| 摘 要： | 真养产碱杆菌突变株Ｂ５１０（ＡｌｃａｌｉｇｅｎｅｓｅｕｔｒｏｐｈｕｓｍｕｔａｎｔＢ５１０）能以葡萄糖为碳源在无机合成培养基上积累聚β羟基丁酸．为消除发酵过程中底物的抑制和优化ＤＯｓｔａｔ分批补料培养中的补料速率，建立了真养产碱菌突变株Ｂ５１０的比生长速率μ和底物浓度Ｓ的数学模型，并求得其生长得率Ｙｘ／ｓ．在此基础上，借助一个计算路线优化出在生长期每一个ＤＯ信号所要补加的底物量为１３ｇ／Ｌ；按中心组合实验设计优化ＰＨＢ积累期最佳葡萄糖浓度为３５ｇ／Ｌ，硫酸铵为１６ｇ／Ｌ．通过ＤＯｓｔａｔ补料分批培养，最终细胞浓度为８０ｇ／Ｌ，ＰＨＢ的产量为６２ｇ／Ｌ．
