离子液体氯化1-辛基-3-甲基咪唑对洋葱的遗传毒性作用

摘    要:为探讨咪唑类离子液体氯化1-辛基-3-甲基咪唑([C_8mim][C1])对洋葱的遗传毒性,用浓度分别为2.5、5、10和20mmol·L~(-1)的[C_8mim][C1]处理洋葱内表皮细胞和洋葱根尖8 h,用浓度为10 mmol·L~(-1)的[C_8mim][C1]处理洋葱内表皮细胞和洋葱根尖4 h、8 h、12 h和24 h,研究[C_8mim][C1]对洋葱内表皮细胞核的影响和对洋葱根尖有丝分裂指数和染色体畸变的影响。结果显示,与对照组相比,随着[C_8mim][Cl]处理浓度和时间的增加,洋葱内表皮细胞核偏移率明显升高,根尖细胞有丝分裂指数显著降低,染色体畸变现象明显,畸变率升高。研究表明,当浓度达到2.5 mmol·L~(-1)时,[C_8mim][Cl]对洋葱具有遗传毒性。

Abstract:The aim of this study was to explore the genotoxicity of imidazole ionic liquid chloride 1-octyl-3-methylimidazolium ([C8mim][C1]). Endothelial cells (EC) and root tip cells (RTC) of onion were treated with [C8mim][C1] at the variable concentrations for 8 hours (2.5, 5, 10 and 20 mmol L-1). Cells were also treated with [C8mim][C1] at the concentration of 10 mmol L-1 for different time period of 4, 8, 12 and 24 hours, respectively. The effect of [C8mim][C1] on onion nucleus was observed, mitotic index was calculated and chromosome aberration of RTC were detected after the treatment of [C8mim][C1]. The results showed that [C8mim][Cl] exposure induced the location change of onion epidermal cell nucleus. In addition, [C8mim][Cl] caused the decrease of mitotic index of RTC and the increase of chromosome aberration ratio. These results demonstrated that the genotoxicity of [C8mim][Cl] on EC and RTC is in a dose- and time-dependent manner when the [C8mim][Cl] concentration reaches 2.5 mmol L-1.
