymol resulted in markedly less-elevated malondialdehyde levels, hemolysis, and destabilization of erythrocytes exposed to hexavalent chromium. Microscopically, thymol markedly reduced hexavalent chromium-induced morphological alterations in rat red blood cells. Conclusively, thymol counteracted hexavalent chromium-induced oxidative damage in rat erythrocytes.  相似文献

利用泥鳅红细胞核的遗传损伤监测福州市内河水质,探讨红细胞核的遗传损伤与水质常规衡量指标间的相关性.结果表明:泥鳅红细胞微核率及核异变率的高低能够反映出河水的综合污染程度,差异达到显著或极显著.在一定范围内河水受污染程度越严重,微核及核异常率越高.但当染毒时间太长时,微核率及核异常率反而有所下降.微核率与水质的几项常规衡量指标间的相关程度依次为:高锰酸盐指数 >溶解氧 >氨氮比 >pH值;总核异常率与河水的几项常规指标间的相关程度依次为:高锰酸盐指数 >氨氮比>溶解氧>pH值.核异常率与微核率达到最高值的处理时间不完全同步.核异常率显著高于微核率,微核率与核异常率相关系数为 0. 44,这反映出生物体本身具有自动修复能力,能通过各种途径修复受损的DNA.此外,还探讨了泥鳅红细胞的微核来源和利用泥鳅红细胞核的遗传损伤监测河水水质的可行性及注意问题. 图 3表 6参 16  相似文献

采用红细胞微核和核异常测试法,研究了除草剂精禾草克、氟乐灵、扫茀特,2-甲-4氯钠水剂对泥鳅红细胞核的遗传毒性.结果表明,4种除草剂单独作用时均不同程度地引起微核细胞率和核异常细胞率等遗传指标的上升(P<0.05或P<0.01). 低浓度的除草剂对泥鳅红细胞的联合诱变作用比高浓度的除草剂明显.除草剂浓度与微核率或核异常率无显著相关,不表现剂量-效应关系.扫茀特的诱变效应大于其它3种除草剂.4种除草剂联合作用时具拮抗性. 图1 表5 参20  相似文献

Blood zinc level, hematological parameters and blood cell morphology were evaluated in common carp immediately after 3 h exposure to 20 mg dm(-3) of zinc (Zn0), and in 24, 48 and 96 hours after the end of it (Zn24, Zn48, Zn96). Blood zinc level in the non-exposed fish was 8 mg dm(-3), reached a maximum of 20 mg dm(-3) in Zn48, while it dropped to 9 mg dm(-3) in Zn96. Zinc caused a stress reaction in fish indicated by an increase in hematocrit value in Zn0, and elevated plasma glucose level and trombocytosis which persisted until the end of the experiment. Zinc-exposed fish showed an increased frequency of abnormal erythrocytes, and a compensatory release of immature erythrocytes to the blood stream. In zinc-treated fish, leukocyte count initially increased and subsequently decreased significantly below the control level due to a drop in lymphocyte number. Lymphocyte viability was reduced, and abnormal lymphocytes appeared. A decreased count of juvenile neutrophiles, and reduced phagocyte activity also occurred. The results indicate possible zinc-induced disturbances in both specific and non-specific immune mechanisms.  相似文献
