土壤细菌荧光原位杂交对比染色技术的构建

摘    要:荧光原位杂交技术(FISH)已经成为研究海洋、湖泊及河流沉积物微生物特征的重要手段,但对土壤的应用尚存在局限性。研究以土壤和堆肥为材料,筛选了FISH对比染色荧光剂,并对其染色条件进行了分析。研究结果表明,1/200稀释SYBR Green(ISGI)可用于FISH的对比染色。其染色时间为1 min,封片试剂为无荧光浸镜油。

Abstract:Fluorescence in situ hybridization(FISH) is widely used to describe bacterial community in marine,lake and river sediments.However,the application of FISH to soil or compopst still need to improve.In this study,the contrast stain for FISH was selected,and the dyeing conditions were analyzed.The results show that 1/200 dilution of SYBR Green I(SGI) is useful as the counterstain for FISH.The dyeing time is 1 min,and non fluorescent immersion oil was recommended to mount.
