摘    要:比较了65℃温育法以及冻融法对蓝绿藻基因聚合酶链式反应(PCR)测定以及毒素酶联免疫吸附反应(ELISA)测定效率的差异.对培养的毒素分泌型的微囊藻细胞分别用两种方法预处理,做成10倍稀释系列梯度,以同样条件进行PCR扩增和ELISA毒素测定.结果表明,65℃温育法相比冻融法可分别提高ELISA和PCR测定效率不低于10倍和100倍;65℃温育法更适用于蓝绿藻细胞内物质的准确测定.

Abstract:In order to enhance the sensitivity of PCR and ELISA determination of blue-green algae, the different influence of 65℃ pretreatment and Freeze-Thaw cycle pretreatment had been compared. The cells of cyanobacteria NIES-90 that could produce toxims were pretreated by both methods and diluted to ten folds series. Then PCR and ELISA were determined under same condition. The results showed that 65℃ pretreatment compared with Freeze-Thaw cycle pretreatment could enhance the efficiency of ELISA and PCR determination to more than 10 and 100 folds respectively. Thus, 65℃ pretreatment should be more applicable for determination of blue-green algae intracellular containment.
